"Latar Belakang: Ketika perawatan saluran akar gagal, perawatan ulang saluran akar nonbedah, sedapat mungkin, adalah pilihan klinis pertama. Material pengisi saluran akar sebelumnya harus dibuang untuk kemudian dilakukan desinfeksi dan pengisian ulang saluran akar. Terdapat berbagai teknik pembuangan material pengisi, termasuk metode heat carrier, instrumen manual, putar, ultrasonik dan pelarut, atau kombinasi. Penghilangan sepenuhnya material pengisi tidak dapat dilakukan dengan hampir sebagian besar material yang tersisa dari perawatan ulang adalah siler. Siler biokeramik berikatan secara kimiawi dengan membentuk hidroksiapatit dan berpenetrasi ke tubulus dentin untuk menciptakan ikatan mikromekanis. Hal ini membuat pembersihan siler biokeramik sulit dilakukan. Tujuan: Mengevaluasi kebersihan saluran akar dengan berbagai metode pembuangan siler biokeramik menggunakan instrumen putar, kombinasi instrumen putarultrasonik, dan kombinasi instrumen putar-heat carrier pasca obturasi dengan teknik hidraulik dan WVC, yang dianalisis menggunakan Micro-CT. Metode: Empat puluh dua sampel gigi premolar rahang bawah pasca ekstraksi dengan akar lurus dan saluran akar tunggal yang diobturasi dengan siler biokeramik AH Plus® Bioceramic (Dentsply, USA) meggunakan dua teknik, hidraulik dan WVC. Pembuangan material pengisi menggunakan instrumen putar, kombinasi instrumen putar-ultrasonik, dan kombinasi instrumen putar-heat carrier. Pemindaian micro-CT setelah obturasi dan setelah pembuangan material dan dianalisis menggunakan perangkat lunak CTAn. Hasil: Terdapat perbedaan persentase kebersihan saluran akar (p<0,05) yang bermakna antara metode instrumen putar, kombinasi instrumen putar-ultrasonik, dan kombinasi instrumen putar-heat carrier. Kesimpulan: Pembuangan siler biokeramik menggunakan kombinasi instrumen putar-ultrasonik lebih efektif dibandingkan metode instrumen putar dan metode kombinasi instrumen putar-heat carrier. Teknik obturasi tidak mempengaruhi kebersihan saluran akar.

Background: When root canal treatment fails, non-surgical retreatment, if possible, is the first clinical choice. Previous root canal filling materials must be removed for subsequent disinfection and obturation of the root canal. There are various techniques for removing filling materials, including heat carrier methods, manual instruments, rotary, ultrasonic and solvent, or combinations thereof. Complete removal of filling materials cannot be achieved with almost all remaining materials from retreatment being sealer. Bioceramic sealers chemically bond by forming hydroxyapatite and penetrate into dentin tubules to create micromechanical bonds. This makes cleaning of bioceramic sealer difficult. Objective: To evaluate the cleanliness of root canal walls in retreatment with various methods of removing bioceramic sealer using rotary instruments, combination of rotary-ultrasonic instruments, and combination of rotary-heat carrier instruments postobturation with hydraulic and WVC techniques, analyzed using Micro-CT. Methods: Forty-two extracted mandibular premolar samples with straight roots and single root canals obturated with AH Plus® Bioceramic sealer (Dentsply, USA) using two techniques, hydraulic and WVC. Removal of filling material using rotary instruments, combination of rotary-ultrasonic instruments, and combination of rotary-heat carrier instruments. Micro-CT scanning after obturation and after removal of material and analyzed using CTAn software. Results: There is a significant difference (p<0.05) in the percentage of root canal cleanliness between the rotary instrument, the combination with ultrasonic, and the combination with heat carrier.Conclusion: Bioceramic sealer removal methods using a combination of rotary-ultrasonic instruments are more effective than rotary instrument methods and combination of rotary-heat carrier methods. The obturation technique does not affect the cleanliness of root canal."

"Latar belakang: Penglepasan ion kalsium oleh material bioaktif dapat berperan penting dalam peningkatan pH yang diperlukan dalam aktivitas antibakteri dan remineralisasi jaringan keras gigi.
Tujuan: untuk menganalisis pelepasan ion kalsium dan peningkatan pH dari MTA modifikasi dan Bioceramic pada periode waktu 1,48,168 jam.
Metode: Sampel n=30 dipersiapkan dengan ukuran diameter 3 mm tinggi 3 mm, terdiri dari 15 sampel MTA modifikasi, 15 sampel Bioceramic direndam dalam air deionisasi 1,48,168 jam diukur kadar pelepasan ion kalsium menggunakan AAS dan nilai pH menggunakan pHmeter, Uji Kruskal Wallis dan Mann Whitney.
Hasil: Terdapat perbedaan yang bermakna diantara semua kelompok dengan nilai signifikansi p le;0,05.
Kesimpulan: Bioceramic terbukti melepaskan ion kalsium dan peningkatan pH lebih besar dibandingkan dengan MTA modifikasi pada waktu pengukuran 1,48,168

Background: Calcium ion release can promote alkalinizing activity and regeneration.
Objective: To analyze calcium ion release and pH changes from modified MTA and Bioceramics as bioactive material.
Methods: 30 samples are prepared with the size of 3 mm in diameter and 3 mm in height. The samples are consist of 15 of modified MTA and 15 of bioceramics. And then immersed in deionized water for an hour which will then be measured in 1, 48, and 168 hours period. And measured atom absorption sphectropometer and pHmeter.
Result: Mann Whitney post hoc rsquo s statistic test result showed a significant discrepancy among all groups, with the significant value of p le 0,05.
Conclusion: Bioceramics was proven to release more calcium ions and more pH elevation compared to modified MTA during the 1 hour, 48 hour, and 168 hours measurements.

Latar belakang: Siler dapat meningkatkan sealing ability Tujuan: Menganalisis dan membandingkan sealing ability pengisian saluran akar dengan siler bioceramic dan resin metakrilat. Metode: Tiga puluh gigi dengan saluran akar tunggal, dipreparasi dengan ProTaper Next, dibagi dalam dua kelompok secara acak; Masing-masing grup diisi dengan gutaperca berlapis, untuk kelompok 1 menggunakan Siler Bioceramic SB dan kelompok 2 Siler Resin-Metakrilat SRM . Pengukuran kebocoran dengan tinta india pada daerah sepertiga apikal apeks yang dipotong secara melintang, kemudian dilihat dengan mikroskop stereo. Hasil: Kelompok siler bioceramic menunjukkan celah tepi yang lebih sedikit jika dibandingkan dengan siler resin-metakrilat. Kesimpulan: Sealing ability pengisian saluran akar menggunakan siler bioceramic SB lebih baik dibandingkan dengan siler resin-metakrilat SRM .
ABSTRAK
Background Sealer may increase sealing ability. Purpose To compare and analyze sealability of obturation using the bioceramic sealer and methacrylate resin based sealer. Methods thirty single rooted teeth were prepared using ProTaper Next and divided randomly into two groups the first group was obturated with Bioceramic Sealer SB while the second group was obturated with methacrylate resin based sealer SRM . Each group used coated gutta percha as a core material. Apical third marginal adaptation was evaluated by observing the dye penetration between the obturation material and the root canal walls on cross sectioned samples. Results SB Showed the least microleakage in the apical third marginal adaptation, followed by SRM. Conclusion Obturation of root canal using bioceramic sealer has the best sealing ability at the apical third, compared to SRM."

"Latar Belakang: E.faecalis isolat klinis merupakan bakteri yang menyebabkan lesi periapeks persisten. Bioceramic dan Mineral Trioxide Aggregate merupakan siler yang mempunyai sifat anti bakteri.
Tujuan: Untuk menganalisis efek anti bakteri siler Bioceramic dan MTA terhadap E.faecalis isolat klinis pada waktu 2 menit segera , 4 jam initial setting , 1 dan 7 hari setelah pengadukan siler.
Metode: Efek anti bakteri siler Bioceramic dan MTA diperiksa dengan direct contact test. Masing-masing siler dikontakkan langsung dengan E.faecalis isolat klinis 2 menit, 4 jam, 1 dan 7 hari setelah pengadukan siler. Suspensi ini dioleskan pada medium agar dan diinkubasi 24 jam untuk melihat koloni bakteri yang tumbuh CFU/ml.
Hasil: Terdapat perbedaan bermakna antara siler Bioceramic dan MTA pada waktu 7 hari, antara siler Bioceramic pada waktu 2 dan 4 jam serta waktu 4 jam dan 7 hari setelah pengadukan siler.
Kesimpulan: siler Bioceramic dan MTA mempunyai efek anti bakteri yang baik terhadap E.faecalis pada saat segera, initial setting, dan 1 hari setelah pengadukan siler, pada waktu 7 hari setelah pengadukan, siler MTA yang paling baik. Background: E.faecalis isolate clinic is a kind of bacteria that cause persistent periapical lesion. Bioceramic and Mineral Trioxide Aggregate are sealers that having antibacterial properties.
Aim: To analyze antibacterial effect of Bioceramic and MTA sealers against E.faecalis isolate clinic at 2 minutes fresh , 4 hours initial setting , 1 day and 7 day after mixed the sealers.
Methods: Antibacterial effect of Bioceramic and MTA sealers was assessed by direct test contact. Each sealer was contacted with E.faecalis isolate clinic at 2 minutes, 4 hours, 1 day and 7 days after mixed the sealers. This suspension was swab in agar medium and incubated for 24 hours. The colony in agar plates is counted with colony forming unit CFU .
Result: The significant differences was shown by Bioceramic and MTA at 7 days, between Bioceramic at 2 minutes fresh and 4 hours initial setting also at 4 hours initial setting and 7 days after mixed the sealer.
Conclusion: Both of Bioceramic and MTA sealers have a good antibacterial effect at fresh, initial setting and 1 hour after mixed the sealer but at 7 days, MTA was the greater. MTA sealer has a constant antibacterial effect until 7 days.

"ABSTRAK
Latar belakang: Salah satu sifat ideal suatu siler saluran akar adalah memiliki kemampuan penutupan yang baik terutama pada sepertiga apeks. Tujuan: Untuk mengevaluasi perbandingan kebocoran antara pengisian saluran akar dengan siler polidimetilsiloksan SP dan siler bioceramic SB . Metode: Tiga puluh enam gigi premolar rahang bawah, dibagi dua kelompok dan diisi dengan teknik kon tunggal menggunakan siler polidimetilsiloksan pada kelompok pertama, dan siler bioceramic pada kelompok ke-dua. Selanjutnya gigi diinkubasi 37?C, selama 24 jam , kemudian dilapisi dengan dua lapis cat kuku kecuali pada 2 mm dari apeks, dan direndam dalam tinta india 7 X 24 jam . Sampel didekalsifikasi, didehidrasi dan dibuat transparan sesuai dengan metode Robertson. Kedalaman penetrasi zat tinta dievaluasi dengan mikroskop stereo. Skor 0 untuk tidak bocor, skor 1 untuk penetrasi zat tinta le; 0,5 mm, skor 2 untuk penetrasi 0,51 ndash; 1 mm, dan skor 3 untuk penetrasi > 1 mm. Hasil: Distribusi proporsi kebocoran terbesar pada kelompok SP terdapat pada skor 2 44.4 , sedangkan pada kelompok SB terdapat pada skor 1 55.6 . Kesimpulan: Tingkat kebocoran pengisian saluran akar dengan siler bioceramic tidak berbeda dengan siler polidimetilsiloksan.

ABSTRACT
Background One of the ideal properties of a root canal sealer is to have a good sealing ability, especially at the apical third of the root. Objective To evaluate the comparison of the apical leakage between obturation using bioceramic sealer SB and polydimethylsiloxane sealer SP . Methods Thirty six mandibular premolars were equally divided into two groups and was obturated with single cone technique. The sealer used for Group I and Group II were SP and SB respectively. After obturation, the samples were incubated 37 C, 24 h , sealed with two coats of nail polish except for 2 mm from the apex, immersed in the Indian ink for 7 days, decalcified, dehydrated and made transparent according to Robertson technique. Dye penetration were evaluated under stereomicroscope. Samples with no dye penetration were given score 0, le 0,5 mm dye penetration were given score 1, 0,051 ndash 1 mm were given score 2, and 1 mm were given score 3. Result The largest proportion distribution in SP group was at the score 2 44.4 , and in group SB was at the score 1 55.6 . Conclusion Bioceramic sealer showed similar apical leakage to polydimethylsiloxane sealer."

"Latar Belakang: Siler biokeramik berbasis kalsium silikat diketahui memiliki biokompabilitas tinggi, mampu beradhesi dengan dinding saluran akar serta dapat menginduksi respon osteogenik, yang dikategorikan sebagai material biomimetik. Akan tetapi, siler biokeramik memiliki kekurangan pada sifat fisiknya. Kitosan larut air merupakan bentuk modifikasi kitosan yang memiliki keunggulan sebagai antioksidan, antibakteri, berperan dalam penyembuhan lesi dan regenerasi jaringan serta sebagai dapat memperbaiki sifat fisik semen. Perpaduan kedua bahan dapat menciptakan semen saluran akar biomimetik.
Tujuan: Melakukan uji karakteristik, sifat fisik dan biologis terhadap novel siler hibrida biokeramik-kitosan larut air (BCC) sebagai semen biomimetik saluran akar.
Metode: Novel siler hibrida BCC dimanipulasi dari semen biokeramik (BC) yang telah melalui proses ball-milling dan sintering, kemudian dicampur kitosan larut air dengan W/P 0,4. Empat variabel penelitian, yaitu semen biokeramik BC, novel siler hibrida BCC, siler biokeramik dan siler epoksi resin dilakukan uji karakterisasi (XRD, SEM/EDS), uji sifat fisik (setting time, daya alir, film thickness), serta uji biologis (sitotoksisitas dan bioaktivitas terhadap hPDLSCs).
Hasil: Novel siler hibrida BCC mengandung unsur dan puncak kristalin yang serupa dengan siler biokeramik (Sure-Seal Root™), memiliki bentuk partikel yang cenderung globular dan homogen dengan jarak antarpartikel lebih rapat serta ukuran partikel deskriptif yang lebih besar dibandingkan Sure-Seal Root™ dan AH Plus® tetapi lebih kecil dibandingkan semen biokeramik BC. Hanya semen biokeramik BC dan novel siler BCC yang memiliki rasio Ca/Si/P. Novel siler BCC menunjukkan perbedaan setting time, daya alir dan film thickness yang bermakna dengan semen biokeramik BC, Sure-Seal Root™ dan AH Plus®. Keempat kelompok menunjukkan sitotoksisitas rendah terhadap hPDLSCs. Bioaktivitas novel siler BCC relatif lebih tinggi dibandingkan semua kelompok perlakuan dan kontrol.
Kesimpulan: Novel siler hibrida BCC berpotensi memberikan implikasi klinis yang baik, menunjukkan sifat fisik yang mendekati standar semen saluran akar dan memiliki bioaktivitas sebagai semen biomimetik saluran akar.

Background: Currently, bioceramic sealers with calcium silicate based have been developed and show high biocompatibility, are able to adhere to root canal dentin and can induce an osteogenic response, which can be categorized as biomimetic materials. On the other hand, water-soluble particle (WSP) chitosan, one of chitosan derivatives, has advantages as an antioxidant, antibacterial, plays a role in wound healing and tissue regeneration as well as a thickening agent. The combination of these two materials can create a biomimetic endodontic sealer.
Objective: To examine the characteristics, physical and biological properties of novel bioceramic-chitosan hybrid sealer (BCC) as biomimetic endodontic sealer.
Methods: Novel BCC sealer were manipulated from bioceramic cement (BC) which had been synthesized through a ball-milling and sintering process, then mixed with WSP chitosan with a W/P of 0,4. Four variables, namely novel BCC sealer, BC bioceramic cement, bioceramic and epoxy resin sealer were tested characterization (XRD, SEM/EDS), physical properties (setting time, flow, film thickness), and biological tests (cytotoxicity and bioactivity on hPDLSCs).
Results: Novel BCC sealer showed bioceramic elements and crystalline peaks similar to bioceramic sealer (Sure-Seal Root™), had a particle shape that tends to be globular and homogeneous with descriptive particle size larger than Sure-Seal Root™ and AH Plus® but smaller compared to BC bioceramic cement. Only BC bioceramic cements and novel BCC sealer had Ca/Si/P ratios. Novel BCC sealer showed significant differences in setting time, flow, and film thickness with BC bioceramic cements, Sure-Seal Root™ and AH Plus®. The four groups showed low cytotoxicity to hPDLSCs. The bioactivity of novel BCC sealer was relatively higher among other groups and control group.
Conclusion: Novel BCC hybrid sealer has good clinical implications, exhibit physical properties close to standard root canal cements and have bioactivity as root canal biomimetic sealer.

"Latar Belakang: Siler saluran akar berfungsi untuk mengisi ruang antara gutaperca dengan dinding saluran akar dan harus bersifat biokompatibel terhadap jaringan periapeks. Siler saluran akar merupakan bahan kimia yang berpotensi menyebabkan mutasi yang dapat dilihat dari ekspresi protein sel tersebut. Tujuan: Mengetahui dan membandingkan potensi mutagenitas siler resin, silikon, dan biokeramik terhadap perubahan ekspresi protein sel limfosit manusia. Metode: Sembilan sampel dari setiap kelompok siler sebanyak 2 ml yang terdiri atas bahan siler dan darah diinkubasi selama 1, 3 dan 7 hari. Kemudian dilakukan isolasi sel limfosit dan pemisahan protein dengan metode elektroforesis. Profil pita protein diobservasi dan data dianalisis secara statistik dengan Kruskal-Wallis dan post-hoc Mann-Whitney. Hasil: Tidak terdapat perbedaan bermakna secara statistik terhadap pembentukan pita protein antara ketiga bahan siler berbahan dasar resin, silikon dan biokeramik. Namun, terdapat perbedaan bermakna antara kelompok siler resin dan silikon pada hari pertama dan ketiga, dan antara kelompok siler silikon dan biokeramik pada hari pertama. Kesimpulan: Terdapat perbedaan potensi mutagenik pada hari pertama, siler resin lebih berpotensi mutagenik diikuti oleh biokeramik kemudian siler silikon. Pada hari ketiga, biokeramik lebih berpotensi mutagenik diikuti oleh resin kemudian silikon. Pada hari ketujuh, biokeramik lebih berpotensi mutagenik diikuti oleh resin dan silikon.
.....Background: Root canal sealers serves to fill the space between the gutta percha and canal wall must be biocompatible with periapical tissue. Root canal sealers are chemicals agent that potentially cause mutations that can be seen from the protein expression of the cells. Objective: To know and compare the potential mutagenicity of resin, silicone, and bioceramic sealers on expression of proteins of human lymphocyte cells. Methods: Nine samples from each group sealer as much as 2 ml of blood are incubated for 1, 3 and 7 days. Then the isolated lymphocytes are observed for protein separation by electrophoresis method. Profile of protein bands observed and data were analyzed statistically by Kruskal-Wallis and post-hoc Mann-Whitney. Results: there is no statistically differences in the formation of protein bands among the resin, silicone and bioceramic sealers. However, there is a statistically differences between the resin and silicone on the first and third, and between silicone and bioceramic on the first day. Conclusion: There were differences in the potential mutagenicity on the first day, resin is more potentially mutagenic followed by bioceramic then silicone. On the third day, bioceramic is more potentially mutagenic followed by resin then silicone. On the seventh day, bioceramic is more potentially mutagenic followed by resin and silicone sealers."
