"ABSTRAK
Pendahuluan. Osteoarthritis (OA) adalah sebuah penyakit sendi degeneratif yang
menyebabkan disabilitas dengan prevalensi yang terus meningkat. Hormon
pertumbuhan memiliki efek regenerasi tulang rawan secara langsung melalui
stimulasi sel kondroblas dan proses morphoangiogenesis juga melalui faktor
pertumbuhan secara sistemik. Penelitian ini bertujuan untuk mengetahui manfaat
suntikan sendi dengan hormon pertumbuhan pada kasus Osteoarthritis.
Metode Penelitian. Penelitian dilakukan di Rumah Sakit Hewan Institut
Pertanian Bogor pada bulan Mei hingga September 2015. Desain penelitian adalah
randomized post test only control group. Sejumlah 21 ekor kelinci Selandia Baru
putih, berat 1.9-2.6kg, usia 7-8 bulan. Kelinci dibagi secara acak menjadi
kelompok kontrol (NaCl 0.9%), suntikan hormon pertumbuhan (4iu), dan suntikan
asam hyaluronat (6mg) . Dengan metode acak tersamar dilakukan suntikan
kolagenase tipe II C. Histolyticum pada hari 1 dan ke 4 pada lutut kiri, kemudian
tindakan penyuntikan dilakukan sebanyak tiga kali dengan selang waktu 1
minggu. Evaluasi dengan periode kepincangan, skoring makroskopis, histologis
dilakukan pada minggu ke-8 pasca penyuntikan pertama.
Temuan Penelitian. Berdasarkan hasil penelitian ditemukan periode kepincangan
pada grup yang diberikan hormon pertumbuhan lebih singkat, dan bermakna
secara statistik dibandingkan dengan grup kontrol (p<0.001), grup asam
hyaluronat (p<0.03), dan grup hormon pertumbuhan (p<0.001). Evaluasi skor
makroskopik dengan skor yoshimi menunjukan bahwa kelompok hormon
pertumbuhan memiliki kerusakan tulang rawan yang lebih ringan jika
dibandingkan dengan grup kontrol (p=0.001) dan grup asam hyaluronat (p=0.04).
Skoring histopatologis menggunakan skor modifikasi Mankin menunjukan pada
kelompok dengan hormon pertumbuhan memiliki angka terendah dibandingkan
grup lainnya (p=0.001), grup kontrol (p=0.001), grup asam hyaluronat (p=0.015).
Kesimpulan. Suntikan hormon pertumbuhan ke dalam sendi memiliki efektifitas
yang lebih baik dibandingkan dengan Asam hyaluronat pada model osteoarthritis.
Hormon pertumbuhan memberikan harapan baru sebagai alternatif dalam terapi
osteoarthritis.ABSTRACT

for patients all over the globe, with an increasing number of patients. Growth
hormone (GH) works trough direct and indirect effect on cartilage regeneration by
chondroblast stimulation, stimulation of growth factors and morphoangiogenesis
process. Further research is needed to know the effects of intra articular joint
injection of growth hormone using validated animal model and reliable outcome
measurement.
Methods. This study was conducted in Animal Hospital of Agricultural Institute
Bogor west Java, from May to September 2015. The design of the study was
randomized posttest only control group. Male white New Zealand rabbit (n=21)
weighted 1.9-2.6kg, age 6-7months were used in this study. The sample was
randomized and divided into three groups. All groups recieved intra articular
injection of type 2 collagenase (Sigma® Missouri) 2mg at the left knee on day 1
and 4. Injections of growth hormone (4iu), hyaluronic acid (HA) (6mg) and saline
(0.6ml) were done at 2 weeks after collagenase injection once a week for
consecutive 3 weeks. Evaluation of weight and lameness periode is done
periodically, histopathological and macroscopic score were done at 8 weeks since
the first injection.
Result. The lameness priode for control group is significantly longer than both of
the experimental groups (p<0.001), HA (p<0.03), and GH (p<0.001).
Macroscopic score evaluation taken from the lateral condyle of the left femur
showed that the GH group received significantly less cartilage damage than the
HA group (P=0.04) and placebo (P=0.01). Histopathological score was also found
lowest at the GH group (p=0.001), with significant difference in control
(p=0.001), and HA group (p=0.015).).
Conclusion. Intraarticular injection of growth hormone is found to be more
effective compared to hyaluronic acid on rabbit osteoarthritis model. This results
showed promising result for intra articular injection of GH as an alternative

" adalah salah satu penyebab penurunan performa fisik pada seseorang yang melakukan program latihan fisik jangka panjang. Kondisi overtraining dihubungkan dengan gangguan dalam regenerasi sel tubuh. Insulin-like growth factor-I (IGF-I) adalah protein yang menstimulasi pertumbuhan dan proliferasi sel. IGF-I bekerja dalam regulasi aksis GH/IGF, dimana kerja IGF dipengaruhi oleh growth hormone (GH) dan insulin-like growth factor binding protein-3 (IGFBP-3). Penelitian ini bertujuan untuk mengetahui respons tubuh terhadap latihan fisik aerobik overtraining dengan menganalisa kadar GH, IGF-I, dan IGFBP-3, mengingat hormon dan protein ini berperan dalam regenerasi sel tubuh, khususnya otot rangka. Subjek penelitian adalah 19 ekor tikus putih jantan galur Wistar (berusia 8-10 minggu, berat 150-250 gr) yang dibagi menjadi 3 kelompok (satu kelompok kontrol dan dua kelompok yaitu kelompok aerobik dan kelompok aerobik overtraining yang diberikan perlakuan masing-masing selama 11 minggu). Perlakuan latihan fisik aerobik dan aerobik overtraining dilakukan dengan Animal treadmill L-6000 sebanyak lima hari dalam seminggu. Setelah hari terakhir perlakuan, seluruh hewan coba dikorbankan. Serum darah diambil dengan cara pungsi jantung. Kadar GH, IGF-I, dan IGFBP-3 dalam serum diukur dengan metode ELISA dan data hasil penelitian dianalisis dengan one way ANOVA dan dilanjutkan dengan uji Post Hoc. Hasil pemeriksaan ELISA menunjukkan kadar IGFBP-3 yang lebih rendah secara signifikan pada kelompok aerobik overtraining dibandingkan dengan kelompok aerobik, sementara tidak ada perbedaan kadar GH dan IGF-I pada ketiga kelompok. Kadar IGFBP-3 dalam serum dapat dipertimbangkan sebagai penanda biologis kondisi overtraining.

Overtraining is one of the causes of decline in physical performance in long-term physical exercise program. Overtraining is associated with impaired regeneration of body cells. Insulin-like growth factor-I (IGF-I) is a stimulator of cell growth and proliferation. IGF act based on the GH/IGF axis, which mean IGF-I act is regulated by growth hormone (GH) and insulin-like growth factor binding protein-3 (IGFBP-3). This study purpose is to determine the body's response to aerobic overtraining exercise by analyzing the levels of growth hormone (GH), insulin-like growth factor-I (IGF-I), and insulin-like growth factor binding protein-3 (IGFBP-3) as those hormone and proteins play a role in the regeneration of body cells, especially skeletal muscle cells. Subjects were 19 white male rats of the Wistar strain (8-10 weeks old, weight: 150-250 g) were divided into 3 groups (one control group and two groups of aerobic and aerobic overtraining group both were given treatment for 11 weeks). Aerobic exercise and aerobic overtraining exercise treatment were conducted five days a week using Animal treadmill L-6000. After the last day of treatment, all experimental animals were sacrificed. Blood serum collected by cardiac puncture. Levels of GH, IGF-I and IGFBP-3 in serum were measured by ELISA. The data were analyzed by one-way ANOVA followed by post hoc test. ELISA results showed significant lower levels of IGFBP-3 in aerobic overtraining group compared to the aerobic group, while there was no difference in the levels of GH and IGF-I in all groups. IGFBP-3 levels in the serum may be a suitable as biological markers for overtraining condition."

"ABSTRACT
Stunting or short stature in children is a significant nutritional problem in developing and underdeveloped countries. Stunting during childhood might affect brain development and impair development cognitive function. Additionally, this condition associated with the increased risk for obesity during adulthood. Several studies have shown that the increment risk of obesity and overweight in children with a short stature was due to their metabolic efficiency. Children with stunting have lower resting energy expenditure compared to non stunting children. Additionally, stunted children has higher respiratory quotient and carbohydrate oxidation but lower fat oxidation compared to non-stunting children. These results might explain why stunted children easily become obese, which is due to lower fat oxidation and leading to tendency to store fat.
This review discussed the current status on studies in the nutrigenetic aspects of the relationship between stunting in the childhood and obesity in adulthood. I hypothesized that stunted children are more likely to become obese in their later life because they have lower metabolic rate and higher tendency of fat storage. There are several candidate genes and pathway involved in obesity and I suspected that ghrelin and its receptor growth hormone secretague receptor (GHSR) were responsible."

"Gen tilapia Growth Hormone (tiGH) merupakan gen pengkode hormon pertumbuhan dari ikan nila yang berperan untuk meningkatkan pertumbuhan. Penelitian bertujuan melakukan kloning dan ekspresi gen tiGH untuk memproduksi protein rekombinan hormon pertumbuhan ikan nila (Oreochromis niloticus). Penelitian meliputi tahapan isolasi gen tiGH dari pMBA_tiGH, ligasi ke dalam pETBlue-2, serta transformasi vektor rekombinan ke dalam sel inang dengan menggunakan elektroporasi.
Hasil penelitian menunjukkan bahwa vektor rekombinan dapat ditransformasi ke dalam sel inang E. coli BL2 dengan efisiensi transformasi 1,12x103 cfu/μg. Ekspresi gen tiGH dilakukan menggunakan induksi IPTG 0,4 mM dan dipurifikasi menjadi protein rekombinan growth hormone dengan berat molekul sebesar 22 kDa.

Tilapia growth hormone gene (tiGH) is a gene encoding growth hormone from the tilapia whose folr is to increase the growth. The research objective is to do cloning and expression tiGH gene to produce growth hormone recombinant proteins of tilapia (Oreochromis niloticus). Stages of research include isolation tiGH gene from pMBA_tiGH, ligation into pETBlue-2, and the transformation recombinant vector into host cells by using electroporation.
The result showed that recombinant vectors have been successfully transformed into the host cell E.coli BL21with transformation efficiency reached 1.12 x103 cfu/μg. Expression tiGH gene performed using 0.4 mM IPTG induction and purified recombinant protein growth hormone with a molecular weight of 22 kDa.
