"Latar Belakang
Penuaan merupakan suatu proses yang tidak dapat dihindari. Namun, pencegahan penyakit terkait usia yang seringkali menyertainya dapat dilakukan dengan memodifikasi faktor risiko utamanya, termasuk inflamasi. Salah satu pendekatan yang dapat digunakan adalah dengan memanfaatkan tanaman yang kaya antioksidan. Rosella (Hibiscus sabdariffa Linn.), terbukti memiliki aktivitas antioksidan yang tinggi. Namun, saat ini belum ada penelitian spesifik yang menilai dampaknya terhadap biomarker khusus penuaan seperti Sirtuin 1.
Metode
Penelitian eksperimental in vivo ini dilakukan pada 18 tikus Sprague Dawley jantan yang dibagi secara acak kedalam 3 kelompok: kontrol (K) yang merupakan tikus muda normal yang diberi pakan standar; Accelerated Aging (AA) yang diberi pakan tinggi lemak dan D-galaktosa; dan Hibiscus sabdariffa Linn. (HSL) yang diberi pakan tinggi lemak, D-galaktosa, dan ekstrak etanol bunga H. sabdariffa dengan dosis 400 mg/KgBB. Perlakuan dilakukan selama 6 minggu. Pada akhir penelitian, kadar SIRT1 diukur menggunakan kit ELISA standar.
Hasil
Perbedaan rerata kadar SIRT1 yang diperoleh antar kelompok tidak signifikan secara statistik.
Kesimpulan
Ekstrak HSL belum mampu meningkatkan kadar SIRT1.

Introduction
Aging is an inevitable process. However, prevention of age-related diseases that often follows can be done by modifying its main risk factors, including inflammation. One approach that can be used is to utilize plants that are rich in antioxidants. Rosella (Hibiscus sabdariffa Linn.), has been shown to have high antioxidant activity. However, there is currently no specific study that assesses its impact on a specific aging biomarker like Sirtuin 1.
Method:
This in vivo experimental study was conducted on 18 male Sprague Dawley rats that were randomly divided into 3 groups: control (K) which were normal young rats that are fed with standard feed; Accelerated Aging (AA) which were fed high fat feed and D-galactose; and Hibiscus sabdariffa Linn. (HSL) which were fed high fat feed, D-galactose, and the ethanol extract of H. sabdariffa flowers at a dose of 400 mg/KgBW. The treatment was carried out for 6 weeks. At the end of the study, SIRT1 levels were measured using a standard ELISA kit.
Results:
The difference in mean SIRT1 levels obtained between groups was not statistically significant.
Conclusion:
HSL extract has not yet been able to increase SIRT1 levels.

"Pendahuluan: Data Susenas tahun 2022 yang dikeluarkan oleh Badan Pusat Statistik (BPS) mencatat bahwa 10,48% penduduk Indonesia adalah lansia dengan 2 dari 5 lansia memiliki keluhan kesehatan. Hal ini bisa disebabkan karena banyaknya faktor ekstrinstik maupun intrinsik yang menyebabkan kerusakan seluler sehingga menginduksi protein p53 untuk melakukan cellular senescence. Senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dalam ekstrak tanaman Plectranthus amboinicus yang kaya akan antioksidan dipercaya dapat menghambat proses cellular senescence sehingga dapat menekan proses penuaan. Dengan itu, tujuan dari penelitian ini adalah untuk mengetahui interaksi antara komponen ligan pada senyawa aktif thymol, carvacrol, dan flavonoid yang terkandung dalam Plectranthus amboinicus dengan protein target p53 yang berkaitan dengan aktivitas cellular senescence pada proses penuaan. Metode: Studi in silico melalui molecular docking (penambatan molekuler) untuk menguji interaksi molekuler dari ligan senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dari tanaman Plectranthus amboinicus terhadap reseptor protein target p53 yang bertanggung jawab dalam proses cellular senescence. Hasil interaksi yang didapat merupakan sebuah histogram yang akan dianalisis dan diinterpretasikan untuk mengetahui afinitas ikatan (binding affinity) dari interaksi yang terjadi. Hasil: Terjadi interaksi antara ligan senyawa thymol, carvacrol, dan flavonoid (quercetin, luteolin, apigenin, rutin, eriodictyol) dari tanaman Plectranthus amboinicus terhadap reseptor protein target p53. Kesimpulan: Plectranthus amboinicus berpotensi menjadi agen anti-aging yang dapat menghambat dan memperlambat proses dari penuaan melalui studi in silico. Akan tetapi, perlu dilakukan penelitian lebih lanjut mengenai mengenai sifat-sifat dari Plectranthus amboinicus untuk mengetahui potensi tanaman ini terhadap tubuh manusia yang belum diketahui.

Introduction: The 2022 Susenas data stated by the Central Statistics Agency (BPS) notes that 10.48% of Indonesia’s population are elderly with 2 out of 5 having health complaints. This could be due to the many extrinsic and intrinsic factors that causes cellular damage thereby inducing the p53 protein to perform cellular senescence. Compounds of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the extract of the Plectranthus amboinicus plant which are rich in antioxidants are believed to have the potential to inhibit the cellular senescence process so as to suppress the aging process. The purpose of this research is to investigate the interaction between the ligand of thymol, carvacrol, and flavonoid compounds from Plectranthus amboinicus with the p53 target protein which is associated with cellular senescence activity in the aging procress. Methods: In silico study through molecular docking to examine the molecular interactions of the ligands of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the Plectranthus amboinicus plant against the target protein receptor p53 which is responsible for the process of cellular senescence. The interaction results obtained is a histogram which will be analyzed and interpreted to determine the binding affinity of the interactions that occured. Results: Interactions existed between the ligands of thymol, carvacrol, and flavonoids (quercetin, luteolin, apigenin, rutin, eriodictyol) from the Plectranthus amboinicus plant against the target protein receptor p53. Conclusion: Plectranthus amboinicus has the potential to be an anti-aging agent that can inhibit and slow down the process of aging through in silico studies. However, it is necessary to carry out further research regarding the properties of Plectranthus amboinicus to determine the unknown potential of this plant to the human body.

"Penuaan kulit merupakan proses alami yang terjadi pada manusia dan ditandai dengan penurunan elastisitas, kelembapan, dan munculnya garis-garis halus serta kerutan. Berbagai upaya dapat dilakukan untuk mencegah penuaan kulit terjadi, salah satunya dengan menggunakan produk perawatan kulit yang mengandung bahan aktif seperti senyawa antioksidan. Kulit buah manggis (Garcinia mangostana L.) diketahui memiliki kandungan senyawa antioksidan, salah satunya, ∝-mangostin yang berpotensi sebagai antioksidan yang dapat mencegah penuaan. Penelitian ini dilakukan bertujuan untuk mendapatkan kondisi operasi yang optimum untuk memperoleh ekstrak kulit buah manggis dalam natural deep eutectic solvent (NADES) berbasis kolin klorida dan 1,2-propanadiol serta mengembangkan formulasi krim topikal anti penuaan dari hasil ekstraksi tersebut. Ekstraksi dilakukan dengan metode pengadukan menggunakan thermoshaker pada suhu ruang, dengan variasi rasio massa padat-cair dan waktu ekstraksi. Kondisi optimum diperoleh pada rasio 1:10 selama 4 jam, menghasilkan kandungan ∝-mangostin sebesar 6,24 mg/mL. Ekstrak kemudian diformulasikan ke dalam krim topikal sebesar 20%, menghasilkan sediaan dengan ukuran partikel rata-rata 376,87 nm, zeta potensial -52,37 mV, dan polydispersity index sebesar ,85. Krim memiliki warna putih kecoklatan, aroma virgin coconut oil, tekstur kental, dan tidak lengket saat diaplikasikan. Uji aktivitas antioksidan menunjukkan kandungan total fenolik sebesar 42,18 mg GAE/g simplisia dan 17,55 mg GAE/g emulgel, serta nilai FRAP sebesar 32,39 μmol Fe²⁺/g simplisia dan 7,53 μmol Fe²⁺/g emulgel. Formulasi ini menunjukkan potensi sebagai krim topikal anti penuaan yang stabil dan efektif.

Skin aging is a natural physiological process in humans, characterized by a decline in skin elasticity and moisture, along with the emergence of fine lines and wrinkles. Preventive strategies to delay skin aging include the use of skincare products formulated with active compounds such as antioxidants. The peel of mangosteen (Garcinia mangostana L.) is known to contain potent antioxidant compounds, notably ∝-mangostin, which exhibits promising anti-aging properties. This study aimed to determine the optimal extraction conditions for mangosteen peel using a Natural Deep Eutectic Solvent (NADES) composed of choline chloride and 1,2-propanediol, and to develop a topical anti-aging cream based on the resulting extract. Extraction was performed using a stirring method with a thermoshaker at ambient temperature, varying the solid-to-liquid mass ratio and extraction time. The optimal condition was identified at a 1:10 ratio for 4 hours, yielding an ∝-mangostin concentration of 6.24 mg/mL. The extract was incorporated into a topical cream at a concentration of 20%, resulting in a formulation with an average particle size of 376.87 nm, a zeta potential of -52.37 mV, and a polydispersity index of 1.85. The cream exhibited a brownish-white color, a virgin coconut oil aroma, a thick consistency, and a non-sticky texture upon application. Antioxidant activity assays revealed a total phenolic content of 42.18 mg GAE/g in the mangosteen extract and 17.55 mg GAE/g in the emulgel, with FRAP values of 32.39 μmol Fe²⁺/g and 7.53 μmol Fe²⁺/g, respectively. These findings indicate that the formulation has strong potential as a stable and effective anti-aging topical cream."
