ABSTRAK
Latar belakang: Cidera saraf perifer sebagai keluaran dari post operatif hingga saat ini belum ditangani dengan maksimal. Penelitian ini ditujukan untuk menentukan potensi dari sekretom pada regenerasi cidera saraf perifer.

Metode: Cidera saraf perifer buatan dilakukan pada tikus dengan melakukan diseksi pada saraf sciatic. Evaluasi perbaikan motorik dilakukan mengunakan Sciatic Functional Index (SFI) pada minggu ke enam (SFI 1), minggu ke sembilan (SFI 2), dan minggu ke dua belas (SFI 3). Rasio berat basah antara otot gastrocnemius kanan dan kiri dibandingkan serta dilakukan histomorphometry saraf sciatic pada tiap kelompok.

Hasil: Kelompok III menunjukan SFI 1 yang lebih baik dibandingkan kelompok I (p=0.017). Kelompok I dan III menunjukan perbedaan SF2 yang signifikan dibandingkan dengan kelompok II dan IV (p<0.001). Rasio tertinggi dari otot gastrocnemius ditemukan pada kelompok I dan III, yang bernilai 0.65 ± 0.059 dan 0.67 ± 0.179 (p<0.001). Pada histomorphometry, akson termyelinisasi paling banyak ditemukan pada kelompok I dan III, yang bernilai p<0.001.

Kesimpulan: Sekretom sel punca mesenkimal korda umbilikalis dapat digunakan sebagai terapi baru untuk menggantikan autograf pada penanganan kerusakan saraf perifer.
ABSTRACT
Background: Currently, the post-surgical outcome of peripheral nerve injury has not been optimal. The purpose of this research is to determine the potency of secretome in peripheral nerve injury regeneration.

Method: The mice had artificially-induced peripheral nerve injury, which was created by dissecting the sciatic nerve. Sciatic Functional Index (SFI) was used to evaluate the motoric recovery on week six (SFI 1), week nine (SFI 2), and week twelve (SFI 3). The mice was sacrificed on week twelve. The wet mass ratios of the right and left gastrocnemius muscle were compared, then the sciatic nerve histomorphometry evaluation was performed on each group.

Results: Group III showed a better SFI 1 result than Group I (p=0.017). Group I and III showed significantly better SFI 2 than group II and IV (p<0.001). The highest ratio of gastrocnemius muscle was found in group I and III, which were 0.65 ± 0.059 and 0.67 ± 0.179 (p<0.001). On histomorphometry, the highest number of myelinated axons were found in group I and III, which were p<0.001.

Conclusion: Umbilical cord mesenchymal stem cell secretome can be used as a new therapy to replace the autograft in peripheral nerve defect management.

Latar Belakang: Di era ini, terapi dengan sel punea untuk mengobati berbagai penyakit semakin dirninati. Sejauh ini, banyak pendapat mengenai keamanan sel punea, yang dikatakan aman untuk manusia. NaIllun" belum ada penelitian lebih jauh mengenai keamanan sel punea yang disuntikan lewat vena, untuk kesehatan pembuluh darah. Tujuan dari peneiitian ini adalah untuk menganalisis efek sel punea yang disuntikkan secara IV, pada pembuluh darah arteri yang nannal. Metode: Data didapatkan dari eksperimen klinik tikus Wistar yang dilaksanakan di Instilut Pertanian Bogor, merupakan studt awal pada tikus dengan tekanan darah normal. Tikus dengan tekanan darah nonnal (140/100 mmHg, diukur dengan CODA) tersebut dibagi menjadi 3 kelompok. Kelompok pertama ada kelompok kontrol, kelompok kedua disumikan secara IV sel punca 1 x 106 , dan kelompok keliga 3xl 06 sel, dalam sekali penyuntikan. Kelompok kedua dan ketiga diamati selama 1 bulan setelah penyuntikan sel punca. Setelah 1 bulan, tikus dinekropsi dalam keadaan anastesi. Selanjutnya, dengan bantuan J Image Software, dilakukan pcngamatan terhadap diameter dan ketebalan dinding dari meri karotis, karotis interna, karotis eksterna, aorta abdominal, iliaka kiri, dan iliaka kanan. Analisis statistik dilakukan dengan metode ANOV A terhadap 3 kelompok tikus untuk. mengukur perbedaan diameter pembuluh darah dan ketebalan dinding pembulu darah. Jika persebaran data tidak rata, maka digunakan metode T Independent Test. Hasil: Hasil analisis menunjukkan bahwa tidak ada perbedaan signifikan dalam diameter dan ketebalan dinding untuk kelompok I, kelompok 2, dan kelompok 3 untuk semua pembuluh darah kecuali arteri iliaka kiri. Arteri iliaka kiri menunjukkan perbedaan yang bennakna dalam diameter dan ketebalan pembuluh darah. Diskusi: Hasil riset ini, memmjukkan keamanan sel punca pada arteri bila disuntikan ke dalam vena. Ini ditunjukkan dengan hasil yang tidak. terdapat perbedaan bermakna dalam 5 pembuluh darah yang dipenksa. kecuali arteri iliaka kiri menunjukkan hasil yang bermaknadan perlu diteliti lebih lanjutdenganjumlah sam pel yang memadai (30 tikus). Setiap kelompok mempunyai sampel minimum 9 ekor, namun dan hasil penelitian masing-masing kelompok mempunya 5 ekor tikus yang dapat dianalisis. Sehingga hasil analisis yang didapatkan tidak bisa mencerminkan hubungan yang kuat secara statistik, namun merupakan suatu kecenderungan.

This reference work presents the basic principles of angiogenesis induction, including the roles of signaling factors such as hypoxia-inducible factors, biophysical stimulation and angiogenic cells. The book also covers lymphogenesis induction. Both the established fundamentals in the field as well as new trends in the vascularization of engineered tissues are discussed. These include pre-vascularization strategies using preparation of channeled scaffolds and preparation of decellularized blood vessel trees, approaches to inducing formation of microvasculature and approaches to inducing the growth of vascular networks. The authors expand on these concepts with current studies of dual-level approaches to engineer vascularized tissue composites. The book concludes with a discussion of current clinical approaches and the use of vascular grafts in the context of providing clinical practice with new tissue engineering strategies.

Cardiovascular diseases are still the leading cause of death in developed countries. Revascularization procedures such as coronary artery and peripheral bypass grafts, as well as access surgery represent a 2$ billion market yearly for the US alone. Despite intense research over many decades, no clinically suitable, shelf-ready, synthetic, vascular, small-caliber graft exists. There is therefore still a quest for such a clinical vascular prosthesis for surgical revascularization procedures and access surgery. Many approaches have been tried and are currently under investigation with promising results. These range from acellular and cell-based, stable or bio-degradable, synthetic scaffolds to biological or decellularized grafts, not forgetting self-assembly technologies for in vitro or in vivo VTE. All these approaches can be further enhanced by functionalization, e.g. with growth factors and drug elution. This updatable book aims to cover all the relevant aspects of Vascular Tissue Engineering (VTE) and novel alternatives to develop vascular grafts for clinical applications. The chapters in this book cover different aspects of manufacturing scaffolds with various polymers, mechanical characteristics, degradation rates, decellularization techniques, cell sheet assembly, 3-D printing and autologous mandril-based VTE. All the necessary in vitro tests such as biocompatibility and thrombogenicity are reviewed. Pre-clinical assessment of in vivo experimental models include patency, compliance, intimal hyperplasia, inflammatory reaction, cellular ingrowth and remodeling. Finally, early clinical trials will be periodically updated regarding results, regulatory aspects and post-marketing quality assessment. Furthermore, the reader should get an insight into various approaches, technologies and methods to better understand the complexity of blood surface and cell interactions in VTE.
