Infeksi dengue masih merupakan masalah kesehatan di Indonesia sebagai negara endemik. Sampai saat ini, belum ditemukan terapi definitif sebagai tatalaksana infeksi dengue. Penelitian untuk pengembangan antivirus dengue sudah banyak dilakukan. Ekstrak Calophyllum canum, sebagai salah satu tanaman yang tumbuh di Kalimantan, ditemukan memiliki efek anti mikroganisme. Penelitian dilakukan untuk menguji efek antiviral dari crude extract batang C. canum fraksti etil asetat terhadap replikasi virus dengue serotipe-2 (DENV-2) pada sel Huh7it-1 dengan focus assay dan MTT assay untuk menguji sitotoksitas pada sel tidak terinfeksi. Nilai IC50 dari ekstrak C. canum pada sel Huh7it-1 adalah 123,96 μg/mL dan nilai CC50 adalah 620,57 μg/mL. Indeks selektivitas adalah 5,01. Pada focus assay terdapat perbedaan bermakna antara tiap perlakuan dengan kontrol (p<0,001) namun pada MTT assay tidak terdapat perbedaan bermakna antara tiap perlakuan dengan kontrol (p≥0,05). Crude extract dari batang C. canum memiliki efek inhibisi terhadap replikasi DENV-2 secara in Vitro pada sel Huh7it-1. ...... Until now, dengue infection is still a health concern in Indonesia. There is no definite treatment for dengue infection. The development of dengue antivirus using natural extract of plant have been conducted. Calophyllum canum, a plant that grow in Kalimantan was discovered to have anti-microorganism effect. In present study, crude extract of C. canum stem in ethyl acetate fraction was used to examine the antiviral effect on dengue virus seroptype-2 (DENV-2) replication on Huh7it-1. The inhibition of DENV-2 replication was determined by focus assay. The cytotoxicity of uninfected cells by MTT assay. The IC50 value of C. canum on Huh7it-1 cells was 123.96 μg/mL and the CC50 value was 620.57 μg/mL. The selectivity index was 5.01. There was significant difference on focus assay between all concentration with control (p<0.001), but there was no significant difference in MTT assay (p≥0.05). Crude extract of C. canum stem showed inhibition effect on DENV-2 replication in vitro on Huh7it-1 cells.

ABSTRACT
Dengue Hemorrhagic Fever (DHF) incidence in Indonesia can be classified as high, as the number of cases at the year of 2013 reached 44 cases and continues to rise. Until now there has no available specific antiviral towards dengue virus (DENV) yet. The aim of this research is to assess effect of isoamyl gallate as gallic acid derivate towards DENV-2 NGC replication in in vitro assessment. Huh-7 cells were cultured in 48 well plate and infected with DENV-2 NGC and combined with various concentration of isoamyl gallate. The CC50 was measured by MTT assay and IC50 was measured by focus assay. The significance of the data taken from each concentration was compared using Kruskall-Wallis and Mann-Whitney test. The result of the experiment showed the value of CC50, IC50, and SI were >80μg/mL, 6.2μg/mL, and 72.8 respectively. Kruskall-Wallis test showed that there was significant difference between each treatment using different concentration. However, for Mann-Whitney test, insignificant difference towards the control was shown in the concentration of 2.5μg/mL. IC50 value support the statement that isoamyl gallate will cause inhibition of DENV-2 NGC replication. With high value of SI, it can be concluded that isoamyl gallate could become an anti-DENV.

Kejadian demam berdarah dengue (DBD) di Indonesia tergolong tinggi, ditunjukkan dengan tingginya jumlah kasus pada tahun 2013 mencapai 44 kasus dan terus meningkat. Masalah yang dihadapi terkait terus berlanjutnya kejadian DBD adalah belum ditemukannya antivirus spesifik terhadap DENV. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh isoamil galat sebagai turunan asam gallat terhadap replikasi DENV-2 NGC secara in vitro. Sel Huh-7 dikultur dalam 48 well plate dan ditambahkan campuran DENV-2 NGC dan ekstrak isoamil galat. Nilai CC50 diukur dengan MTT assay sedangkan nilai IC50 diukur dengan uji fokus. Perbedaan yang signifikan antar data yang diambil dari masing-masing konsentrasi dibandingkan dengan uji Kruskall-Wallis dan Mann-Whitney. Hasil penelitian menunjukkan nilai CC50, IC50, dan SI adalah >80μg/mL, 6.2μg/mL, and 72.8 secara berurutan. Uji Kruskall-Wallis menunjukkan bahwa ada perbedaan yang signifikan (p value<0.05) antara masing-masing perlakuan dengan menggunakan konsentrasi yang berbeda. Namun, untuk uji Mann-Whitney, perbedaan yang tidak signifikan terhadap kontrol ditunjukkan pada konsentrasi 2,5μg/mL. Nilai IC50 yang rendah dan CC50 yang tinggi memnunjukkan bahwa Isoamil galat akan menyebabkan penghambatan replikasi DENV-2 NGC dan tidak toksik pada sel. Nilai SI didapati secara signifikan tinggi (SI>10) sehingga ada kemungkinan bahwa Isoamil galat bisa menjadi anti-DENV

Pertukaran data secara peer-to-peer telah berhasil mendapatkan popularitas dengan cepat terutama di industri pengiriman media digital seperti musik dan video. Beberapa penelitian menunjukkan bahwa penggunaan bandwidth untuk jaringan peer-to-peer mencapai 60% dari total lalu lintas internet. Meningkatnya penggunaan aplikasi peer-to-peer, mengakibatkan dependensi antara peer di dalam sistem. Satu peer saja yang hilang atau mati bisa mengakibatkan kegagalan bagi peer-peer lainnya karena data yang ada di peer tersebut ikut hilang. Penelitian ini bertujuan mengembangkan strategi replikasi dan pencarian dokumen yang memanfaatkan konsep lokalitas dan popularitas dokumen. Dokumen-dokumen populer yang dimiliki oleh suatu peer dibuat replikanya ke peer lain yang memiliki kemiripan koleksi dengan peer tersebut. Penulis melaksanakan simulasi untuk menguji strategi yang diusulkan dengan memodifikasi simulator untuk locality-aware peer-to-peer system yang telah dikembangkan sebelumnya oleh peneliti lain (Hamzah, 2007). Hasil penelitian menunjukkan bahwa strategi yang diusulkan penulis menunjukkan peningkatan performa dibandingkan dengan strategi lain yang merepresentasikan bentuk default dari strategi server-side replication dan client-side replication. Penelitian juga menunjukkan bahwa strategi ini menunjukkan performa yang baik di situasi dimana sebaran query lebih diutamakan untuk dokumen yang populer dan sesuai dengan specialty dari peer yang menginisiasi query. ......Peer-to-peer system has achieved a huge popularity, especially in the digital media industry. Research has shown that 60% of the bandwidth usage in the internet is accumulated by peer-to-peer system. With the increasing usage and demand of peer-to-peer system, comes a high dependency between user or peer in the system. Even one dead peer could cause a failure to many other peers because the dead peer took along all of its data with it. This research aims to develop a search and replication strategy that utilize the concept of locality and document popularity. A popular document that is owned by a peer will be replicated to other peer that share some similarity with the original peer's document collection. A simulation is done by modifying an existing simulator made by another researcher in the field of locality-aware peer-to-peer system (Hamzah, 2007). The result is that the strategy proposed by this research have shown improvement in performance compared to the other strategy that is supposed to represent server-side replication and client-side replication in its default form. This research has also shown that the strategy proposed performed better in a situation where the query prioritize documents with better popularity and have the same topic as the peer's specialty.

Demam berdarah dengue (DBD) merupakan masalah global yang disebabkan oleh infeksi virus dengue (DENV) dengan vektor nyamuk Aedes spp. Dengan nilai selectivity index (SI) sebesar 128,400, senyawa murni katekin memiliki potensi dalam menghambat replikasi DENV dan dalam pencarian penobat antivirus yang spesifik dan efektif untuk mengobati infeksi DENV. Penelitian ini dilakukan secara eksperimental untuk mengetahui perbandingan aktivitas senyawa murni katekin ketika menempel pada reseptor sel Vero dan menempel pada DENV. Senyawa katekin digunakan dengan konsentrasi sebesar dua kali IC50, yaitu 42,914 μg/ml. Efektivitas inhibisi dan viabilitas sel dievaluasi dengan metode focus assay dan MTT assay. Pada penghambatan reseptor sel Vero, didapatkan persentase efektivitas inhibisi sebesar 46,31±11,39% dengan viabilitas sel sebesar 103,5±2,51%. Pada penempelan DENV, persentase efektivitas inhibisi sebesar 54,67±18,01% dengan viabilitas sel sebesar 84,73±13,05%. Pemberian senyawa murni katekin pada reseptor sel Vero tidak berbeda bermakna secara statistik dengan penempelan DENV dalam menginhibisi virus.  Akan tetapi, penempelan DENV memilki toksisitas yang berbeda bermakna dengan penghambatan reseptor sel Vero. Dengan efektivitas dan toksisitas yang baik, senyawa murni katekin berpotensi sebagai antivirus DENV di masa depan. Penelitian lebih lanjut diperlukan untuk menjaga bioavailabilitas katekin dan juga mentukan target spesifik dari senyawa katekin agar efektif dan tetap aman saat dikonsumsi dalam tubuh.

 


Dengue hemorrhagic fever (DHF) is a global problem caused by dengue virus infection with Aedes spp. as its vector. With selectivity index (SI) value of 128,400, pure compounds of catechins have the potential to inhibit DENV replication in the search for specific antiviral treatments that are effective for treating DENV infection.  This research was carried out experimentally to determine the comparison of the activity of pure catechin compounds when attached to Vero cell receptors and attached to DENV. The catechin compound has a concentration of twice the IC50, which is 42,914 μg/ml with Vero cell as the experiment medium. Inhibition efectivity and cell viability are evaluated by focus assay and MTT assay methods. In inhibition of Vero cell receptors, the percentage of inhibition effectiveness was 46,31±11,39% with cell viability of 103.5±11,39%. In the DENV envelope attachment, the percentage of inhibition effectiveness was 54,67±18,01% with cell viability of 84,73±13,05%. Statistically, administration of pure catechin compounds to Vero cell receptors did not differ significantly from the attachment of DENV in inhibiting the virus. However, the attachment of DENV has a significantly different toxicity from the inhibition of Vero cell receptors. With good effectiveness and toxicity, pure catechin compounds have the potential to be a prophylactic antiviral DENV in the future. Further research is needed to maintain the bioavailability of catechins and also determine specific targets of catechin compounds to be effective and safe when consumed in the body.

ABSTRAK
Infeksi virus dengue DENV masih menjadi masalah serius baik di dunia maupun di Indonesia. Spektrum klinis dengue yang terdiri atas demam berdarah dengue DBD dan sindrom syok dengue SSD menyebabkan kematian. Terapi definitif belum tersedia saat ini, sehingga masih mengandalkan terapi suportif. Berbagai tanaman sebagai antivirus telah banyak dieksplorasi. Salah satu tanaman diketahui memiliki efek antivirus berasal genus Calophyllum. Penelitian ini bertujuan untuk menganalisis potensi inhibisi replikasi virus dengue serotipe 2 oleh fraksi air Calophyllum nodosum pada sel Huh7it. Dua metode yang digunakan antara lain MTT 3- 4, 5-dimethylthiazolyl-2 -2, 5-diphenyltetrazolium bromide assay dan focus assay. MTT assay menilai viabilitas sel pada sel yang tidak terinfeksi, sedangkan focus assay menilai titer virus yang menggambarkan hambatan infektivitas DENV. Hasil menunjukkan antara perlakuan dan kontrol pada kelompok viabilitas tidak berbeda bermakna dengan nilai half cytotoxic concentration CC50 2729 g/mL, sedangkan perbedaan bermakna ditemukan pada kelompok infektivitas dengan nilai half inhibitory concentration IC50 10,87 g/mL. Indeks Selektivitas IS yang didapatkan sebesar 251,06. Fraksi air Calophyllum nodosum berpotensi menghambat replikasi DENV-2 secara in vitro.
ABSTRACT
Infection by Dengue virus DENV is still a serious problem around the world including in Indonesia. Clinical Dengue spectrum from bleeding dengue fever to dengue shock syndrome lead to mortality. Definitive therapy is not available so that the treatment still relies on supportive therapy. Various herbs as antiviral have been widely explored. Calophyllum genus is known to have an antiviral effect. This study aims to analyze and assess inhibition potential of dengue virus serotype 2 replication by Calophyllum nodosum water fraction in Huh7it cell. Two methods were used including MTT 3 4, 5 dimethylthiazolyl 2 2, 5 diphenyltetrazolium bromide assay and focus assay. MTT assay was to assess uninfected cell viability, whereas focus assay aims to assess virus titer which describes DENV infectivity. The results showed viability group did not has a significant difference with half cytotoxic concentration CC50 2729 g mL, whereas infectivity group was significantly different with half inhibitory concentration CC50 10,87 g mL. The selectivity index was 251,06. Calophyllum nodosum water fraction is potential to inhibit DENV 2 replication in vitro.

Infeksi virus dengue (DENV) dapat menyebabkan penyakit demam berdarah dengue (DBD) yang ditularkan melalui nyamuk betina Aedes aegypti. Penyakit ini meningkat setiap tahunnya di dunia pada tahun 2010 sekitar 2,2 juta orang dan sekitar 3,34 orang juta pada tahun 2016. Permasalahan yang terjadi ialah ketersediaan obat spesifik DENV belum ada hingga sekarang. Salah satu pengobatan yang sedang berkembang di Indonesia ialah pengobatan herbal, seperti Piperin. Piperin merupakan senyawa murni dari buah lada (Piper nigrum, Piper longum, dan Piper retrovractum) dan diketahui memiliki kemampuan untuk dapat menghambat replikasi virus DENV-2 pada sel Vero. Namun, belum diketahui spesifik pada mekanisme apa piperin menghambat replikasi DENV-2 tersebut. Penelitian ini akan fokus pada mekanisme efektivitas hambatan replikasi virus pada pre-post infeksi dan post infeksi dan juga viabilitas sel. Efektivitas hambatan dan viabilitas sel dilihat dengan metode focus assay dan MTT assay. Berdasarkan penelitian, didapatkan hasil efektivitas hambatan pada mekanisme penghambatan pre-post infeksi dan post infeksi ialah 89,74%  2,56 dan 84,53%  3,44 secara berurutan. Sedangkan untuk viabilitas didapatkan hasil 100,03% dan 102,00% untuk mekanisme pre-post infeksi dan post infeksi. Hal ini menunjukkan piperin efektif menghambat DENV-2 dengan persentase penghambatan lebih tinggi pada mekanisme pre-post infeksi.
Infection by dengue virus (DENV) can cause dengue hemorrhagic fever (DHF) which is transmitted through female Aedes aegypti mosquitoes. This disease keeps rising every year around the world, in 2010 approximately 2.2 million people to 3.34 million people in 2016 got infected. The occurring problem is no specific antiviral drug to DENV. One of the treatments that are developing in Indonesia is herbal medicine, Piperine. Piperine is one of the pure compounds of pepper (Piper nigrum, Piper longum, dan Piper retrovractum). It is believed to be able to inhibit DENV-2 viral replication in Vero cell. The effectivity of Piperine as antiviral against DENV-2 has been proven in the thesis conducted by Heidi. However, its specific mechanisms inhibit DENV-2 replication has not been known yet. Therefore, this is an experimental study of Piperine against DENV- 2 strain NGC viral replication in Vero cell. The focus of research is looking at the effectivity of inhibition in pre-post infection, post infection, and also cell viability. The effectivity of inhibition is seen by the focus assay method. Based on the research, the results for the effectivity in pre-post infection is 89,74%  2,56, and 84,53%  3,44 in post infection. As for the cell viability, the results are 100,03%  2,74 in pre-post infection and 102,00%  3,23 in post infection. These show that Piperine is effective in inhibiting DENV-2 and the effectivity of inhibition is higher in pre-post mechanism.
