Berdasarkan data dari WHO masih terdapatnya peningkatan angka disabilitas yang disebabkan karena cedera pada sistem saraf tepi. Setelah terjadinya cedera saraf tepi, maka akan terjadi serangkaian proses seluler maupun molekuler sebagai respon terhadap adanya cedera. Salah satu respon molekuler yang penting yaitu aktivasi jalur persinyalan balik, menyebabkan terinduksinya sintesis berbagai protein, salah satunya adalah HSP 70, berperan sebagai neuro proteksi dan mencegah terjadinya apoptosis neuron lebih lanjut. Tujuan penelitian ini yaitu untuk menilai pengaruh pemberian PRP terhadap proses regenerasi saraf tepi dengan melihat hubungan ekspresi HSP 70 terhadap fungsi berjalan (TFI & PFI). Penelitian ini menggunakan sampel bahan biologis tersimpan yaitu Medulla Spinalis tikus berjumlah 36, terbagi menjadi dua kelompok besar berdasarkan hari terminasi nya yaitu H-7 dan H-42. Hasil penelitian ini menunjukan bahwa tidak terdapatnya hubungan (p>0,05) antara ekspresi HSP 70 terhadap fungsi berjalan melalui pemeriksaan TFI dan PFI pada kelompok skiatika yang diberi PRP maupun yang tidak diberi PRP, berarti bahwa tidak adanya hubungan secara langsung antara ekspresi HSP 70 dengan perbaikan fungsi motorik. ......According to WHO data, the number of disabilities brought on by damage to the peripheral nervous system is continually rising. Following a peripheral nerve damage, a number of cellular and molecular reactions will take place. One significant biological reaction is the activation of the reverse signaling pathway, which triggers the creation of several proteins, including HSP 70, which serves as neuroprotection and stops more neuronal apoptosis. The aim of this study was to assess the effect of PRP administration on the process of peripheral nerve regeneration by examined the correlation between HSP 70 expression with the motoric function (TFI & PFI). This research used samples of stored biological material, namely 36 rat spinal cords, divided into two large groups based on termination day, namely H-7 and H-42. The results of this study showed that there was no correlation (p>0.05) between HSP 70 expression and walking function through TFI and PFI examinations in the sciatica group who were given PRP and those who were not given PRP, indicating that there is no direct correlation between HSP 70 expression with motor function improvement.

Cedera saraf tepi merupakan salah satu penyebab disabilitas di dunia. Cedera saraf tepi akan merangsang badan neuron motorik untuk mengalami kromatolisis. PRP saat ini dianggap bermanfaat untuk regenerasi sel saraf karena mengandung berbagai faktor pertumbuhan yang berperan dalam neuron survival dan pertumbuhan akson. Penelitian ini bertujuan untuk mengetahui peran PRP terhadap fungsi berjalan melalui regerasi neuron motorik di medulla spinalis. Penelitian menggunakan enam kelompok tikus wistar jantan yang diberi perlakuan operasi sham, model skiatika, dan model skiatika yang diberi PRP. Tikus dilakukan terminasi pada hari ke-7 dan 42. Model skiatika dibuat dengan melakukan penjepitan di nervus ischiadicus selama 3 menit. Pemeriksaan histologi menggunakan pewarnaan hematoksilin eosin dan toluidine blue pada sediaan medulla spinalis tikus untuk menilai densitas badan nissl. Ekspresi marker regenerasi menggunakan pemeriksaan imunohistokimia GAP-43. Fungsi motorik dinilai setiap minggu menggunakan Sciatic Functional Index (SFI) dan Foot Fault Test (FFT). Terdapat perbedaan signifikan pada pemeriksaan densitas badan nissl neuron motorik di medulla spinalis pada hari ke-42. Pemeriksaan motorik dengan pengukuran SFI dan FFT menunjukkan perbedaan signifikan pada hari ke-7 dan 14. Hasil penelitian menunjukkan bahwa pemberian terapi PRP dapat mempercepat regenerasi saraf perifer yang ditandai dengan peningkatan badan nissl dan perbaikan fungsi motorik. ......Peripheral nerve injury is one of the leading causes of disability. Peripheral nerve injury stimulates motor neuron bodies to undergo chromatolysis. PRP is currently considered beneficial for nerve cell regeneration. It contains growth factors that affect neuron survival and axon growth. This study aims to determine PRP’s role on walking function through motor neuron regeneration in spinal cord. The study used six groups of male wistar rats treated with sham surgery; sciatica model; sciatica+PRP. The rats were terminated on days 7 and 42. Sciatica model was made by clamping the sciatic nerve for 3 minutes. Histological examination using hematoxylin eosin and toluidine blue staining on rat spinal cord to assess Nissl body density. Expression of regeneration markers using GAP-43 immunohistochemistry. Motor function was assessed weekly with Sciatic Functional Index (SFI) and Foot Fault Test (FFT). There was a significant difference in the examination of the density of the nissl body of motor neurons on day 42. Motor examination with SFI and FFT measurements showed significant differences on the 7th and 14th days. The results showed that PRP therapy could accelerate peripheral nerve regeneration which was characterized by an increase in nissl body and motor function improvement.

The introduction includes history of nerve repair research and establishes key concepts and terminology and will be followed by sections that represent the main areas of interest in the field: (1) Biomaterials, (2) Therapeutic Cells, (3) Drug, Gene and Extracellular Vesicle Therapies, (4) Research Models and (5) Clinical Translation. Each section will contain 3 - 6 chapters, capturing the full breadth of relevant technology. Bringing together diverse disciplines under one overarching theme echoes the multidisciplinary approach that underpins modern tissue engineering and regenerative medicine. Each chapter will be written in an accessible manner that will facilitate interest and understanding, providing a comprehensive single reference source.
