"Protein Transaktivator transkripsi (Tat) adalah protein regulator HIV-1 berfungsi sebagai aktivator transkripsi genom HIV-1. Varian protein Tat-Eli adalah aktivator transkripsi paling kuat daripada varian lain melalui induksi promotor LTR HVI-1. Kemampuan tersebut digunakan sebagai kontrol positif dalam pengembangan uji infeksitivitas HIV-1 berbasis gene reporter eGFP diregulasi LTR HIV-1. Pengembangan uji infeksivitas tersebut menawarkan waktu deteksi infeksi lebih singkat daripada uji p24 pada uji fenotipik. Penelitian ini bertujuan untuk mengekspresikan protein rekombinan Tat-Eli di sistem ekpresi prokariot dan mempurifikasinya sehingga dapat dijadikan kontrol positif penginduksi promotor. Pada penelitian ini dilakukan pengklonaan gen sintetik Tat-Eli ke vektor pQE80L. Protein rekombinan Tat-Eli dipurifikasi menggunakan Ni-NTA. Pengklonaan ulang gen reporter eGFP disisipkan setelah promotor LTR HIV-1. Aktivitas protein rekombinan Tat-Eli terhadap ekspresi eGFP di sel mamalia dinilai berdasarkan persentase sel pengekspresi eGFP dan intensitas cahaya eGFP.Konstruksi plasmid rekombinan membawa gen Tat-Eli, pQETat, berhasil dibuat, diekspresikan dan dipurifikasi kondisi native. Plasmid pengekspresi eGFP  dengan promoter HIV-1, pLTReGFP berhasil dikonstruksi. Penambahan Tat-Eli rekombinan pada sel mamalia yang ditransfeksi pLTReGFP menunjukkan perbedaan intensitas cahaya eGFP yang bermakna dan paling tinggi dari semua perlakuan. Protein rekombinan Tat-Eli dapat diekspresikan dan dipurifikasi secara optimal dari E.coli. Penambahan protein Tat-Eli pada sel yang ditransfeksi pLTReGFP meningkatkan intensistas cahaya eGFP.

Transcriptional Transactivator Protein (Tat) is an HIV-1 regulatory protein functioning as an activator of HIV-1 genome transcription. The Tat-Eli protein variant was the most potent transcriptional activator than other variants through the induction of the HVI-1 LTR promoter. This ability was used as a positive control in the development of an HIV-1 infection test based on the eGFP reporter gene regulated by LTR HIV-1. The development of the infectiousness test offers a shorter infection detection time than the p24 test in the phenotypic test. This study aims to express Tat-Eli recombinant protein in the prokaryotic expression system and to purify it so that it can be used as a positive control inducer of the promoter. In this study, synthetic Tat-Eli gene was cloned into the pQE80L vector. Tat-Eli recombinant protein was purified using Ni-NTA. Recloning of the eGFP reporter gene was inserted after the HIV-1 LTR promoter. The activity of Tat-Eli recombinant protein on eGFP expression in mammalian cells was assessed based on the percentage of eGFP-expressing cells and eGFP light intensity. The recombinant plasmid construction carrying the Tat-Eli gene, pQETat was successfully generated, expressed and purified in native conditions. An eGFP-expressing plasmid with HIV-1 promoter, pLTReGFP was successfully constructed. The addition of recombinant Tat-Eli to mammalian cells transfected with pLTReGFP showed a significant difference in eGFP light intensity and was the highest of all treatments. Tat-Eli recombinant protein can be optimally expressed and purified from E. coli. The addition of Tat-Eli protein in pLTReGFP-transfected cells increased eGFP light intensity."

"Latar Belakang: Ti-6Al-4V adalah salah satu bahan implan yang paling umum digunakan. Dalam upaya mengurangi biaya produksi implan titanium, bahan dipanaskan untuk mengoptimalkan produk akhir mereka. Salah satu kondisi yang perlu dipenuhi untuk bahan implan baru adalah biokompatibilitas bahan untuk jaringan di sekitarnya.
Tujuan: Untuk memahami efek implantasi material implan Ti-6Al-4V ELI yang dipanaskan pada 10500C pada tulang paha tikus Sprague Dawley melalui studi histologis.
Metode: Desain penelitian eksperimental digunakan dalam penelitian ini dengan tikus Sprague Dawley betina yang dibagi menjadi tiga kelompok: kelompok perlakuan; kelompok kontrol normal; dan kelompok kontrol cacat, yang dibuat lubang pada tulang paha. Dua tikus digunakan di masing-masing kelompok. Pada kelompok yang diberi perlakuan, defek tulang paha diimplantasikan dengan material implan Ti-6Al-4V ELI yang dipanaskan 10500C. Tikus dikorbankan setelah empat minggu dan dievaluasi secara histologis menggunakan Salkeld Histological Scoring yang dimodifikasi.
Hasil: Skor histologis kelompok sampel 1 yang diobati adalah 3 yang berarti regenerasi tulang yang hampir sempurna, ditunjukkan oleh penampilan tulang rawan mineral. Nilai untuk kelompok perlakuan sampel 2 dan kelompok kontrol cacat sampel 1,2 adalah 2, yang berarti regenerasi tulang hanya mencapai fase pembentukan fibrocartilage.
Kesimpulan: Regenerasi tulang dapat diperoleh di sekitar lokasi implantasi 10500C yang dipanaskan Ti-6Al-4V ELI material implan yang ditunjukkan oleh adanya jaringan fibrosa, fibrocartilage, dan tulang rawan termineralisasi.

Background: Ti-6Al-4V is one of the most commonly used implant materials. In an effort to reduce the cost of producing titanium implants, the material is heated to optimize their final product. One of the conditions that need to be met for new implant material is the biocompatibility of the material for the surrounding tissue.
Objective: To understand the effect of implantation of TiI 6Al-4V ELI material that was heated at 10500C on the femur of Sprague Dawley rats through histological studies.
Methods: The experimental research design used in this study with female Sprague Dawley mice was divided into three groups: the treatment group; normal control group; and the deformed control group, which made a hole in the femur. Two mice were used in each group. In the treated group, femur defects were implanted with the implant material 10500C EL-Ti-6Al-4V. Mice were sacrificed after four weeks and evaluated histologically using modified Salkeld Histological Scoring.
Results: The histological score of the sample group 1 treated was 3, which means almost perfect bone regeneration, indicated by the appearance of mineral cartilage. The values ​​for the sample 2 treatment group and the sample defect control group 1,2 are 2, which means bone regeneration only reaches the fibrocartilage formation phase.
Conclusion: Bone regeneration can be obtained around the implantation site of ELI Ti-6Al-4V implant material heated by 10500C which is indicated by the presence of fibrous tissue, fibrocartilage, and mineralized cartilage.

"This article asks the question how Banda displaced from the Banda Islands due to colonization of Banda by the VOC in 1621, maintain their existence as a sustainable cultural group. Banda communities play an important role in maritime commerce in the eastern part of Indonesia at the beginning of the colonial period. They survive as one cultural group in two villages on the islands of Kei. The traditional songs of the two villages center on the sea voyage. The argument is that Banda people are mobilized by oral traditions that reveal the kinship ties of Banda people with their partners in commerce in distant lands."
