ABSTRAK
Oksiresveratrol merupakan salah satu kelompok stilben yang berpotensi digunakan sebagai agen pencerah kulit, serta memiliki aktivitas antioksidan, antiviral, dan antibakteri. Oksiresveratrol dapat ditemukan pada bagian ranting tanaman murbei Morus alba L. Ekstraksi oksiresveratrol dari tanaman ini dapat dilakukan dengan berbagai metode. Pada penelitian ini, ekstraksi dilakukan dengan metode Microwave-Assisted Extraction MAE menggunakan pelarut surfaktan, yaitu polioksietilen 20 sorbitan monolaurat, atau yang biasa disebut dengan Tween 20, untuk mendapatkan kadar oksiresveratrol optimum dari ranting murbei. Optimasi dilakukan terhadap beberapa faktor yaitu waktu ekstraksi, rasio pelarut terhadap sampel, dan konsentrasi pelarut, dengan menggunakan Response Surface Methodology RSM. Selain itu, dilakukan juga ekstraksi oksiresveratrol dengan pelarut etanol 96 secara maserasi, sebagai pembanding hasil ekstraksi. Penetapan kadar oksiresveratrol dilakukan menggunakan Kromatografi Cair Kinerja Tinggi KCKT dengan fase gerak larutan asam asetat 0,5 -asetonitril 3:1 v/v dan dideteksi pada panjang gelombang 320 nm. Kondisi optimum diperoleh pada waktu ekstraksi 5 menit, rasio pelarut-sampel 40:1 mL/g, dan konsentrasi pelarut 100 mM dengan kadar oksiresveratrol 0,0172 mg/g simplisia. Sementara, pada ekstraksi menggunakan metode maserasi dengan pelarut etanol 96, diperoleh kadar oksiresveratrol sebesar 1,5704 mg/g simplisia.
ABSTRACT
Oxyresveratrol belongs to stilbenoid group that has been reported possess many biological activities such as whitening agent, antioxidant, antiviral, and antibacterial. Oxyresveratrol can be found in white mulberry Morus alba L. The highest content was found in the twigs. There are many methods can be used to extract oxyresveratrol from white mulberry twigs. In this study, polyoxyethylene 20 sorbitan monolaurate Tween 20 used as solvent with Microwave Assisted Extraction MAE as extraction method to obtain optimum oxyresveratrol content from mulberry twigs. Extraction parameters were extraction time, liquid solid ratio, and solvent concentration. The parameters were optimized using response surface methodology RSM. This research also comparing the content of oxyresveratrol with conventional extraction methods. The method was maceration with 96 ethanol as solvent. Oxyresveratrol content was determined using High Performance Liquid Chromatography HPLC with 0.5 acetic acid acetonitrile 3 1 v v as mobile phase and detection wavelength was 320 nm. Based on the result, the optimum condition was extraction time 5 minutes, liquid solid ratio 40 1 mL g, and Tween 20 concentration 100 mM. Oxyresveratrol content from optimum condition was 0.0172 mg g dry weight. Meanwhile, the result from maceration method was 1.5704 mg g dry weight.

Transfersom adalah agregat yang sangat mudah beradaptasi dan elastis. Piroksikam sebagai salah satu NSAID merupakan pilihan yang sangat layak dalam mengobati nyeri. Namun penggunaan piroksikam sering dikaitkan dengan sejumlah efek samping, terutama pada gastrointestinal, dan termasuk dalam sistem klasifikasi biofarmasetika obat dengan kelarutan yang rendah. Penelitian ini bertujuan untuk memperoleh formulasi transfersom piroksikam menggunakan Tween 20 dan Tween 80 sebagai edge activator dan membandingkan penetrasi in vitro transfersom piroksikam dengan Tween 20 dan Tween 80 dalam sediaan gel. Pembuatan transfersom menggunakan metode hidrasi lapis tipis. Transfersom piroksikam dengan Tween 20 menghasilkan ukuran partikel sebesar 170,5 nm, PDI sebesar 0,357, zeta potensial sebesar -28,7 mV, efisiensi penjerapan 32,442%, dan indeks deformabilitas sebesar 0,429, sementara transfersom piroksikam dengan Tween 80 menghasilkan ukuran partikel sebesar 133,6 nm, PDI sebesar 0,260, zeta potensial sebesar -30,6 mV, efisiensi penjerapan sebesar 34,3041%, dan indeks deformabilitas sebesar 0,269. Uji penetrasi sediaan gel transfersom piroksikam dengan Tween 20 menghasilkan jumlah kumulatif 1769,2085 ± 406,226 μg/cm2 dengan persen penetrasi sebesar 47,6434 ± 9,644 % dan fluks sebesar 191,8 ± 51,84 μg/cm2.jam sementara dengan Tween 80 jumlah kumulatif yang dihasilkan sebesar 1500,8199 ± 297,983 μg/cm2 dengan persen penetrasi sebesar 40,6249 ± 7,43 % dan fluks sebesar 186,12 ± 42,85 μg/cm2.jam. Kedua formulasi tranfersom memberikan hasil yang baik dan uji penetrasi secara in vitro menunjukkan bahwa formulasi gel transfersom piroksikam dengan surfaktan Tween 20 memberikan hasil yang lebih baik dibanding Tween 80. ......Transfersomes are highly adaptable and elastic aggregates. Piroxicam as an NSAID is a very feasible option in treating pain. However, the use of piroxicam is often associated with a number of side effects, especially gastrointestinal, and piroxicam is included in the biopharmaceutical classification system with low solubility. This study aimed to obtain a formulation of piroxicam transfersome using Tween 20 and Tween 80 as edge activator and to compare in vitro penetration of piroxicam transfersome with Tween 20 and Tween 80 in a gel preparation. Transfersome making used thin layer hydration. The piroxicam transfersome with Tween 20 resulted a particle size of 170,5 nm, a PDI of 0,357, a zeta potential of -28,7 mV, an entrapment efficiency of 32,442%, and a deformability index of 0,429, while the piroxicam transfersome with Tween 80 resulted a particle size of 133,6 nm, PDI of 0,260, zeta potential of -30,6 mV, entrapment efficiency of 34.3041%, and deformability index of 0,269. The penetration test for transfersome piroxicam gel preparations with Tween 20 resulted in a cumulative amount of 1769,2085 ± 406,226 μg/cm2 with a percent penetration of 47,6434 ± 9,644% and a flux of 191,8 ± 51,84 μg/cm2.hour, meanwhile the cumulative amount with Tween 80 resulted 1500,8199 ± 297,983 μg/cm2 with a percent penetration of 40,6249 ± 7,43% and a flux of 186,12 ± 42,85 μg/cm2.hour. Both tranfersome formulations given good results and in vitro penetration tests showed that the formulation of piroxicam transfersome gel with surfactant Tween 20 gave better results than Tween 80.

Studies on the determination of Critical Micelle Concentration (CMC), of mixed surfactants were carried out at 30°C for hexadecyltrimethylammonium bromide (HTAB) and polyoxyethylene sorbitan monolaurate (Tween 20), hexanol and water. From the phase diagram, a composition of 0.2 to 1.0 fraction of water is the best area for normal micelle region for HTAB and 0.3 to 1.0 fraction of water for Tween 20 with curving upward the 1-hexanol apex maximum solubility of 1-hexanol. From the fluorescence method, the CMC value for pure T20 and HTAB were 1.6293×10-3 M and 2.7439×10-3 M respectively. While for CMC1 and CMC11 value for mixed surfactant system at mole ratio 0.2: 0.8 (T20: HTAB) were CMC1: 2.358×10-3 M and CMC11: 7.0741×10-3 M. These finding were observed upon the theoretical values, which indicate the synergistic behavior between both surfactants.
