"Hiperurisemia adalah keadaan dimana terjadi peningkatan kadar asam urat dalam darah. Xantin oksidase berperan dalam oksidasi hipoxantin dan xantin menjadi asam urat. Salah satu pengobatan hiperurisemia adalah menghambat xantin oksidase dalam proses pembentukan asam urat. Tujuan penelitian ini untuk mengetahui penghambatan aktivitas xanthine oxidase pada senyawa hasil isolasi. Serbuk simplisia di maserasi dengan metanol, kemudian dilakukan partisi dengan pelarut heksan, kloroform, etil asetat, n-Butanol dan air. Fraksi n-butanol dengan nilai IC50 3,68 μg/mL, fraksi ini dilakukan pemisahan secara kromatografi kolom dengan eluen metanol air. Pada uji aktivitas, isolat memiliki aktivitas penghambatan terhadap xantin oksidase sebesar 2,79 μg/mL. Uji kinetika enzim menunjukkan bahwa isolat mempunyai aktivitas penghambatan kompetitif. Dari hasil identifikasi yang dilakukan diduga isolat yang diperoleh merupakan glikosida dengan aglikon berupa flavonoid.

Hyperuricemia is increasing of uric acid in blood. Xanthine oxidase is an enzym that plays a role in the oxidation hypoxanthine and xanthine into uric acid. One of the hyperuricemia remedies is inhibit xantin oxidase to produce uric acid.

The purpose of this study was to find inhibitory activity xanthine oxidase of compound from isolation. The sample powder was maserated by methanol solvent, and the extract was partitioned by n-hexane, chloroform, ethyl acetat, and n-buthanol. n- Buthanol fraction with IC50 values 3,68 μg/mL, this fraction was separated process by column chromatography with methanol/water as eluent.

Activity assay showed that the isolate has activity to inhibit xanthine oxidase with IC50 values 2,79 μg/mL. The result of enzym kinetics showed that isolate has a competitive inhibitory activity. Phytochemical identification indicated that isolate contained glycoside with flavonoid as aglycon."
