Penelitian keragaman genetik udang mantis di Perairan Pelabuhan ratu dan Cirebon telah dilakukan pada bulan Februari ndash; November 2016. Penelitian ini bertujuan untuk mengetahui keragaman genetik udang mantis di perairan Pelabuhan ratu dan Cirebon. Identifikasi udang mantis menggunakan karakter morfologi dan DNA barcoding dengan menggunakan Cytochrome oksidase sub unit I COI . Analisis karakter morfologi menggunakan software PAST v.3.14 Paleontological Statistics dengan metode cluster. Rekonstruksi pohon filogenetik menggunakan software MEGA 6 dengan metode Neighbour Joining berdasarkan model Tamura-3 paramater dengan bootstrap 1000 kali. Hasil penelitian menunjukkan bahwa stomatopoda yang ditemukan dari lokasi pengambilan sampel terdiri atas Harpiosquilla harpax, Oratosquilla oratoria, Oratosquillina gravieri dan Harpiosquilla annandalei. Rata-rata kelimpahan larva Stomatopoda di perairan Cirebon pada stasiun I, II, III dan IV masing-masing 0,047; 0,018; 0,003 dan 0,003 ind/m3sedangkan larva di perairan Pelabuhan ratu hanya ditemukan di stasiun IV sebanyak 0,003 ind/m3. Hasil dendogram karakter morfometrik terdiri atas tiga kelompok, yaitu kelompok H. harpax Cirebon - H.harpax Pelabuhan ratu , kelompok O. oratoria-H. annandalei, dan kelompok O. gravieri. Kesamaan pada kelompok H. harpax dari Cirebon dan Pelabuhan ratu sebesar 94,5 sedangkan H. annandalei ndash; O.oratoria sebesar 92,5 . Hasil rekonstruksi filogenetik yang dibentuk berdasarkan sekuen yang sudah dicocokkan pada Gene bank yaitu terdiri atas 2 genus yaitu Harpiosquilla dan Oratosquilla. ...... The study of genetic diversity mantis shrimp in the Pelabuhan Ratu and Cirebon waters was conducted in February November 2016. This study aimed to determine the genetic diversity of the mantis shrimp in the Pelabuhan Ratu and Cirebon waters. Mantis shrimp was identified using morphological characters and DNA barcoding used Cytochrome Oxidase subunit I COI . Analysis character morphological were done using PAST software v.3.14 Paleontological Statistics cluster method. Reconstruction of the phylogenetic tree used MEGA software 6 with Neighbour Joining method based on the model of Tamura 3 parameters by bootstrapping 1000 times. The results showed that stomatopods found from sampling sites consist of Harpiosquilla harpax, Oratosquilla oratoria, Oratosquillina gravieri and Harpiosquilla annandalei. The average abundance of larvae stomatopoda were found in Cirebon waters at station I, II, III and IV 0,047 0,018 0.003 and 0.003 ind m3, respectively, while in the Pelabuhan ratu water fourth station were found as much as 0,003 ind m3. Dendogram of morphometric character consists of three groups, namely H. harpax Cirebon H. harpax Pelabuhan ratu , O. Oratoria H. annandalei group, and the group O. gravieri. Similarities were found H.harpax group of Cirebon and Pelabuhan Ratu as much as 94.5 while H. annandalei O. oratoria was 92.5 . The results of phylogenetic reconstruction were formed by sequences that have been matched in the Gene bank which consists of two genera, Harpiosquilla and Oratosquilla.

Penelitian mengenai kajian keragaman genetik, aspek pertumbuhan dan reproduksi udang mantis di perairan Teluk Banten Serang Propinsi Banten sebagai upaya konservasi dalam pengelolaan sumber daya udang mantis, khususnya Harpiosquilla raphidea telah dilakukan dari bulan Desember 2011 sampai dengan September 2013. Penelitian bertujuan untuk mengkaji dan mengetahui aspekaspek biologi populasi (keragaman genetik, aspek pertumbuhan dan reproduksi). Penelitian dilakukan mengunakan metode survei dan purposive sampling. Lokasi pengambilan sampel udang mantis Harpiosquilla raphidea di perairan Teluk Banten (Provinsi Banten). Hasil Penelitian menunjukkan bahwa keragaman morfologi udang mantis (H. raphidea) yang terekspresi dari perbedaan karakter ukuran tubuh, dari 22 karakter morfologi 20 karakter morfologi berbeda sangat nyata sedangkan 2 karakter lainnya tidak berbeda nyata yaitu lebar abdomen ruas kelima (ASL) dan ruas keenam (ASN). Analisis sekuens nukleotida daerah COI mtDNA udang mantis H. raphidea diperoleh sekuens udang mantis H. raphidea asal Indonesia yang saat ini belum ada di Gene Bank. Pola pertumbuhan udang mantis relatif sama antara jantan dan betina Allometrik (-). Laju pertumbuhan udang mantis Harpiosquilla raphidea tidak jauh berbeda dengan udang mantis lainnya dari Ordo Stomatopoda. Rentang hidup udang mantis di Teluk Banten relatif lebih panjang (5,6 -- 6,7 tahun), dengan nilai pertumbuhan (K) udang mantis jantan lebih tinggi dibandingkan betina. Faktor kondisi udang betina lebih tinggi dari pada jantan dengan nilai pertumbuhan (L¥) antara jantan dan betina pada kisaran yang sama adalah sebesar 369,08 mm. Tingkat kematangan gonad (TKG) udang mantis H. raphidea terindentifikasi dalam empat tingkat perkembangan. Ukuran matang gonad pertama udang mantis betina 199 mm dan udang mantis jantan adalah 230 mm. Udang mantis betina pada TKG II, III dan IV memiliki fekunditas berkisar 24,600--76,809 dengan rata-rata fekunditas 54,082±13,456 butir, sedangkan diameter telur pada TKG III, 510 ?675 μm dengan rata-rata diameter 574 ±6.98 μm. Pengelolaan sumber daya udang mantis di Teluk Banten diarahkan pada kegiatan perbaikan populasi melalui kegiatan konservasi atau upaya domestikasi. Upaya konservasi polulasi melalui pengendalian penangkapan berdasarkan ukuran tangkapan dengan perbaikan habitat serta upaya domestikasi. Populasi Teluk Banten dapat dijadikan sebagai sumber induk dan dapat disilangkan dengan populasi Jambi sebagai selective breeding. ...... Genes related to starch synthesis and the metabolism contribute to a variety of physicochemical properties that determine the eating/cooking qualities of rice. Our previous study suggested that a set of molecular markers was able to estimate the eating quality of japonica rice. The present study reports the genetic diversity of 22 japonica rice varieties based on markers corresponding to starch synthesizing genes. The mean of the polymorphic information content (PIC: 0.135) value and the diversity index (0.171) indicated a low genetic diversity in these varieties. The phylogenetic tree clearly demonstrated three main clusters: 1) cluster I contained seven varieties with similar physicochemical properties; 2) cluster II only showed a Japanese variety, Koshihikari, and 3) cluster III included the most Korean varieties (14 varieties). This phylogenetic analysis did not completely represent the physicochemical properties differentiation of the japonica varieties, although it did reveal an initial clue to the close relationship between Korean rice and the Japanese and Chinese varieties. Notably, these markers were also able to identify a premium japonica rice. The molecular markers and information concerning the genetic relationship would be useful in improving the japonica rice along with its starch quality of in breeding program.

Keragaman Genetik Padi Japonica berdasarkan Marka terkait Gen Sintesis Pati. Gen terkait sintesis dan metabolism pati berkontribusi pada berbagai sifat fisiko-kimia yang menentukan mutu rasa dan hasil masak (cooking) beras. Penelitian sebelumnya menunjukkan bahwa satu set marka molekuler mampu memprediksi mutu rasa beras japonica. Pada studi ini dilaporka keragaman genetik 22 varietas padi japonica berdasarkan marka untuk gen-gen pensintesis pati. Rata-rata nilai polymorphic information content (PIC:0,135) dan indeks keragaman (0,171) menunjukkan keragaman genetik yang rendah dalam varietas padi ini. Pohon filogenetik menunjukkan tiga kelompok utama yang dibentuk: 1) klaster I terdiri dari tujuh varietas dengan sifat fisikokimia yang mirip; 2) klaster II hanya terdiri dari verietas premium Jepang, Koshihikari, dan 3) klaster III mengelompokkan sebagian besar varietas Korea (14 varietas). Analisis filogenetik ini belum sepenuhnya menggambarkan diferensiasi varietas japonica berdasarkan sifat fisiko-kimia, namun hasil ini mengungkapkan petunjuk awal korelasi yang erat antara padi Korea dengan varietas Jepang dan Cina. Marka-marka tersebut juga mampu mengidentifikasi beras premium japonica. Marka molekuler dan informasi kekerabatan genetik ini akan berguna dalam membantu mengembangkan padi japonica terkait dengan mutu pati dalam program pemuliaan

Penelitian ini bertujuan untuk mengetahui keragaman genetik dan kekerabatan ikan cupang menggunakan sekuens Cytochrome Oxidase I. Keragaman genetik diketahui menggunakan sekuens Cytochrome Oxidase I. Amplifikasi Cytochrome Oxidase I dilakukan menggunakan primer HCO dan LCO dengan hasil sekuens yang diperoleh berukuran 680 pb. Hasil alignment sekuens menunjukkan keragaman genetik yang kurang signifikan pada B. imbellis, sedangkan pada B. pallifina, B. patoti, dan B. unimaculata menunjukkan keragaman yang cukup signifikan. Tiga spesies ikan cupang dari Kalimantan memiliki kekerabatan yang dekat. Namun demikian, kekerabatan ikan cupang dari Kalimantan dengan ikan cupang dari Sumatera cukup jauh. ...... This study aims to determine the genetic diversity and kinship of fighting fish using Cytochrome Oxidase I. Cytochrome Oxidase I is used to know the genetic diversity of fighting fish. Primer HCO and LCO were used to amplify 680 bp of Cytochrome Oxidase I. The results of sequence alignment showed less significant genetic diversity for B. imbellis, whereas B. pallifina, B. patoti, and B. unimaculata show significant diversity. Three species of fighting fish from Kalimantan are closely related. However, the kinship of fighting fish from Kalimantan with fighting fish from Sumatra are far enough.

Studi keragaman genetik dilakukan untuk mengetahui hubungan kekerabatan bakteri asam laktat indigenos Indonesia yang memiliki kemampuan resistansi terhadap chloramphenicol dan erythromycin. Hasil uji resistansi menunjukkan bahwa isolat DH1, DH7, dan S34 resistan terhadap chloramphenicol (5 μg/ml), sedangkan isolat T8 resistan terhadap erythromycin (15 μg/ml). Isolat D2, S23, dan T8 diketahui resistan terhadap kombinasi chloramphenicol dan erythromycin (1 μg/ml). Analisis BLAST menunjukkan bahwa isolat bakteri asam laktat terdiri atas Lactobacillus plantarum, L. fermentum, Pediococcus acidilactici, dan P. pentosaceus. Analisis pohon filogenetik diketahui bahwa isolat D2, S12, S34, T3, dan T8 memiliki kekerabatan yang dekat dengan L. plantarum. Isolat R31 dan DH1 memiliki kekerabatan yang dekat dengan L. fermentum. Isolat LK14, S23, R24, DH7,DS13, GR3, HB3 memiliki kekerabatan yang dekat dengan genus Pediococcus.
Study on genetic diversity was useful to determine the kinship of Indigenous Indonesia lactic acid bacteria which have the capability of resistance to chloramphenicol and erythromycin. The resistance test showed isolates DH1, DH7, and S34 that were resistant to chloramphenicol (5 μg/ml), whereas T8 was resistant to erythromycin (15 μg/ml). Isolates D2, S23, and T8 were remain resistant to the combination of chloramphenicol and erythromycin (1 μg/ml). The results of BLAST analysis showed that there were four different species of lactic acid bacteria, such as Lactobacillus plantarum, L. fermentum, Pediococcus acidilactici, and P. pentosaceus. The results of phylogenetic trees analysis showed that isolates D2, S12, S34, T3, and T8 have a close kinship with L. plantarum, whereas isolates R31 and DH1 have a close kinship with L. fermentum. Moreover, isolates LK14, S23, R24, DH7, DS13, GR3, HB3 have a close kinship to the genus Pediococcus.
