ABSTRAK
Pembatasan industri rokok harus disertai dengan adanya pengembangan produk alternatif non-rokok berbahan dasar tembakau untuk menjamin keberlangsungan petani tembakau. Salah satu produk alternatif tersebut adalah ekstrak tembakau sebagai biopestisida dengan nikotin sebagai bahan aktifnya. Pada penelitian ini, metode ekstraksi digesti dilakukan dalam berbagai variasi suhu sementara metode refluks dilakukan dalam berbagai variasi waktu untuk mendapatkan kondisi ekstraksi nikotin yang optimum. Yield nikotin didapatkan dengan mengalikan yield ekstrak, yang merupakan perbandingan massa ekstrak terhadap bahan, dengan kadar nikotin ekstrak yang diperoleh menggunakan HPLC (High Performance Liquid Chromatograhy). Hasil akhir menunjukkan bahwa pada ekstraksi digesti menggunakan pelarut etanol selama 2 jam, suhu yang optimal adalah 50°C dengan yield nikotin 3,92%. Sedangkan untuk ekstraksi refluks menggunakan pelarut metanol, waktu ekstraksi yang optimal adalah 6 jam yield nikotin 6,23%. Uji antifungi menunjukkan bahwa ekstrak tembakau dapat menghambat pertumbuhan fungi Aspergillus niger sehingga berpotensi sebagai fungisida. Diharapkan penelitian ini dapat menjadi pemicu bagi industri untuk memproduksi biopestisida berbahan baku tembakau sehingga dapat menyerap hasil panen tembakau demi keberlangsungan petani tembakau di Indonesia.
ABSTRACT
Cigarette industry’s restriction must be accompanied by development of non-cigarette, tobacco-based product to ensure the future of tobacco farmers. One of the prospective utilization of tobacco is as biopesticide with nicotine as its active ingredient. In this study, the extration method of digestion was conducted in a variety of temperatures while the reflux method was carried at a variety of extraction time to obtain the optimum condition of nicotine extraction. Nicotine yield was obtained with multplying the extraction yield by the concentration of nicotine in the extract which was obtained through HPLC (High Performance Liquid Chromatograhy). The result showed that digestion extraction using etanol for 2 hours has optimum temperature 50°C with nicotine yield of 3.92%. For reflux extraction with metanol, the optimum time is 6 hours with 6.23% yield. Antifungal test showed tobacco extract inhibited the growth of Aspergillus niger thus it could to be utilized as fungicide. It is expected that the study may trigger industries to produce tobacco biopesticides so it can utilize the harvest from tobacco farmers to sustain their livelihood.

Sintesis ZnO-NP, WO3-NP, dan nanomaterial ZnO-WO3 dilakukan menggunakan ekstrak daun tembakau sebagai sumber basa lemah dan capping agent. Nanopartikel dan nanomaterial hasil sintesis dikarakterisasi menggunakan FTIR, UV-Vis DRS, dan XRD. Hasil karakterisasi dengan FTIR menunjukkan adanya ikatan Zn-O dan W-O pada nanomaterial ZnO-WO3. Hasil karakterisasi UV-Vis DRS menunjukkan bahwa ZnO-NP memiliki band gap 3.21 eV, WO3-NP memiliki band gap 2.30 eV, dan nanomaterial ZnO-WO3 memiliki band gap 2.60 eV. Hasil karakterisasi XRD menunjukkan bahwa ZnO-NP memiliki struktur kristal heksagonal, WO3-NP memiliki struktur kristal triklinik, dan nanomaterial ZnO-WO3 memiliki peak gabungan ZnO-NP dan WO3-NP. Adanya peak baru mengindikasikan terbentuknya struktur baru jejaring ZnO dan WO3. ZnO-NP hasil sintesis memiliki ukuran kristalin sebesar 37.31 nm dan WO3-NP hasil sintesis memiliki ukuran kristalin sebesar 25.35 nm. Aktivitas fotokatalitik nanopartikel ZnO, WO3, dan nanomaterial ZnO-WO3 dilakukan pemodelan degradasi terhadap metilen biru. Aktivitas fotodegradasi metilen biru terjadi peningkatan dibawah sinar tampak dengan nanomaterial ZnO-WO3. Hasil menunjukan bahwa selama 90 menit, degradasi metilen biru menggunakan nanomaterial ZnO-WO3 sebesar 84.19 , sedangkan degradasi metilen biru menggunakan ZnO-NP sebesar 37.86 dan degradasi metilen biru menggunakan WO3-NP sebesar 40.65.
In this research, we synthesize ZnO NP, WO3 NP, and nanomaterial ZnO WO3 using tobacco leaves extract as a weak base source and capping agent. The synthesized nanoparticle and nanomaterial are characterized using FTIR, UV Vis DRS, and XRD. FTIR characterization shows that there are Zn O and W O bond for nanomaterial ZnO WO3. UV Vis DRS characterization shows that ZnO NP has a band gap of 3.21 eV, WO3 NP has a band gap of 2.30 eV, and nanomaterial ZnO WO3 has a band gap of 2.60 eV. XRD characterization shows that ZnO NP has a hexagonal crystal structure, WO3 NP has a triclinic crystal structure, and nanomaterial ZnO WO3 has some peaks combination of ZnO NP and WO3 NP, also there are some new peaks indicates there is a new structure which is ZnO NP and WO3 NP make a network. ZnO NP has a crystalline size of 37.31 nm and WO3 NP has a crystalline size of 25.35 nm. Photocatalytic activity of nanoparticles and nanomaterial is observed by degrading methylene blue as a model in this research. It shows an enhancement of fotodegradation activity of methylene blue under visible light using nanomaterial ZnO WO3 if it is compared with the ZnO NP and WO3 NP, which is under irradiation through 90 minutes, we obtain degradation of ZnO WO3 is 84.19 , whereas for ZnO NP is 37.86 and for WO3 NP is 40.65.

The hypersensitive reaction (HR) jn plants is a plant defense system against pathogen attack. HR is characterized by the rapid death of infected cells accompanying the formation of necrotic lessions in which pathogens are thought to be enclosed. Following the early signaling events activated by pathogen attack, the elicitor signals are often amplified through the generation of secondary signal molecules such as salicylic acid (SA) and Jasmonate acid (JA). Some studies have shown thai SA- and JA-signaling pathways are interconnected in complicated ways and it is still not clear how defense response is modulated by SA and JA. To understand better the involvement of JA. the action of MeJA on harpin-induced HR was investigated by using tobacco plant (wildtype and transgenic carrying a chimeric gene of PSPAL2 promoter and GUS gene. Leaves of of tobacco were infiltrated with harpin and MeJA. The results from HR assay and GUS histochemical staining showed that induction of HR by harpin (250 ug/ml) seems to be modulated by 100 uM MeJA. These results suggest that exogenenously application of MeJA will affect the endogenous concentration in plant that will become a switching point for MeJA to lead the potentiation or suppression of defense response.

Penelitian dalam disertasi ini mengusulkan metode ekstraksi panas refluks etanol untuk mendapatkan kadar nikotin yang tinggi dalam ekstrak hasil refluks etanol EHRE . Kandungan tertinggi nikotin dalam ekstrak diperoleh sebesar 6,30 b/b , yang dicapai pada 6 jam ekstraksi dengan suhu optimum 70 C, kecepatan pengadukan 150_rpm dan rasio solid terhadap pelarut sebesar 1:5. Nikotin yang dihasilkan dari literatur hanya berkisar 0,30 ndash; 3,60 berat berdasarkan berat kering daun tembakau b/b . Pengembangan teknik ekstraksi dalam penelitian ini ternyata mampu meningkatkan kadar nikotin 60 lebih tinggi daripada hasil yang ada dalam literatur. Peningkatan yang signifikan ini sangat penting untuk keperluan data lebih lanjut pada pemurnian nikotin. Ekstrak yang diperoleh kemudian diaplikasikan sebagai biopestisida terhadap hama penggerek buah kopi Hypothenemus hampei . Aplikasi EHRE sebagai biopestisida menunjukkan intensitas rata-rata serangan hama penggerek buah kopi hanya 1,83 dibandingkan dengan kontrol 12,58 selama 6 minggu pengamatan. Hasil tersebut menunjukkan bahwa EHRE N. tabacum var. Virginia memiliki potensi kuat dan efektif digunakan sebagai biopestisida melawan hama penggerek buah kopi. Simulasi produksi pabrik biopestisida skala komersial berikut studi kelayakan ekonominya juga dilakukan dalam penelitian ini. Data diambil dari literatur maupun hasil penelitian pada skala pilot. Berdasarkan simulasi dan analisis menggunakan software SuperPro Designer v9.0, diperoleh kapasitas produksi sebesar 433 kg.batch-1 atau 570.282 kg.tahun-1. Nilai ROI; Payback period; IRR; dan NPV berturut-turut sebesar 30,57 ; 3,27 tahun; 30,55 ; dan 16.580.000 USD.
This study proposed a heat reflux extraction HRE method to increase the yield of nicotine from ethanolic heat reflux extract EHRE of N. tabacum. The highest yield of nicotine 6.30 wt was achieved at 6 hours, optimum temperature at 70 C, 150 rpm and fixed solid-to-solvent ratio at 1:5. Nicotine yields from the literatures were obtained between 0.30 to 3.60 wt of nicotine per dry weight of tobacco leaves. Development of the HRE increased the nicotine yield to 60 more than the highest yield reported in the literatures. This significant improvement is important for further purification in nicotine production. The EHRE was then applied as a biopesticide against coffee berry borer Hypothenemus hampei . Application of the EHRE as a biopesticide shows that average intensity of the coffee berry borer attack was 1.83 compared with the control 12.58 for six weeks observation. These results indicated that the EHRE of N. tabacum var. Virginia has strong potential and effective as a biopesticide against coffee berry borer. Simulation of production of biopesticide plant at commercial scale and its feasibility study was accomplished in this research. Data were taken from the literature as well as pilot scale results. The simulation results showed that capacity of biopesticide production was 433_kg.batch-1_or 570,282_kg.year-1, while values of ROI; payback period; IRR; and NPV were 30.57 ; 3.27 years; 30.55 ; and 16,580,000 USD.

Penggantian pestisida sintetis dengan pestisida nabati sangat diperlukan karena lebih ramah lingkungan. Tembakau banyak dieksploitasi untuk produksi rokok, sehingga perlu dikembangkan untuk produk alternatif lain, salah satunya adalah insektisida karena mengandung nikotin dan senyawa toksik lainnya. Penelitian ini bertujuan untuk mengetahui efektivitas tembakau sebagai insektisida untuk Galleria mellonella dan Gryllus bimaculatus. Daun tembakau Virginia diekstraksi menggunakan teknik Ethanolic Heat Reflux Extraction (EHRE) selama 6 jam. LC50 dianalisis terhadap serangga selama 24 jam. Fraksinasi menggunakan corong pisah dilakukan dengan menggunakan empat rasio volume pelarut diklorometana. Karakterisasi oleh GC-MS dilakukan untuk mengidentifikasi dan membandingkan komponen di dalam fraksi dan ekstrak. Berdasarkan hasil penelitian, metode EHRE menghasilkan 23.16% yield. Nilai LC50 adalah 36.6 mg/ml untuk Galleria mellonella dan 38.5 mg/ml untuk Gryllus bimaculatus. Karakterisasi oleh GC-MS menunjukkan bahwa nikotin adalah komponen yang dominan. Fraksinasi meningkatkan konten nikotin sebesar 62.07% (1:1), 60.06% (1:2), 64.31% (1:3) dan 45.33% (1: 4). Konten nikotin pada fraksi lebih tinggi dibandingkan konten nikotin dari EHRE (38,8%). ......Replacement of synthetic pesticide with natural pesticide is highly demanding because it is more environmentally friendly. Tobacco is majorly exploited for cigarettes production. Therefore, it needs to be developed for other alternative products, one of which is insecticide due to nicotine content and other toxic compounds. This research aimed to determine the effectiveness of tobacco as insecticide for Galleria mellonella and Gryllus bimaculatus. Virginia tobacco leaves were extracted using Ethanolic Heat Reflux Extraction (EHRE) technique for 6 hours. The LC50 was analyzed against the insects for 24 hours. Fractionation by shake flask method was done using four volume ratios of dichloromethane solvent. Characterization by GC-MS was conducted to identify and compare the components inside the fractions and the extract. From the result, EHRE method produced 23.16% of yield. The LC50 values were 36.6 mg/ml for Galleria mellonella and 38.5 mg/ml for Gryllus bimaculatus. Characterization by GC-MS showed that nicotine was the dominant component. Fractionation increased nicotine content by 62.07% (1:1), 60.06% (1:2), 64.31% (1:3) and 45.33% (1:4), which were higher than the nicotine content from EHRE (38.8%). This study proved the effectiveness of tobacco extract as insecticide for Gryllus bimaculatus and Galleria mellonella.
