Abstrak :
Acrolejeunea fertilis merupakan lumut hati bentuk daun yang termasuk dalam famili Lejeuneaceae dengan potensi yang luas namun memiliki biomassa terbatas. Kultur in vitro merupakan solusi untuk perbanyakan A. fertilis. Pengaplikasian Zat Pengatur Tumbuh (ZPT) dengan variasi konsentrasi pada medium ½ MS diharapkan dapat meningkatkan persentase pertumbuhan A. fertilis. Tujuan penelitian ini adalah untuk mengetahui kombinasi dan konsentrasi ZPT yang optimum untuk kultur in vitro A. fertilis. Jenis ZPT yang digunakan yaitu 2,4- Dichlorophenoxyacetic acid (2,4-D) dan Kinetin dengan 12 variasi konsentrasi yaitu 0—1 mg/L untuk 2,4-D dan 0— 2 mg/L untuk Kinetin. Setiap perlakuan terdiri dari 14 ulangan. Pengamatan kualitatif berupa perubahan warna eksplan, pertumbuhan tunas, dan keberadaan kontaminasi. Pengamatan kuantitatif berupa persentase pertumbuhan tunas, rerata panjang tunas, jumlah tunas yang muncul dari setiap eksplan, serta persentase kontaminasi. Data jumlah eksplan yang membentuk tunas dan data panjang tunas dianalisis menggunakan ANOVA satu arah dan dilanjutkan Duncan Multiple Range Test (DMRT), α 0,05. Hasil penelitian menunjukkan bahwa terdapat variasi jumlah tunas yang tumbuh dari setiap ekpslan yang merespons. Penambahan Kinetin dengan konsentrasi 0,1—1 mg/L dan 2,4-D 1 mg/L menghasilkan pertumbuhan tunas yang paling signifikan. Pengaplikasian ZPT dengan kombinasi dan konsentrasi yang tepat mampu meningkatkan pertumbuhan tunas gametofit A. fertilis ......Acrolejeunea fertilis is a leavy liverwort, part of Lejeuneaceae with lots of potensials yet its biomass is limited. In vitro culture might be an alternative solution for A. fertilis’ multiplication. The application of growth regulator in ½ MS culture media are expected to increase A. fertilis’ shoot growth. The aim of this work is to discover the optimum concentration of growth regulator for A. fertilis’ in vitro culture. Type of growth regulators used in this research were 2,4-Dichlorophenoxyacetic acid (2,4-D) and Kinetin with 12 different concentration, range of 0—1 mg/L for 2,4-D and 0—2 mg/L for Kinetin. The qualitative parameters observed in this research were explant’s pigmentation, shoot growth, and presences of contaminations. The quantitative parameters were shoot growth percentage, average shoot length, number of shoots emerged , and percentage of contaminations. All data were analyzed with One Way ANOVA and Duncan Multiple Range Test (DMRT), α 0,05. Results showed that there were variations in shoot growth per explant. Addition of 0,1—1 mg/L Kinetin and 2,4-D 1 mg/L was the most significant concentrations for A. fertilis’ shoot growth. Addition of growth regulator with exact concentration to ½ MS media considered to increase shoot growth of A. fertilis.

This proceedings volume chronicles the papers presented at the 35th CIB W78 2018 Conference: IT in Design, Construction, and Management, held in Chicago, IL, USA, in October 2018. The theme of the conference focused on fostering, encouraging, and promoting research and development in the application of integrated information technology (IT) throughout the life-cycle of the design, construction, and occupancy of buildings and related facilities. The CIB – International Council for Research and Innovation in Building Construction – was established in 1953 as an association whose objectives were to stimulate and facilitate international cooperation and information exchange between governmental research institutes in the building and construction sector, with an emphasis on those institutes engaged in technical fields of research. The conference brought together more than 200 scholars from 40 countries, who presented the innovative concepts and methods featured in this collection of papers.
