ABSTRAK
Udang windu (Penaeus monodon Fab.) mempunyai pertumbuhan yang baik pada salinitas 10-25 ppt. Tetapi di Indonesia hanya sedikit tambak yang selama setahun penuh dengan kisaran salinitas tersebut. Pada musim hujan, salinitas tambak cenderung turun menjadi 5-10 ppt dan di musim panas salinitas tambak naik menjadi 34-70 ppt.

Penelitian ini bertujuan untuk mengetahui: (1) Mortalitas dan batas toleransi udang windu stadium juwana terhadap salinitas; (2) Kisaran preferensi udang windu stadium juwana terhadap salinitas; (3) Pengaruh salinitas terhadap jumlah pakan yang dikonsumsi dan lamanya pakan berada dalam tubuh udang windu stadium juwana.

Dari grafik Lethal Dose 50 % (LD50) diketahui bahwa udang windu stadium juwana pada salinitas rendah mempunyai batas toleransi 3,6 ppt dan salinitas tinggi pada 44,5 ppt. Dari hasil uji preferensi dapat disimpulkan bahwa udang windu stadium juwana menyenangi kisaran salinitas 19-23 ppt. Sedangkan dari hasil uji anava satu faktor menunjukkan bahwa salinitas tidak berpengaruh terhadap jumlah pakan yang dikonsumsi dan lamanya pakan berada dalam tubuh udang windu stadium juwana. BACKGROUND: Black tiger shrimp Penaeus monodon is one of the common causes of shellfish allergy that is increasing worldwide. One of the important problems in the management of shellfish allergy is the lack of accurate diagnostic assay because the biological and immunological properties of allergens in black tiger shrimp have not been well characterized. This study aims to detect proteins with the ability to bind and cross-link immunoglobulin E (IgE) from black tiger shrimp by enzyme-linked immunosorbent assay (ELISA), Western blot, and a humanized rat basophilic leukemia reporter cell line RS-ATL8. METHODS: Sera from shrimp allergic subjects were subjected to ELISA and Western blots using raw or cooked shrimp extract as antigens. Pooled sera were used to sensitize the RS-ATL8 reporter cell line and cells were activated by shrimp extract. Eluted protein extracts separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) were tested on the RS-ATL8 cell line and subjected to mass spectrometry to identify potential candidate allergens. RESULTS: Allergic sera reacted stronger to raw shrimp extract than cooked shrimp extract (P=0.009). Western blot demonstrated that major IgE reactivity protein bands were at 32-39 kDa and 91-230 kDa in both raw and cooked shrimp extracts. The eluted protein bands at the molecular weight of 38 and 115 kDa from raw shrimp extract induced IgE cross-linking as assayed by the RS-ATL8 cell line. These protein bands were subjected to mass spectrometry for analysis. Ubiquitin-activating enzyme and crustacyanin were identified as potential candidate novel shrimp allergens. CONCLUSIONS: The RS-ATL8 reporter cell line can be used to identify potential new shrimp allergens that can functionally cross-link IgE and induce mast cell degranulation.

Segara Anakan Lagoon (SAL) is widely known as a traditional fishing ground for many aquatic organisms and is located in Central Java, in an area with high rainfall rates. The objectives of this study were to determine the effects of rainfall on the distribution of Penaeus monodon Fabricius in SAL and/or mangrove waters and to explain the cause of yearly fluctuations in this area’s fish catch during a 13-year period from 1998 to 2011. The effects of rainfall on the local distribution and abundance of shrimp in SAL, Cilacap, Central Java, Indonesia, were examined using the Anco method for three periods, namely: first period (i.e., commercial catch production, 1998–2011), second period (December 2010–November 2011) and third period (December 2011–April 2012) as part of a shrimp fishery and eco-biology study in this region. The marked increase in rainfall from 557 mm during the East Monsoon (June–August) to 1,225 mm in the second transition season (September–November) and West Monsoon (December–February) in the Segara Anakan region enhanced the seasonal movement of shrimp into the Zone IV fishing ground and produced an initial increase in the abundance of adults (CL>25 mm) in the region from 312 to 2,630 individuals. This initial increase in adult abundance enhanced the shrimp’s reproductive potential, while heavy rainfall indirectly assisted the recruitment of young shrimp into the estuary, their growth, and survival, to increase shrimp abundance in the following year. Lower rainfalls from July to September) adversely affected shrimp population and usually resulted in smaller populations (312 individuals). Statistical analysis of the relationship between shrimp catch and annual rainfall showed a high level of significance at 1%.

Lumpur bor berbasis air dengan aditif Barit (B) dan KCl (K) berpotensi toksik, sehingga Lemigas berupaya melakukan pencegahan berdasarkan pengujian TCLP dan LC50 - 96 jam terhadap Penaeus monodon. Kondisi uji disesuaikan pada Sumur Bangau #1 di Sesulu PSC, Selat Makassar. Dengan kombinasi Bmin, Bmax, Kmin, dan Kmax, konsentrasi Cu pada setiap formula (Bmin-Kmin: 26,17 ppm; Bmin-Kmax: 39,74 ppm; Bmax-Kmin: 21,47 ppm; Bmax-Kmax: 31,7 ppm) dan Pb pada Bmin-Kmin (9,369 ppm) melewati baku mutu lingkungan. LC50 Formula Bmin-Kmin memenuhi baku mutu lingkungan (44.058 ppm), sedangkan Formula Bmax-Kmax tidak memenuhi baku mutu lingkungan (13.269 ppm). Hal ini dipengaruhi oleh komposisi logam berat, toksisitas KCl, dan kondisi lingkungan. WBM jenis ini lebih baik digunakan pada pengeboran off-shore.
Water based mud with Barite (B) and KCl (K) as additives have toxicity potential, therefore Lemigas performed testing prevention effort based on TCLP and LC50 – 96 hours on Penaeus monodon. Testing condition environment was adapted to Sumur Bangau #1 at Sesulu PSC, Makassar Strait. Cu concentration in Bmin, Bmax, Kmin, and Kmax combinations (Bmin-Kmin: 26,17 ppm; Bmin-Kmax: 39,74 ppm; Bmax-Kmin: 21,47 ppm; Bmax-Kmax: 31,7 ppm) and Pb in Bmin-Kmin (9,369 ppm) are above the threshold. LC50 Bmin-Kmin Formula fulfill the threshold (44.058 ppm) while Bmax-Kmax Formula did not (13.269 ppm). It is influenced by heavy metals composition, KCl toxicity, and environmental condition. This type of WBM is better used in off-shore drilling operation.
