Abstrak :
Penelitian dilakukan untuk mengetahui pengaruh kombinasi gliserol 6% dengan beberapa konsentrasi kuning telur terhadap kualitas spermatozoa ikan kerapu kertang Epinephelus lanceolatus (Bloch, 1970) pascakriopreservasi. Semen ikan kerapu kertang didapatkan dengan metode pengurutan. Larutan pengencer terdiri atas marine fish ringer, gliserol 6%, dan berbagai konsentrasi kuning telur (0%; 5%; 10%; 15%; 20%; dan 25%). Ekuilibrasi dilakukan selama 10 menit pada suhu 4 ºC. Pembekuan dalam freezer (-20 ºC) selama 48 jam. Pencairan pada suhu 45 ºC selama 30-60 detik. Evaluasi semen dilakukan secara makroskopis (warna, volume, dan pH) dan secara mikroskopis (motilitas, viabilitas, dan abnormalitas) serta kemampuan fertilisasinya terhadap telur ikan kerapu macan. Berdasarkan hasil uji ANAVA yang dilanjutkan dengan uji Tukey, terdapat perbedaan nyata (P>0,05) terhadap motilitas, viabilitas, dan kemampuan fertilisasi spermatozoa ikan kerapu kertang pascakriopreservasi, akan tetapi tidak berpengaruh nyata terhadap abnormalitas spermatozoa pascakriopreservasi (P>0,05). Hasil penelitian menunjukkan bahwa perlakuan kuning telur 15% merupakan konsentrasi optimum karena menghasilkan nilai persentase rata-rata motilitas, viabilitas, dan kemampuan fertilisasi tertinggi masing-masing sebesar 83,64 ± 1,72%, 81,44 ± 2,06%, dan 77,31 ± 1,90%. Nilai rata-rata terendah pada persentase abnormalitas sebesar 21,50 ± 1,20%.
Research on the effect of combination 6% glycerol and eggyolk as cryoprotectant of giant grouper Epinephelus lanceolatus (Bloch, 1970) spermatozoa quality postcryopreservation. Giant grouper cement is obtained by sorting method. The diluent solution used consist of marine fish ringer, 6% glycerol, and varian egg yolk concentrations (0%; 5%; 10%; 15%; 20%; dan 25%). Equilibration is carried out for 10 minutes at 4ºC. Freezing in the freezer (-20 ºC) for 48 hours. Thawing at 45 ºC for 30-60 seconds. Cement evaluation is carried out macroscopically (color, volume, and pH) and microscopically (motility, viability, and abnormality) and also the ability to fertilize the egg of tigger grouper. Based on the ANAVA statistical test followed by Tukey, there were significant differences effect on motility, viability, and fertilization ability of spermatozoa of giant grouper postcryopreservation (P> 0.05), but abnormality was significantly affected (P <0.05). (P> 0.05). The optimum concentration of egg yolk is 15% because it produces the highest percentage value of motility, viability, and fertilization ability of 83.64 ± 1.72%, 81.44 ± 2.06% and 77.31 ± 1.90% respectively. The lowest average value of abnormalitiy percentage is 21.50 ± 1.20%.

Penelitian dilakukan untuk mengevaluasi pengaruh penggunaan kombinasi etilen glikol (EG) sebagai krioprotektan intraseluler dan kuning telur sebagai krioprotektan ekstraseluler dengan variasi konsentrasi dalam mempertahankan jumlah dan morfologi folikel preantral ovarium tikus setelah vitrifikasi selama 48 jam. 20 Sprague-Dawley betina berumur 12 minggu digunakan sebagai hewan model penelitian. Folikel yang diuji merupakan folikel tahap preantral yang berasal dari ovarium kanan saja. Sampel ovarium utuh (n = 20) dibagi ke dalam 7 kelompok yaitu KK, KKP 1, KKP 2, KKP 3, KP 1, KP 2, dan KP 3. Kelompok kontrol (KK) merupakan ovarium segar tanpa vitrifikasi. Kelompok kontrol perlakuan (KKP 1, KKP 2, dan KKP 3) merupakan ovarium yang diberi perlakuan vitrifikasi dalam etilen glikol (EG) dengan konsentrasi berturut-turut 3,75%; 7,5%; dan 15%. Kelompok perlakuan (KP 1, KP 2, dan KP 3) merupakan ovarium yang diberi perlakuan vitrifikasi dalam kombinasi EG dan kuning telur (1 : 1) dengan konsentrasi masing-masing berturut-turut 3,75%; 7,5%; dan 15%. Setelah diisolasi dari tikus, ovarium KK segera difiksasi sedangkan ovarium KKP dan KP dibekukan dalam nitrogen cair (−196 ° C). Setelah 48 jam, sampel ovarium dicairkan dan difiksasi. Seluruh kelompok ovarium difiksasi selama 48 jam. Masing-masing ovarium lalu dibuat sayatan histologis dengan ketebalan 5 um, kemudian diwarnai dengan pewarna hematoksilin eosin (HE). Folikel preantral ovarium diamati keutuhan morfologinya dan dihitung jumlahnya. Data hasil pengamatan menunjukkan bahwa berdasarkan uji Kruskall-Wallis (P < 0,05) tidak terdapat perbedaan yang signifikan pada jumlah folikel preantral ovarium 48 jam pascavitrifikasi. Berdasarkan hal tersebut, dapat disimpulkan bahwa vitrifikasi menggunakan kombinasi etilen glikol (EG) dan kuning telur tidak berpengaruh dalam mempertahankan folikel preantral ovarium pascavitrifikasi.
The study was conducted to evaluate the effect of combination of ethylene glycol (EG) as intracellular cryoprotectant and egg yolk as extracellular cryoprotectant with varied concentrations in maintaining morphology and counts of ovarian pre-antral follicles after vitrification for 48 hours. Twenty 12 week old Sprague-Dawley female rats were used as animal models. The follicles tested were preantral stage follicles from right ovary only. Whole ovary samples (n = 20) were divided into 7 groups, namely NC; TCG 1; TCG 2; TCG 3; TG 1; TG 2; and TG 3. The control group (NC) consists of fresh ovaries without vitrification. The treatment control group (TCG 1; TCG 2; and TCG 3) consists of ovaries treated with vitrification in ethylene glycol (EG) with a concentration of 3.75%; 7.5%; and 15%, respectively. The treatment group (TG 1; TG 2; and TG 3) were ovaries treated with vitrification in a combination of EG and egg yolk (ratio 1: 1) with a concentration of 3.75%; 7.5%; and 15%, respectively. After being isolated from the rats, the NC ovaries were immediately fixated while the TCG and TG ovaries were frozen in liquid nitrogen (−196 °C). After 48 hours, the ovary sample is thawed and fixated. All ovarian groups were fixated for 48 hours. Each of the ovaries was then made a histological incision with a thickness of 5 µm, then stained with a hematoxylin eosin (HE). Ovarian pre-antral follicles were observed for the integrity of the structure and counted. The data based on Kruskall-Wallis test (P < 0.05) show that there were no significant differences in the number of ovarian preantral follicles 48 hours after vitrification. It can be concluded that vitrification using a combination of ethylene glycol (EG) and egg yolk has no effect in maintaining ovarian preantral follicles after vitrification.

Produk emulsifier lesitin merupakan agen penstabil yang baik digunakan untuk industri makanan, farmasi dan kosmetik. Dalam industri makanan, sebagain besar egen pengemulsi yang digunakan merupakan tipe emulsi minyak dalam air. Dalam penelitian ini, lesitin yang telah diekstrak dari kuning telur, dimodifikasi melalui reaksi hidrolisis enzimatik menggunakan enzim papain. Modifikasi ini akan merubah struktur molekulnya yang menjadikan lesitin lebih stabil dalam tipe emulsi minyak dalam air O/W. Penelitian ini dilakukan untuk menentukan kondisi reaksi hidrolisis yang optimum, meliputi waktu reaksi dan jumlah enzim papain yang digunakan. Penentuan hasil lesitin termodifikasi dilakukan dengan beberapa pengujian seperti uji stabilitas, bilangan saponifikasi, bilangan asam, tegangan permukaan dan zeta potensial. Dari hasil uji yang telah dilakukan, diperoleh yield lesitin tertinggi sebesar 10,42 melalui ekstraksi menggunakan pelerut etanol absolut. Stabilitas emulsi untuk jenis O/W dicapai pada lesitin dengan waktu reaksi selama 40 menit yang mampu stabil hingga 43 jam dan lesitin dengan pemakaian enzim papain sebanyak 4 yang stabil hingga 31 jam. Tegangan permukaan terendah dicapai pada lesitin hasil hidrolisis selama 40 menit dengan nilai sebesar 48,52 dyne/cm serta lesitin hasil hidrolisis dengan 2 enzim papain dengan nilai tegangan permukaan sebesar 48,68 dyne/cm. Nilai zeta potensial dari lesitin hasil hidrolisis selama 40 menit memiliki nilai 99,2 mV dan pada lesitin hasil hidrolisis dengan 2 enzim papain memiliki nilai 94,8 mV. Hasil tersebut diperoleh sebagai akibat dari putusnya rantai asam lemak tunggal pada struktur senyawa lesitin yang dapat ditunjukkan dengan instrumentasi FTIR.
The lecithin emulsifier product is a good stabilizer agent used for food, pharmaceutical and cosmetic industries. In the food industry, most of the emulsifying emulsifier used is a type of oil in water emulsion. In this study, lecithin extracted from egg yolk was modified by enzymatic hydrolysis reaction using papain enzyme. This modification will change the molecular structure which makes lecithin more stable in the type of oil in water emulsion O W. This study was conducted to determine the optimum hydrolysis reaction conditions, including reaction time and amount of papain enzyme used. Determination of modified lecithin yield was performed by several tests such as stability test, saponification number, acid number, surface tension and potential zeta. From the results of tests that have been done, obtained the highest lecithin yield of 10,42 through extraction using absolute ethanol solvent. Emulsion stability for the O W type was achieved in 40 minute hydrolyzed lecithin which stable up to 43 hours and hydrolyzed lecithin using 4 papain enzyme with stable up to 31 hours. The lowest surface tension was achieved in 40 minute hydrolyzed lecithin with value of 48,52 dyne cm and hydrolyzed lecithin using 4 papain enzyme with surface tension value 48,68 dyne cm. The potential zeta value of 40 minute hydrolyzed lecithin has a value of 99.2 mV and on hydrolyzed lecithin using 4 papain enzyme has a value of 94,8 mV. The results are obtained as a result of the breaking of a single fatty acid chain from the structure of lecithin which can be demonstrated by FTIR instrumentation.

The objective of the research is to develop a vision machine to detect eggs fertility 0f 5 days in incubation process. A visual sensor, a webcam connected to a computer, was used as image acquisition of some eggs. The eggs were near a source, called IRLED, which transmitted the infrared wave. The images were then processed by software to give useful information for user if the eggs are fertile or not. The system was designed to give information and decision which is not only more accurate but also faster than manual process. The result of research shows that the fertile egg image has gray grade on maximum frequency, between 23 and 27. The infertile one has gray grade between 34 and 43. This different gray grade of the fertile and infertile image is because of different near infrared absorption between the fertile and fertile one. The vision machine could also detect the fertile and infertile 144 samples with the accuracy 92.36%. The reasons of error are wrong position of some eggs, sun ray disturbance and the thick of eggeshell. The detection rate of the machine is 360 eggs per minute.

Penelitian pengaruh pemberian berbagai konsentrasi kuning telur ayam kampung terhadap kualitas spermatozoa ikan koi Cyprinus carpio, Linnaeus 1758 48 jam pascakriopreservasi dilakukan untuk mengetahui konsentrasi optimum kuning telur ayam kampung yang dapat digunakan untuk menjaga kualitas spermatozoa ikan koi. Semen ikan koi diperoleh dengan cara stripping pada bagian urogenital ikan koi. Evaluasi semen ikan koi dilakukan secara makroskopis volume, pH, dan warna semen dan mikroskopis persentase motilitas, viabilitas, dan abnormalitas. Penelitian menggunakan larutan pengencer metanol 10 dan berbagai konsentrasi kuning telur ayam kampung dengan perbandingan semen dan pengencer 1:4. Perlakuan yang diberikan yaitu kuning telur ayam kampung dengan konsentrasi 0 kontrol, 5, 10, 15, 20, dan 25. Pembekuan dilakukan pada suhu -34 oC selama 48 jam. Data yang diperoleh menunjukkan distribusi normal dan bervariasi homogen setelah dilakukan uji normalitas Shapiro-Wilk dan uji homogenitas Levene. Hasil uji Analisis Variansi ANAVA satu arah menunjukkan pengaruh perlakuan kuning telur ayam kampung.
Research about various effect of free range chicken egg yolk concentration to spermatozoa quality of Koi Fish Cyprinus carpio, Linnaeus 1758 48 hours post cryopreservation was done to know optimum concentration of free range chicken egg yolk that can still keep the quality of koi fish spermatozoa. Milt sperm koi fish was collected by stripping at urogenital area of the fish. Koi fish sperm evaluation was done macroscopically volume, pH, milt color and microscopically percentage of motility, viability, and abnormality of spermatozoa. The research was using dilution solution methanol 10 and various concentrations of free range chicken egg yolk with ratio 1 4. The given concentration of free range chicken egg yolk was 0 control, 5, 10, 15, 20, and 25. Freezing was done at 34 oC for 48 hours. The data obtained showed normal and homogenous after processed with the Shapiro Wilk normality test and Levene homogenity test. The one factor ANOVA showed that various concentration of free range chicken egg yolk had effect.
