Inflammation is tissue respons of the body to injury and infection, is ssually cure by medicine class AINS that has seriously side effect, such as disturbance digestion. Mangkokan leaf (Nothopanax scutellarium Merr.) in empirical can be used as antyinflammation. The aim of this study was to determine the optimal dose that had greatest inhibition edema effect in plantar of male white rats furrow Sprague–Dawley induced by 2% carrageenan. This study used Winter method that had modified at 30 male rats which had been divided into five groupes. First groupe had been given with CMC 0.5% as negative control, groupe II, III, and IV had been given with variation dose 0,9; 1,8 dan 3,6 g dried powder/200 g BW, and groupe V had been given diclofenac sodium as positive control, and each of them had been given orally. The measuring volume edema continued every one hour during seven hours. The result show that infusa of mangkokan leaf at dose 3,6 g dried powder/200 g BW has greatest inhibition percentage, about 38,65% and statitical value (p<0.05) showed significant differences with negative control at third until fourth hour after injection carrageenan.
Inflamasi merupakan suatu respon jaringan pada tubuh terhadap cedera dan infeksi, yang pada umumnya diterapi dengan obat golongan AINS yang memiliki efek samping serius, diantaranya gangguan pencernaan. Daun mangkokan (Nothopanax scutellarium Merr.) secara empiris digunakan sebagai antiinflamasi. Tujuan penelitian ini adalah untuk menentukan dosis optimal yang dapat memberikan efek penghambatan udem terbesar pada telapak kaki tikus putih jantan galur Sprague-Dawley yang diinduksi karagenin 2%. Pada penelitian ini digunakan metode Winter yang telah dimodifikasi pada 30 ekor tikus putih jantan, yang dibagi menjadi 5 kelompok. Kelompok pertama diberikan CMC 0,5% sebagai kontrol negatif, kelompok II, III, dan IV diberikan variasi dosis ekstrak, yaitu 0,9; 1,8 dan 3,6 g serbuk kering/200 g BB tikus, serta kelompok V yang diberikan natrium diklofenak sebagai kontrol positif secara per oral. Pengukuran volume udem berturut-turut dilakukan setiap jam, selama 7 jam. Hasil penelitian menunjukkan bahwa infus daun mangkokan memiliki persentase penghambatan tertinggi pada dosis 3,6 g serbuk kering/200 g BB tikus sebesar 38,65% dan berdasarkan uji statistik (p< 0,05) menunjukkan bahwa terdapat perbedaan yang bermakna dengan kontrol negatif pada jam ketiga hingga keempat.

Daun mangkokan memiliki khasiat untuk mengatasi kerontokan rambut yang telah dibuktikan oleh penelitian sebelumnya. Namun, metode ekstraksi yang digunakan kurang efektif untuk mengektraksi flavonoid yang diduga sebagai senyawa aktif yang bertanggungjawab atas masalah kerontokan rambut. Flavonoid juga memiliki kelarutan lipid yang buruk sehingga dibutuhkan sistem penghantaran yang baru, yaitu fitosom. Tujuan penelitian ini adalah untuk membuat formulasi sediaan losio fitosom ekstrak daun mangkokan (Nothopanax scutellarium, Merr.) menggunakan metode ekstraksi dengan green solvent yang optimal. Ionic liquids meliputi; (BMIM)Br, (BMIM)BF4, (BMIM)Cl, (BMIM)HSO4, (HMIM)Br digunakan untuk mengekstraksi senyawa flavonoid dibandingkan dengan metode konvensional. Pelarut (BMIM)Cl dengan garam pemisahan NaCl memiliki kadar total flavonoid sebesar (360.57 mg/g). Penetapan kadar kuersetin diperoleh pelarut yang optimum adalah (BMIM)BF4 dengan garam pemisahan NaCl dengan kadar (26.13 mg/g). Hasil menunjukkan bahwa metode ekstraksi konvensinal, yaitu ekstrak metanol daun mangkokan memiliki kadar total flavonoid (411.08 mg/g) dan kuersetin (127.1 mg/g) yang lebih tinggi dibandingkan ionic liquid. Namun dalam sifat ramah lingkungan, dan efisiensi waktu pelarut ionic liquid perlu dipertimbangkan. Formula losio fitosom yang diuji aktivitas pertumbuhan rambutnya dengan parameter panjang rambut, dan bobot rambut. Data dianalisis menggunakan anova two way diperoleh hasil losio fitosom memiliki aktivitas yang lebih baik dibandingkan dengan minoxidil 2% dengan nilai signifikansi (p=0,0001). ...... Mangkokan leaves have efficacy to curing hair loss which has been proven by previous studies. However, the extraction method used is not effective for extracting flavonoids that are thought to be active compounds responsible for hair loss problems. Flavonoids have poor lipid solubility. New delivery system called phytosome was needed. The purpose of this study was to formulate phytosome lotion of mangkokan leaves extract (Nothopanax scutellarium, Merr.) using an optimal green solvent extraction method. Ionic liquids include; (BMIM) Br, (BMIM)BF4, (BMIM)Cl, (BMIM)HSO4, (HMIM)Br were used to extract flavonoid compounds. (BMIM)Cl solvent with NaCl separation salt has the higher total flavonoid level of (360.57 mg/g). Determination of quercetin levels obtained that (BMIM)BF4 with NaCl salt separation as the optimum solvent with quercetin levels (26.13 mg/g). The results showed that the convensinal extraction method; methanol extract of mangkokan leaves had the highest total flavonoid (411.08 mg/g) and quercetin (127.1 mg/g) levels among ionic liquids. Ionic liquids characterization such as environmentally friendly, and the time efficiency of extraction were needed to be considered. The phytosome lotion formula was tested for hair growth activity with parameters of hair length and hair weight. Data were analyzed using two way ANOVA. The result obtained that phytosome lotion had better activity than 2% Minoxidil with a significance value (p = 0.0001).

Nothopanax scutellarium is one of plant that has several biological activities such as obstetrics, kidney disease, inflammatory disease, and promotion of hair growth. Ethosomes are the ethanolic phospholipid vesicles which are used mainly for transdermal drug delivery of drugs. Ethosomes have higher penetration rate through skin. It contains soft vesicle, composed of hydroalcoholic or hydroglycolic phospholipid in which concentration of alcohol is relatively high. Ethosomes are a novel carrier for enhanced skin delivery. The aims of this research were to formulate and obtain ethosome which formed by thin layer hydration method and to know the safety of gel ethosome which contain ethyl acetate fraction. Safety test measured by scoring and categorizing within chorioallantoins membran. (HET CAM). The formulation of ethosome that using 20% ethanol and the ratio of phosphatidylcholine and ethyl acetate fraction were 1:2. The characterization from the ethosome suspension indicated that the entrapment efficiency value of 52.79% with particle size distribution value (Z-Average) 110.5 nm, polydispersity index (PdI) 0.393 and zeta potential value -11.0 mV. The result of safety test showed that 0.5% and 1% gel ethosome contain ethyl acetate fraction of scutellarium leaves have mild irritation effect.

Mangkokan Nothopanax scutellarium Merr. diketahui memiliki aktivitaspertumbuhan rambut berdasarkan penggunaan secara tradisional dan ilmiah.Kandungan flavanoid di dalam daun mangkokan diduga sebagai senyawa yangberperan dalam pertumbuhan rambut. Etosom adalah suatu sistem penghantaryang dapat berpenetrasi ke dalam lapisan kulit dan terdiri atas fosfolipid, alkoholdalam konsentrasi yang relatif tinggi dan air. Tujuan dari penelitian ini adalahmengetahui aktivitas pertumbuhan rambut dari gel etosom fraksi etil asetat daunmangkokan dibandingkan dengan gel non etosom fraksi etil asetat daunmangkokan serta kemampuannya untuk berpenetrasi ke dalam kulit dan keamanandari gel etosom fraksi etil asetat daun mangkokan. Pengujian aktivitaspertumbuhan rambut diukur berdasarkan 3 parameter yaitu panjang rambut,ketebalan rambut, dan bobot rambut. Hasil yang didapatkan menunjukkan geletosom 1 memiliki aktivitas pertumbuhan rambut yang lebih baik dibandingkangel etosom 0,5 dan gel non etosom, namun setara dengan kontrol potisif gelminoxidil 2 . Uji penetrasi secara in vitro menggunakan sel difusi Franzmenunjukkan bahwa gel etosom 1 memiliki jumlah kumulatif kuersetinterpenetrasi lebih tinggi dibandingkan gel non etosom, yaitu 3,3175 0,02 ?gcm-2untuk gel etosom 1 dengan nilai fluks 0,4147 0,01 ?gcm-2jam-1 dan2,7663 0,19 ?gcm-2 untuk gel non etosom dengan nilai fluks 0,3458 0,02 ?gcm-2jam-1. Hasil uji keamanan dengan metode HETCAM menunjukkan gel etosom0,5 dan 1 bersifat mengiritasi ringan pada pemakaian secara topikal.
Mangkokan Nothopanax scutellarium Merr. is traditionally and scientificallyknown for its activity in hair growth. The content of flavonoids in the mangkokanleaves is suspected as a vital compound in hair growth. Ethosome which consist ofphospholipids, high concentrate of alcohol, and water, are functioning as atransmission system in skin layers rsquo penetration. The aims of this study were toknow the hair growth activity from the ethosomal gel of ethyl acetate fractioncompared to the non ethosomal gel of ethyl acetate fraction, as well as its abilityto penetrate into the skin and its safety from ethasomal gel testing. The hairgrowth activity was measured by three parameters hair length, hair thickness, andhair weight. The results obtained are shown that the ethosomal gel withconcentration 1 have better hair growth activity rather than the ethosomal gelwith concentration 0.5 and the non ethosomal gel, but it has similarity to thepositive control minoxidil gel with concetration 2 . In vitro penetration testusing Franz Diffusion cells shown that the ethosomal gel with concentration 1 has cumulative penetration of quercetin higher than non ethosomal gel, whichvalue was 3,3175 0,02 gcm 2 for the ethosomal gel with concentration 1 and2,7663 0,19 gcm 2 for the non ethosomal gel with flux values for the ethosomalgel with concentration 1 and the non ethosomal gel respectively are0,4147 0,01 gcm 2jam 1 and 0,3458 0,02 gcm 2jam 1. As for the safety testusing HET CAM have shown that both concentrations 0.5 and 1 of ethosomalgel are able to generate mild irritation for topical application.
