"Lipase merupakan salah satu enzim yang penting di industri enzim, dan banyak digunakan dalam pembuatan zat aditif makanan, kosmetik, dan industri farmasi. Penelitian sebelumnya yang dilakukan di PTB-Laptiab BPPT, pengklonaan gen sintetik T. lanuginosus lipase pada B. substilis memiliki aktivitas lipase yang rendah yaitu sebesar 1,488 U/mg.
Penelitian ini bertujuan untuk mengklona gen sintetik T. lanuginosus lipase TLL ke dalam vektor ekspresi Pichia pastoris menggunakan sinyal peptida asli TTL. Gen TLL yang mengandung sinyal peptida asli diamplifikasi dengan PCR, dan disisipkan ke dalam pPICZ? A di antara situs XhoI dan XbaI, kemudian ditransformasikan ke dalam sel kompeten E. coli DH5?.
Hasil transformasi dipilih dua rekombinan positif untuk dilakukan analisis sekuensing. Hasil sekuensing, kedua rekombinan mengandung gen target lipase. Plasmid yang telah dikonfirmasi kemudian dilinearisasi dan ditransformasikan ke dalam P. pastoris X-33 dengan menggunakan metoda elektroporasi. Gen T. lanuginosus lipase berhasil diintegrasi ke dalam kromosom P. pastoris X-33, yang ditunjukkan dengan terbentuknya zona bening pada media Yeast extract Peptone Dextrose Tributyrin YPD.TB agar yang mengandung zeocin.
Thermomyces lanuginosus lipase memiliki daerah open reading frame ORF 916 bp yang mengkode 291 asam amino dengan massa molekul teoritis 35 kDa. Enzim rekombinan T. lanuginosus memiliki suhu optimum 80 C dan pH optimum 8.

Lipase is one of the most important industrial enzymes, which is widely used in the preparation of food additives, cosmetics, and pharmaceutical industries. In the previous study, we have cloned synthetic Thermomyces lanuginosus lipase gene into Bacillus subtilis and Escherichia coli and resulting low expression of enzyme activity.
The aim of this research was to construct the T. lanuginosus lipase TLL gene into P. pastoris vector expression with TLL original signal peptide. Thermomyces lanuginosus lipase gene was amplified by PCR and contained original signal peptide and then inserted into pPICZ A between XhoI and XbaI site, and transformed into competent cell E. coli DH5. From the transformant, two of positive recombinants were analyzed by sequencing analysis.
As the result, both of two recombinant have a positive target gene which has lipase gene. The correct plasmid was linearized and then was transformed into P. pastoris X 33 by electroporation method. Thermomyces lanuginosus synthetic gene lipase has been successfully integrated into chromosome of P. pastoris X 33, which revealed by clear zones around the colony on Yeast extract Peptone Dextrose Tributyrin YPD.TB plate with zeocin.
The Thermomyces lanuginosus lipase had an open reading frame of 916 bp encoding TLL of 291 amino acids with theoretical molecular mass of 35 kDa. The recombinant enzyme, T. lanuginosus lipase had optimal temperature at 80 C and optimal pH at pH 8.0.

"Data Kematian Bayi di Kota Bogor mengalami peningkatan dari 26 menjadi 62 kasus pada tahun 2013. Posyandu merupakan salah satu salah satu wadah dalam masyarakat yang menjalankan program kesehatan dimana salah satu tujuannya adalah melaksanakan kegiatan untuk mempercepat penurunan angka kematian ibu dan bayi. Permasalahan pada posyandu di Kota Bogor adalah jumlah kader aktif yang terus menurun kemudian kaderisasi untuk kader sendiri kurang maksimal, belum berfungsinya pokja dan pokjanal posyandu.
Hasil perhitungan laporan kematian bayi tahun 2013 dan 2014 mendapatkan bahwa lebih dari separuh (55% dan 52,7%) posyandu yang wilayah kerjanya mengalami kasus kematian bayi adalah posyandu yang tingkat perkembangannya pratama dan madya. Penelitian ini ingin melihat bagaimana efektivitas UKBM Posyandu melalui faktor kepemimpinan (komunikasi, kompetensi), fungsi manajemen (perencanaan, pengorganisasian, pelaksanaan dan pengendalian), lingkungan kerja (dukungan sistem, ketersediaan sarana penunjang dan insentif) serta karakteristik responden (klasifikasi posyandu, umur, lama menjabat dan pekerjaan).
Penelitian ini menggunakan unit analisis posyandu dan sebagai respondennya adalah ketua kader posyandu sebanyak 70 posyandu. Rancangan penelitian menggunakan pendekatan cross sectional metode kuantitatif. Uji yang digunakan korelasi regresi untuk melihat hubungan faktor kepemimpinan, fungsi manajemen, lingkungan kerja dan karakteristik responden terhadap efektivitas posyandu. Analisis regresi linier ganda digunakan untuk melihat faktor yang paling menentukan efektivitas posyandu.
Hasil analisis menunjukkan bahwa ada hubungan faktor kepemimpinan, fungsi manajemen, lingkungan kerja dan karakteristik responden terhadap efektivitas posyandu. Hubungan berpola positif yang artinya semakin baik faktor kepemimpinan, fungsi manajemen, lingkungan kerja dan karakteristik responden semakin efektif posyandu. Tidak ada hubungan faktor kepemimpinan (kompetensi), karakteristik responden (klasifikasi posyandu, lama menjabat ketua posyandu) terhadap efektivitas posyandu. Peningkatan intervensi kompetensi dan pengorganisasian akan meningkatkan efektivitas posyandu dengan mengontrol variabel umur dan pekerjaan.

Data infant mortality in the city of Bogor has increased from 26 to 62 cases in 2013. IHC is one of one of the containers in a society that runs the health program where one of his goals is to carry out activities to accelerate the reduction in maternal and infant mortality. Problems on the Posyandu in Bogor is the number of active cadres who continued to decline then regeneration for their own cadres less than the maximum, not the functioning of working groups and Pokjanal Posyandu.
Report the results of the calculation of infant mortality in 2013 and 2014 found that more than half (55% and 52.7%) Posyandu whose jurisdiction experienced cases of infant mortality is Posyandu Pratama and middle level of development. This study wanted to see how the effectiveness UKBM Posyandu through leadership factors (communication, competence), the functions of management (planning, organizing, implementation and control) and working environment (support system, the availability of supporting infrastructure and incentives).
This study uses the unit of analysis is the Posyandu and as chairman of the cadre's respondents were 70 Posyandu. The design of the study using cross sectional quantitative methods. Correlation regression test used to see the relationship between leadership, management functions and working environment on the effectiveness of the Posyandu. Multiple linear regression analysis was used to look at the factors which most determine the effectiveness of the Posyandu.
The analysis shows that there is a correlation between leadership (communications), management functions (planning, organizing, implementation and control) and working environment (support system, the availability of supporting infrastructure and incentives) on the effectiveness of Posyandu. Patterned positive relationship means the better the leadership factor (communications), management functions (planning, organizing, implementation and control) and working environment (support system, the availability of supporting infrastructure and incentives) more effective Posyandu. There is no correlation between leadership (competence) on the effectiveness of Posyandu. Determinants of the efectiveness of Posyandu is organising and controlling. Improved organization and control interventions will improve the effectiveness of Posyandu.

"ABSTRAK
Sekarang ini, tingkat kematangan buah pisang Musa sp diklasifikasikan secara manual berdasarkan warna kulitnya. Pada penelitian ini, akan diperkenakan sistem otomatis tingkat kematangan buah pisang menggunakan teknologi hyperspectral. Sistem perangkat keras yang digunakan terdiri dari satu set alat pengukuran, sumber cahaya halogen dan kamera hyperspectral yang terhubung ke PC melalui Camera Link. Perangkat lunak sistem terdiri dari pengukuran hasil reflektansi citra, ekstraksi ciri, dan algoritma klasifikasi. Citra reflektansi permukaan pisang dihitung berdasarkan citra yang didapat, white reference dan dark reference. Feature extraction ekstraksi ciri didapatkan menggunakan principal component analysis pada semua range panjang gelombang hyperspectral. Dengan demikian, tingkat kematangan diklasifikasikan menggunakan artificial neural network menjadi 3 kelas yaitu, mentah, matang dan sangat matang. Sampel yang digunakan ialah 15 pisang ambon Musa acuminate colla dan 15 pisang raja Musa textilla yang masing-masing berisi 5 sampel pada setiap tingkat kematangan. Hasil penelitian ini menunjukan bahwa sistem yang diusulkan dapat mengkalsifikasikan tingkat kematangan buah pisang cukup akurat.

ABSTRACT
Nowadays, the maturity of banana is classified manually based on the surface color of banana. In this study, an automatic system was introduce using hyperspectral technology system. The hardware of system consist of a set of measuring system, light source and hyperspectral camera that connect to PC via Camera Link. The software of system consists of reflectance image profile measurement, feature extraction and classification algorithm. The reflectance image profile of the banana surface was calculated based on current image, white and dark image reference. The feature sets were computed using a principal component analysis on full wavelength range of HIS spectra. Thus, the maturity stage of banana was classified artificial neural network into 3 classes i.e. immature, mature and very mature. The samples used were 15 sampel Musa acuminate collaa and 15 sampel Moses textilla which is consist 5 samples for each aturity stage.The results show that the proposed system can classify the banana maturity stage perfectly. "

"ABSTRACT
Visualisasi pertulangan daun telah banyak dilakukan menggunakan citra RGB dan metode pengolahan yang digunakan adalah pemrosesan morfologi. Hasil dari metode tersebut dapat menampilkan pola pertulangan daun atau venasi dengan baik, namun sangat terbatas pada resolusi kamera yang digunakan serta keterbatasan informasi spektral citra daunyan dihasilkan. Pada penelitian kali visualisasi venasi berhasil dilakukan dengan citra hyperspectral dengan panjang gelombang 400-1000nm. Sistem visualisasi pada penelitian kali ini menerima input citra hyperspectral dan menghasilkan output berupa citra venasi. Proses automasi mendapatkan citra venasi menggunakan model klasifikasi. Model klasifikasi dibuat berdasarkan infomasi panjang gelombang dari vena dan bagian helaian daun. Tujuan model klasifikasi ini adalah memprediksi bagian vena pada citra hyperspectral Algoritma klasifikasi yang digunakan pada penelitian ini adalah Support Vector Machine SVM , Multi Layer Perceptron Classifier MLPC , serta Decision Tree DT . Hasil akurasi dari model mencapai 97 pada model SVM, 95 pada model MLPC, dan 81 pada model DT. Model SVM dan MLPC selanjutnya digunakan untuk memprediksi citra hyperspectral untuk menghasilkan citra venasi daun bayam merah. Hasil akhir, berupa citra venasi menggunakan model SVM lebih baik karena mampu memvisualisasikan bagian vena primer dan vena sekunder dibandingkan citra venasi dengan model MLPC.

ABSTRACT
Venation visualization broadly have been done by RGB images using morphological image processing. The result of that method can visualizing leaf venation properly, but it depends on camera resolution and limited spectral information. In this research, we developing venation visualization system using hyperspectral image on band 400 1000nm. Our system visualizing red amaranth leaf venation as a output and hyperspectral image for input. To automated identifying venation region, we built classification model to predict based on spectral information. Classification model take every hyperspectral image pixel to predict leaf vein. In this work, we made 3 classification model namely SVM Support Vector Machine , MLPC Multi Layer Perceptron Classifier , and DT Decision Tree . Our model trained by 5 fold cross validation. Average accuracy score for SVM model up to 97 , 95 for MLPC and 81 on DT. Regard this accuracy result, SVM and MLPC model used for constructed venation image and DT model fall on overfitting state. The final result, SVM perform better than MLPC by visualizing primary vein and secondary vein."

"ABSTRAK
Pekarangan adalah salah satu lanskap khas pedesaan, yang memiliki berbagai fungsi krusial. Pekarangan juga merupakan tempat konservasi berbagai sumberdaya hayati lokal. Pekarangan di Kecamatan Pujon telah mulai di kelola kembali sejak adanya kegiatan wisata. Penelitian ini bertujuan untuk mengetahui keanekaragaman vegetasi penyusun pekarangan di Kecamatan Pujon dan juga mendokumentasikan pengetahuan lokal mengenai manfaatnya. Penelitian ini dilaksanakan pada bulan April-November 2018. Sebanyak 90 pekarangan telah dijadikan sampel. Terdiri dari 30 pekarangan di dekat sungai, 30 pekarangan di dekat akses jalan dan 30 pekarangan di dekat hutan. Pekarangan tersebut dikelompokkan ke dalam tiga kategori ukuran yaitu besar, sedang dan kecil. Data diambil menggunakan wawancara terstruktur dan semi terstruktur terhadap pemilik pekarangan. Data dianalisis secara kualitatif dengan statistika deskriptif untuk menggambarkan pengetahuan lokal masyarakat. Data tumbuhan dianalisis dengan menghitung Nilai Indeks Penting (INP), Indeks Shannon-Wiener, Indeks kesamaan dan ketidaksamaan. Data pengetahuan lokal dianalisis dengan menghitung nilai kepentingan lokal (Local Users Value Index, LUVI) dan nilai kultural (Index of Cultural Significance, ICS). Terdapat 5 lanskap di Kecamatan Pujon yaitu sawah, tanah tetelan, tegalan, pekarangan dan hutan. Pekarangan merupakan lanskap terpenting keempat dari kelima lanskap tersebut. Terdapat 13 kategori guna tanaman pada pekarangan di Kecamatan Pujon. Tiga belas kategori guna tersebut adalah pangan (PDM=13,3), sayuran (PDM=11,6), bumbu (PDM=9,4), buah (PDM=8,6), minuman (PDM=8,1), obat (PDM=7,9), pakan ternak (PDM=7,7), ornamental (PDM=7,6), papan (PDM=7,1), ritual (PDM=6,5), pagar (PDM=5,3), pewarna (PDM=4,6) dan tanaman pengganggu (2,3). Pekarangan di dekat sungai memiliki nilai INP paling tinggi, diikuti oleh pekarangan di dekat hutan dan pekarangan di dekat jalan. Berdasarkan Indeks Shannon-Wiener, keanekaragaman jenis tanaman pada pekarangan di Kecamatan Pujon termasuk ke dalam kategori sedang-tinggi. Indeks kesamaan antara pekarangan berdasarkan ukurannya, lebih kecil dari pada indeks ketidaksamannya. Sebanyak 39 tanaman yang terdiri dari 3 tanaman penting dalam masing-masing kategori telah dihitung nilai kulturalnya. Bagi masyarakat di Kecamatan Pujon tanaman yang memiliki nilai ICS tinggi adalah klopo (Cocos nucifera) (ICS=53,42) dan gedang (Musa x paradisiata) (ICS=45,83). Pekarangan di Kecamatan Pujon memiliki berbagai jenis tanaman yang berguna bagi pemiliknya. Pekarangan juga memberikan ecosystem services terhadap lingkungan disekitarnya.

ABSTRACT
Home garden is one of the rural traditional landscapes, which has various crucial functions. Home garden also a place to conserve various local resources. Home garden in Pujon Sub-district has begun to be managed again since the existence of tourism activities. This research was conducted in April-November 2019. In total 90 home gardens were sampled. It consists of 30 home gardens near the river, 30 home gardens near the road access and 30 home gardens near the forest. These home garden grouped into three categories there are large, medium and small sizes. Data was taken using structured and semi-structured interviews with the home garden owner. Data were analyzed qualitatively with descriptive statistics to analyze local knowledge. Vegetation data were analyze by calculating Important Value Index (IVI), Shannon-Wiener indeks, simillarity and dissimilarity index. Local knowledge data were analyzed by calculating Index Cultural Significance (ICS), and Local User Value Index (LUVI). There are 5 landcapes in Pujon Sub-district there are, sawah, tanah tetelan, tegalan, home garden and forests. Home garden is the fourth important lanskap in Pujon Sub-district. There are 13 categories of plants used in the home garden in Pujon Sub-district. There are food (PDM = 13.3), vegetables (PDM = 11.6), spices and herbs (PDM = 9.4), fruit (PDM = 8.6), beverages (PDM = 8.1), medicinal plant (PDM = 7.9), fodder (PDM = 7.7), ornamental (PDM = 7.6), home material (PDM = 7.1), ritual and spiritual (PDM = 6.5), fence (PDM = 5.3), natural coloring for foods (PDM = 4.6) and weeds and grasses (2,3). Home garden near the river have hightest IVI, followed by home garden near the forest and home garden near the road. Based on Shannon-Wiener Index, the flora diversity of home garden In Pujon Subdistrict are medium-rich. Simillarity index between home garden based on their sizes, are smallest than the dissimilarity index. In total 39 plants have analyzed using ICS, its consist of 3 plant from each categories. For the people in Pujon Sub-district, the plants that have high ICS were klopo (Cocos nucifera) (ICS = 53.42) and gedang (Musa x paradisiata) (ICS = 45.83). Home garden in Pujon Subdistrict consist of many plant species that have important role for the owner. Home garden also provide ecosystem services for the environtment.

"Salah satu penyebab kesakitan dan kematian ibu adalah masih rendahnya
pengetahuan ibu hamil. Guna mengatasi hal tersebut maka Kementerian
Kesehatan RI merencanakan Kelas Ibu Hamil (KIH) yang bertujuan untuk
meningkatkan pengetahuan ibu hamil. Penelitian ini dilakukan untuk mengetahui
pengaruh KIH di Puskesmas Cipaku Kota Bogor terhadap pengetahuan kesehatan
maternal. Penelitian ini menggunakan metode penelitian kuantitatif guna
mengetahui pengaruh KIH terhadap pengetahuan serta kualitatif untuk
mengetahui gambaran pelaksanaan KIH. Hasil penelitian menunjukan
peningkatan pengetahuan sesudah pelaksanaan KIH, dimana peningkatan lebih
tinggi terjadi pada kelompok intervensi. Selain itu terjadi penurunan retensi
pengetahuan pada 7 hari setelah KIH sebesar 4%. Temuan kuantitatif ini didukung
dengan temuan kualitatif yang menunjukan masih ditemukannya berbagai
hambatan pada pelaksanaan KIH. Uji instrumen menunjukan kuesioner KIH
belum memiliki validitas dan reliabilitas yang baik sehingga perlu dilakukan
perbaikan.

One of the issues causing maternal morbidity and mortality is the lack of
knowledge among pregnant mothers. In order to overcome this problem, the
Ministry of Health of the Republic of Indonesia has planned a program named
Kelas Ibu Hamil (KIH) or a class for pregnant mothers designed to improve their
knowledge. This study aims at finding out the effects of KIH held at Puskesmas
Cipaku Kota Bogor – on knowledge about maternal health. Using quantitative
method, this study discovers how KIH affects knowledge; meanwhile, the
qualitative method describes the implementation of KIH at this particular area.
Results indicate increasing knowledge among pregnant mothers after the
implementation of KIH, as more considerable improvemens were found within
exposed group. However, 7 days after KIH, their knowledge retention decreased
4%. These quantitative results are supported by some qualitative findings, which
indicate several obstacles coming across the program. Instrument test shows that
the validity and reliability of the KIH questionnaire has not yet been confirmed
yet, therefore requires further improvements

"[ABSTRAK
Kebugaran merupakan prediktor dari penyakit degeneratif seperti penyakit
kardiovaskular, diabetes mellitus dan lain sebagainya. Hasil tes kebugaran pada
siswa sekolah menengah atas di kota Bogor yang dilakukan oleh Dinas Kesehatan
pada tahun 2014, menyebutkan 91.29% siswa berada pada tingkat kebugaran
kurang dan kurang sekali. Perilaku merokok, jenis kelamin, status gizi, frekuensi
olahraga, serta lingkar pinggang, kadar lipid dan tekanan darah, diperkirakan
menjadi determinan kebugaran, menurut laporan Survei Kepatuhan terhadap KTR
di Kawasan Sekolah tahun 2014, terdapat 15.18% siswa yang merokok. Penelitian
ini bertujuan untuk melihat apa saja determinan kebugaran kardiorespirasi pada
siswa di 18 sekolah menengah atas di Kota Bogor. Disain penelitian ini crosssectional
menggunakan tiga data sekunder Dinkes Kota Bogor, tes kebugaran
menggunakan metode TKJI untuk usia 16-19 tahun. Sampel penelitian didapatkan
354 responden yang tersebar pada 18 sekolah. Pada analisis regresi logistik
ganda, ditemukan bahwa variabel jenis kelamin, status gizi, perilaku merokok dan
lingkar pinggang merupakan determinan kebugaran kardiorespirasi, dengan
variabel jenis kelamin yang dominan berhubungan dengan kebugaran
kardiorespirasi. Perlu dibuat program gerakan hidup aktif untuk penanganan
masalah gemuk dan obesitas agar adanya peningkatan kebugaran jasmani. Bagi
penelitian selanjutnya, perlu penggunaan metode pengukuran kebugaran TKJI
secara lengkap atau dengan metode pengukuran yang lain seperti single-test, dan
pengukuran aktivitas fisik yang lebih baik lagi.

ABSTRACT
Fitness is predictor of degenerative diseases, such as cardiovascular disease,
diabetes etc. Result on fitness test among high school students at Bogor, which
was conducted by the city district health office in 2014, mentioned that 91.29%
students were on poor fitness level. Smoking behavior, sex, nutritional status,
sport frequency, blood lipid, waist circumference and blood pressure were
estimated as determinant to fitness level, according to a report from Survei on
Adherence of Non-Smoking Area on School Area in 2014, there are 15.18% of
students were smoking. This research aims to see which factors are determinant to
student?s cardiorespiratory fitness in 18 high school at Bogor. Design of this
research was cross-sectional using three secondary data from Bogor District
Health Office, fitness test using the TKJI method for the age of 16-19 years.
Sample research obtained 354 respondents were scattered in 18 schools. On
multiple logistic regression analysis, it was found that sex, nutritional status,
smoking behavior and waist circumference are determinant to cardiorepiratory
fitness, with sex were the dominant variable associate with cardiorepiratory
fitness. The program on active lifestyle are needed to managing on overweight
and obesity problem, thus increase level of fitness. For further research, full
method on TKJI or other fitness measurement methods such as single-test are
needed , and measurement of physical activity needs to be better again., Fitness is predictor of degenerative diseases, such as cardiovascular disease,

research was cross-sectional using three secondary data from Bogor District
Health Office, fitness test using the TKJI method for the age of 16-19 years.
Sample research obtained 354 respondents were scattered in 18 schools. On
multiple logistic regression analysis, it was found that sex, nutritional status,
smoking behavior and waist circumference are determinant to cardiorepiratory
fitness, with sex were the dominant variable associate with cardiorepiratory
fitness. The program on active lifestyle are needed to managing on overweight
and obesity problem, thus increase level of fitness. For further research, full
method on TKJI or other fitness measurement methods such as single-test are
needed , and measurement of physical activity needs to be better again.]"
