Kanker serviks adalah salah satu penyebab kematian yang besar di dunia. Sehingga penelitian untuk menemukan terapi sebagai alternatif untuk obat-obatan sitotoksik terus dikembangkan. Hal ini karena, obat-obatan sitotoksik konvensional memiliki efek samping seperti; malnutrisi, gejala mual, penurunan daya tahan, dan lainya. Citronellol telah dilaporkan dapat menginduksi apoptosis pada sel kanker darah dan sel kanker payudara dengan menginduksi caspase-3. Citronellol juga telah dilaporkan memiliki potensi anti-bakteri, anti-oksidan, anti-inflamasi dan kemampuan untuk meningkatkan imun dari pasien kanker. Untuk menguji potensi Citronellol digunakan sel HeLa yang merupakan cell line kanker serviks yang memiliki sifat kuat, immortal, dan mudah dipropagasi dalam lab. Konsentrasi Citronellol yang digunakan adalah 10 µg/mL, 25 µg/mL, 50 µg/mL, dan 100 µg/mL. Untuk menguji potensi Citronellol terhadap viabilitas dan ultrastruktur sel HeLa digunakan uji Trypan Blue untuk melihat jumlah sel hidup dan mati, Scepter™ 3.0 cell counter untuk membandingkan rata-rata ukuran diameter sel dan melihat ultrastruktur sel dengan Scanning Electron Microscope (SEM). Viabilitas sel HeLa diketahui semakin menurun seiring semakin tinggi konsentrasi Citronellol. Hal ini dibuktikan dari semakin sedikit jumlah sel yang hidup ketika konsentrasi Citronellol semakin tinggi pada uji Trypan Blue. Pada pengujian dengan Scepter™ 3.0 cell counter diameter sel makin mengecil ketika konsentrasi Citronellol semakin tinggiHasil uji statistik ANOVA menunjukkan penurunan viabilitas pada uji Trypan Blue memiliki nilai yang signifikan namun, pengujian dengan Scepter™ 3.0 cell counter memiliki nilai yang tidak signifikan. Hasil pengamatan dengan SEM, sel HeLa terlihat memiliki karakteristik sel yang sedang berada di fase apoptosis ketika sel diberi Citronellol. ......Cervical cancer is one of the major causes of death in the world. Therefore, research is conducted trying to find alternative therapies. Reason for further research is due to the many side effects caused by conventional cytotoxic drugs used in those conventional therapy such as; such as; malnutrition, symptoms of nausea, decreasing immunity and more. Citronellol has been reported to induce apoptosis in blood cancer cells and breast cancer cells by inducing caspase-3. Citronellol has also been reported to have potential anti-bacterial, anti-oxidant, anti-inflammatory and immune-boosting abilities in cancer patients. In order to test the potential of Citronellol HeLa cells are used due to it being a cervical cancer cell and with characteristics like; strong, immortal and easily propagated in the lab. The amount of Citronellol used are 10 µg/mL, 25 µg/mL, 50 µg/mL, dan 100 µg/mL. In order to test the potential of Citronellol on the viability and ultrastructure of HeLa cells, the Trypan Blue test was used to see the number of live and dead cells, the Scepter™ 3.0 cell counter to compare the average cell diameter sizes and Scanning Electron Microscope (SEM) is used to see the cell ultrastructure. HeLa cell viability is known to decrease as the concentration of Citronellol increases. In the Trypan Blue test this is evident shown by the fewer number of viable cells as the concentration of Citronellol is higher. In the test with Scepter ™ 3.0 cell counter, the diameter of the cell gets smaller as the concentration of Citronellol got higher. The results of ANOVA statistical test, the decrease in viability in the Trypan Blue test had a significant value, but not with the Scepter™ 3.0 cell counter SEM pictures showed HeLa cells with characteristics of cells undergoing apoptosis when given Citronellol.

Cancer cells have telomerase activity is relatively high compared to normal cells, so the cancer cells have the ability to continue to proliferate. Cancer cells undergo uncontrolled mitosis and have high telomerase activity compared to cells normal. Telomerase is an enzyme responsible for telomere length, a segment of DNA that is the tip of chromosomes in eukaryotic cells. Telomeres are associated with the process of aging and carcinogenesis. The purpose of this study was to determine the expression of telomerase in some cells such as breast cancer, cervical cancer, and lung cancer. Methods: The research method is experimental studies in several cancer cell cultures in the form of cell line. Cancer cells used were: HeLa (cervical cancer), MCF7 and T47D (breast cancer), WiDr (lung cancer), and Raji (lymphoma) with culture medium RPMI, DMEM, and M199. Vero cells is used (fibroblast cells) as a control (normal cells). Expression of telomerase enzyme was measured by the Immunohystochemistry (IHC) method. Results: The results showed that the cancer cells have activity/higher telomerase expression were highly significant (p<0.01) compared to normal cells (Vero cells). Similarly, the expression of telomerase in HeLa versus WiDr, WiDr versus T47D, T47D versus Raji, and Raji versus MCF7 also showed highly significant differences (p < 0.01). Telomerase expression between cancer cells that showed significant difference (HeLa cells versus Raji cells; HeLa cells versus MCF7 cell; T47D cells versus MCF7 cells) (p < 0.05). No significant difference was found in the group of HeLa cells versus T47D, WiDr versus Raji cells, and WiDr versus MCF7. Conclusions: It was concluded, that the cancer cells have telomerase expression of specific and different from each other, depending on the type of cell. T47D breast cancer cells have telomerase expression of the highest, followed by cervical cancer cells (HeLa). Lung cancer cells (WiDr) with cell lymphoma (Raji) has almost the same expression and both have lower expression.;

Cervical cancer is the second most frequently occurring cancer among females all over the world. Since the best strategy against cancer is to kill the cancer cells without endangering the normal cells, discovering a potentially selective anticancer agent from plants has become very challenging for researchers worldwide. A previous study on Anredera cordifolia, known as binahong in Indonesia, revealed its cytotoxic activity on HeLa cervical cancer cells with IC50 75 µg/mL. However, the selectivity of the chemical agent and its molecular target was not investigated. Thus, this study was aimed at determining the selectivity of ethanolic extract of Anredera cordifolia leaf (EAC) on Vero cells and its molecular target on HeLa cells. The extracts were prepared by macerating A. cordifolia leaf powder in 70% ethanol. The viability of Vero cells was assessed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. While the cell cycle of HeLa cells was analyzed using the flow cytometry, the molecular target of the extract was investigated by using the immunocytochemical staining. The results exhibited the selective cytotoxicity of EAC on HeLa cells compared to Vero cells with a Selectivity Index (SI) of 17.36. It arrested the G1/S phase of the cell cycle and suppressed the Bcl–2 expression, the anti-apoptotic protein, which also regulates the cell cycle. These findings confirmed the use of A. cordifolia leaf extract as a promising anticancer agent against cervical cancer, particularly the HeLa cells.

Hingga 2018 32.469 kasus kanker serviks diambil pada wanita Indonesia, perbedaan 17,2% dari total kasus kanker di Indonesia. Berbagai upaya dilakukan untuk mengurangi kematian akibat penyakit ini, menentang pengembangan sistem pemberian obat nanoteknologi. Dalam penelitian ini, konjugasi doxorubicin pada partikel nano magnetik Fe3O4 (MNPs) dengan agen capping berbasis karboksilat digunakan untuk menstabilkan partikel nano Fe3O4 untuk melihat pengaruhnya terhadap pemuatan obat dan pelepasan obat dari MNPs. Nanopartikel yang disintesis dikarakterisasi dengan spektroskopi Fourier Transform Infrared (FTIR), Difraksi Sinar-X (XRD), Transmission Electron Microscope (TEM), Particle Size Analyzer (PSA), dan Vibrating Sample Magnetometer (VSM). Pengukuran efisiensi pemuatan dan pelepasan obat dari tiga nanokomposit menghasilkan hasil yang baik, dengan persentase efisiensi pemuatan terbesar, yaitu 90.230% dicapai oleh AA-MNPs. Sementara itu, persentase terbesar dari obat yang dilepaskan adalah 98,590% yang disetujui oleh SA-MNPs. Hasil tes MTT pada sel HeLa menunjukkan bahwa nanokomposit CA-MNP memiliki efisiensi pelepasan obat terbaik, dengan nilai persen sel hidup hanya 23,460%.hr> ABSTRACT
ntil 2018 32,469 cases of cervical cancer were taken in Indonesian women, a difference of 17.2% of the total cancer cases in Indonesia. Various attempts were made to reduce mortality due to this disease, opposing the development of nanotechnology drug delivery systems. In this study, doxorubicin conjugate on Fe3O4 magnetic nanoparticles (MNPs) with carboxylic-based capping agents used to stabilize Fe3O4 nanoparticles to see their effect on drug loading and drug release from MNPs. The synthesized nanoparticles were characterized by Fourier Transform Infrared (FTIR) spectroscopy, X-Ray Diffraction (XRD), Transmission Electron Microscope (TEM), Particle Size Analyzer (PSA), and Vibrating Sample Magnetometer (VSM). Measurement of drug loading & release efficiency from three nanocomposites produced good results, with the largest percentage loading efficiency, ie 90,230% achieved by AA-MNPs. Meanwhile, the largest percent of drug released was 98.590% approved by the SA-MNPs. MTT test results on HeLa cells showed that the CA-MNP nanocomposite had the best drug release efficiency, with a live cell percent value of only 23.460%.

Kanker serviks merupakan salah satu jenis penyakit kanker dengan prevalensi dan mortalitas yang tinggi di Indonesia. Eucheuma cottonii merupakan salah satu jenis rumput laut yang banyak dijumpai di perairan laut Indonesia dan dapat dikembangkan sebagai agen anti kanker. Penelitian ini bertujuan untuk mengetahui komposisi fitokimia dan efek sitotoksik ekstrak makroalga Eucheuma cottonii terhadap sel kanker serviks HeLa yang dinyatakan dengan nilai IC50. Pada penelitian ini, Eucheuma cottonii diekstraksi masing-masing menggunakan pelarut etanol, etil asetat, n-heksana, dan kloroform. Ekstrak yang diperoleh kemudian diuji kandungan fitokimianya, dan menunjukkan hasil positif mengandung flavonoid. Sedangkan analisis kualitatif dengan kromatografi lapis tipis, menunjukkan bahwa ekstrak mengandung tiga hingga lima senyawa kimia. Selanjutnya, masing-masing ekstrak sebanyak 20 L dengan 5 variasi konsentrasi, yaitu 3,125 g/ml; 6,25 g/ml; 12,5 g/ml; 25 g/ml; dan 50 g/m, dimasukkan ke lini sel HeLa yang sudah ditambahkan 100 L DMEM dan diinkubasi selama 24 jam. Setelah diinkubasi selama 24 jam, dilakukan uji MTT assay dengan panjang gelombang 492 nm. Data yang diperoleh kemudian dianalisis sehingga diperoleh nilai IC50 untuk keempat ekstrak Eucheuma cottonii yang diujikan. Hasil menunjukkan bahwa keempat ekstrak Eucheuma cottonii memiliki nilai IC50
ABSTRACT
Cervical cancer is one kinds of cancer with high prevalence and mortality in Indonesia. Eucheuma cottonii is one kind of seaweed which is commonly found in Indonesian marine and can be developed as anticancer agent. This research aims to know the phytochemical composition and cytotoxic effect of extract of makroalgae Eucheuma cottonii on HeLa cervical cancer cells that is expressed by IC50 value. In this research, Eucheuma cottonii were extracted each using ethanol, ethyl acetate, n hexane, and chloroform. The obtained extracts were then tested for its phytochemical content, and showed positive result containing flavonoids. While qualitative analysis with Thin Layer Chromatography TLC , showed that the extract contains three to five chemical compounds. Furthermore, 20 L of each extract in five variation of concentration, i.e. 3,125 g ml 6,25 g ml 12,5 g ml 25 g ml and 50 g ml, inserted into the HeLa cell line that has been added 100 L of DMEM and incubated for 24 hours. After 24 hour incubation, MTT assay with a wavelength of 492 nm was performed to generate data which was then analyzed to obtain IC50 value for the four extracts tested of Eucheuma cottonii. The results showed that all four Eucheuma cottonii extract had IC50 values less than 100 g ml, so it can be concluded that all extracts have cytotoxic activity against HeLa cervical cancer cell HeLa. Among four extracts of Eucheuma cottonii, ethyl acetate extract has the lowest IC50 value and shows the most potent cytotoxic activity against HeLa cervical cancer cells. Thus, ethyl acetate extract of Eucheuma cottoniiis potential to be developed as an anticervical cancer agent.Key word Phytochemical, macroalgae, Eucheuma cottonii , cytotoxic, HeLa cells

Kanker serviks adalah penyebab paling umum kematian akibat kanker pada wanita. Kanker serviks sering dikaitkan dengan human papilloma virus. Pengembangan senyawa yang berasal dari asam risinoleat banyak dilakukan karena diketahui memiliki berbagai manfaat seperti antioksidan, antibakteri dan salah satunya adalah antikanker. Penelitian ini bertujuan untuk menguji aktivitas antiproliferasi terhadap sel HeLa senyawa alkanolamida (R)-12-hidroksi-N,N-bis(2-hidroksietil)stearamida. Pertama-tama metil risinoleat direduksi dengan gas H2 dan katalis Pd/C. Selanjutnya dilakukan reaksi amidasi dengan dietanolamina untuk menghasilkan senyawa alkanolamida dan dilanjutkan dengan pemurnian menggunakan kromatografi kolom dan dimonitor dengan kromatografi lapis tipis (KLT). Diperoleh %rendemen senyawa alkanolamida dari hasil amidasi sebesar 97,85%. Berdasarkan hasil karakterisasi dengan FTIR, munculnya puncak dari gugus C-N pada bilangan gelombang 1135 cm-1, gugus C=O pada 1735 cm-1, dan O-H (regangan) berupa jangkauan dari 3700-3000 cm-1, dan O-H (tekuk) pada 1250 cm-1. yang mengkonfirmasi produk alkanolamida berhasil terbentuk. Selanjutnya senyawa alkanolamida (R)-12-hidroksi-N,N-bis(2-hidroksietil)stearamida diuji aktivitas antiproliferasinya menggunakan metode MTT terhadap sel HeLa, untuk menguji aktivitasnya sebagai agen antikanker. Diperoleh nilai IC50 sebesar 114,9 μM, menunjukkan bahwa senyawa (R)-12-hidroksi-N,N-bis(2-hidroksietil)stearamida memiliki aktivitas antiproliferasi yang termasuk dalam kategori toksik lemah. ......Cervical cancer is the most common cause of death from cancer in women. The development of compounds derived from ricinoleic acid is widely carried out because it is known to have various benefits such as antioxidants, antibacterials and one of them is anticancer. This study aims to examine the antiproliferative activity of HeLa cells with the alkanolamide compound (R)-12-hydroxy-N,N-bis(2-hydroxyethyl)stearamide. First of all, methyl ricinoleate is reduced with H2 gas and Pd/C catalyst. Furthermore, the amidation reaction was carried out with diethanolamine to produce alkanolamide compounds and continued with purification using column chromatography and monitored by thin layer chromatography (TLC). The % yield of alkanolamide compounds from amidation was 97.85%. Based on the results of characterization with FTIR, the emergence of peaks from the C-N group at wave number 1135 cm-1, the C=O group at 1735 cm-1, and O-H (stretch) are in the range of 3700-3000 cm-1, and O-H (bending) at 1250 cm-1. which confirmed that the alkanol amide product was successfully formed. Furthermore, the alkanolamide compound (R)-12-hydroxy-N,N-bis(2-hydroxyethyl)stearamide was tested for its antiproliferation activity using the MTT method against HeLa cells, to test its activity as an anticancer agent. The IC50 value of 114.9 μM was obtained, indicating that the compound (R)-12-hydroxy-N,N-bis(2-hydroxyethyl)stearamide has an antiproliferative activity which is included in the weak toxic category.

Kanker serviks menjadi kanker penyebab kematian tertinggi ketiga di Indonesia setelah kanker paru-paru dan payudara. Metode pencegahan seperti vaksin HPV dan metode pengobatan seperti pembedahan, kemoterapi, dan radioterapi telah dikembangkan. Akan tetapi, masih dibutuhkan pengembangan pengobatan alternatif, salah satunya adalah dari tumbuhan herbal dengan senyawa aktif citronellol. Penelitian-penelitian terdahulu membuktikan bahwa citronellol dapat menurunkan viabilitas dengan menginduksi apoptosis pada beberapa sel kanker, seperti sel kanker paru-paru, payudara, dan darah. Tujuan penelitian ini adalah untuk mengetahui pengaruh citronellol terhadap viabilitas sel HeLa menggunakan mikroskop fluoresens. Variasi konsentrasi 10, 25, 50, dan 100 µg/mL citronellol dengan waktu inkubasi 48 jam diujikan pada sel HeLa. Uji viabilitas dilakukan dengan uji WST-1 dan deteksi apoptosis dilakukan dengan pewarnaan Annexin V-FITC/PI menggunakan mikroskop fluoresens. Hasil uji dengan ANAVA satu arah menunjukkan tidak terdapat pengaruh signifikan dari citronellol terhadap viabilitas sel HeLa. Konsentrasi 10 µg/mL cenderung menurunkan viabilitas sel HeLa dibandingkan kontrol. Pengamatan dengan mikroskop fluoresens menunjukkan terdeteksinya sel apoptosis yang lebih tinggi dibandingkan dengan sel nekrosis dan perubahan morfologi pada sel late apoptosis. Penelitian lebih lanjut diperlukan untuk mengetahui lebih jauh pengaruh citronellol terhadap sel HeLa. ......Cervical cancer is the third leading cause of cancer death in Indonesia following lung cancer and breast cancer.  Prevention methods such as the HPV vaccine and treatment methods such as surgery, radiotherapy, and chemotherapy have been developed. However, the development of alternative medicine is still needed, one of which is from herbal plants with the active compound of citronellol. Previous studies have shown that citronellol can reduce viability by inducing apoptosis in some cancer cells, such as lung, breast, and blood cancer cells. This study aims to determine the effects of citronellol on HeLa cells viability and to detect apoptosis by Annexin V- FITC/PI staining using a fluorescence microscope. Concentration variations of 10, 25, 50, and 100 µg/mL citronellol with an incubation time of 48 hours were treated on HeLa cells. A viability test was carried out by WST-1 test and apoptosis detection was carried out by Annexin V-FITC/PI staining using a fluorescence microscope. The results of the one-way ANOVA test showed that there was no significant effect of different concentrations of citronellol on the viability of HeLa cells. The concentrations of 10 µg/mL of citronellol tended to decrease HeLa cells viability. Observations with fluorescence microscope showed a higher detection rate of apoptotic cells than necrotizing cells and a more visible morphological changes in late apoptotic cells. Furthermore, fluorescence microscopy detected apoptotic cells. Further study is required to get a more comprehensive information on the effects of citronellol on HeLa cells.

Kanker serviks menempati posisi kedua kanker yang paling umum diderita oleh wanita di Indonesia. Penelitian senyawa antikanker dengan menggunakan tanaman herbal banyak dikembangkan karena dilaporkan memiliki efek samping yang rendah. Senyawa citronellol dan eucalyptol merupakan senyawa yang berpotensi menjadi antikanker karena telah dilaporkan dapat menurunkan viabilitas beberapa jenis sel kanker. Namun, pengaruh konsentrasi citronellol dan eucalyptol pada sel dari kanker serviks belum diketahui. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi (10, 25, 50, dan 100 μg/mL) senyawa citronellol dan eucalyptol terhadap viabilitas sel HeLa. Metode analisis yang digunakan yaitu dengan flow cytometry. Hasil pengujian dengan Two-way ANOVA pada tingkat kepercayaan 0,05 membuktikan bahwa tidak terdapat perbedaan signifikan antar pemberian variasi konsentrasi senyawa citronellol dan eucalyptol terhadap viabilitas sel HeLa. Namun, diketahui bahwa senyawa citronellol dengan konsentrasi 50 μg/mL diketahui memiliki kecenderungan untuk menurunkan viabilias sel HeLa paling tinggi. ......Cervical cancer is the second most common cancer suffered by women in Indonesia. The test results using Two-way ANOVA at a confidence level of 0.05 proved there was no significant difference between the concentration variation of citronellol and eucalyptol compounds on the viability of HeLa cells. However, citronellol compound with a concentration of 50 μg/mL is known to have the highest tendency to decrease HeLa cell viability.

Abstrak :
Kanker serviks termasuk salah satu jenis kanker dengan tingkat prevalensi kasus yang tinggi di Indonesia. Berdasarkan GLOBOCAN 2020, ditemukan 36.633 (36% dari total kasus kanker pada wanita) kasus kanker serviks baru dan 21.003 kematian. Pengobatan terhadap kanker serviks saat ini masih menimbulkan efek samping seperti resistensi obat pada sejumlah kasus sehingga diperlukan alternatif pengobatan. Salah satu senyawa yang memiliki potensi antikanker, yaitu Eucalyptol. Eucalyptol adalah senyawa utama dalam tumbuhan eukaliptus seperti Eucalyptus globulus. Studi pengaruh konsentrasi eucalyptol terhadap viabilitas dan deteksi apoptosis sel HeLa dilakukan dengan konsentrasi 25, 50, 100 dan 200 μg/mL. Uji viabilitas dengan WST-1 menunjukkan bahwa variasi konsentrasi eucalyptol tersebut belum dapat menekan viabilitas sel HeLa (p>0,05 dengan uji ANOVA). Selanjutnya, deteksi apoptosis (menggunakan pewarna Annexin V-FITC) dengan mikroskop fluoresens menunjukkan bahwa variasi konsentrasi eucalyptol dapat menginduksi apoptosis dibandingkan nekrosis. Meskipun demikian, belum dilakukan pengamatan pada sel hidup sehingga tidak diketahui persentase sel apoptosis dan nekrosis dalam populasi total. Pengujian lanjutan dengan pewarna DAPI yang mewarnai sel hidup dan metode kuantifikasi lainnya untuk validasi hasil pengamatan dengan mikroskop fluoresens diperlukan. ......Cervical cancer is one type of cancer with a high case prevalence rate in Indonesia. One of the compounds that have anticancer potential is Eucalyptol. Eucalyptol is the main compound in eucalyptus plants such as Eucalyptus globulus. The study of the effect of eucalyptol concentration on the viability and detection of apoptosis of HeLa cells was carried out at concentrations of 25, 50, 100 and 200 g/mL. The viability test with WST-1 showed that the variation in eucalyptol concentration had not been able to suppress HeLa cell viability (p>0.05 by ANOVA test). Furthermore, detection of apoptosis (using Annexin V-FITC dye) by fluorescent microscopy showed that variations in eucalyptol concentration could induce apoptosis rather than necrosis. However, no observations have been made on living cells so that the percentage of apoptotic and necrotic cells in the total population is unknown. Further testing with DAPI dye that stains living cells and other quantification methods for validation of observations by fluorescent microscopy is required.
