Abstrak :
Hipertensi merupakan masalah kesehatan yang prevalensinya masih tinggi di Indonesia. Mayoritas pengidap hipertensi mengandalkan obat golongan penghambat ACE (angiotensin-converting enzyme) sebagai terapi obat hipertensi mereka. ACE dapat dihambat dengan cara mengikat ion Zn2 atau asam amino pada situs aktif ACE. Penelitian sebelumnya menunjukkan bahwa ekstrak biji melinjo memiliki aktivitas penghambatan ACE secara in vitro. Dalam penelitian ini, dilakukan penambatan molekuler pada beberapa senyawa dalam ekstrak biji melinjo terhadap makromolekul ACE guna menganalisis senyawa mana dari dalam ekstrak biji melinjo tersebut yang memiliki aktivitas penghambatan ACE. Hasil penambatan molekuler dengan AutoDock4Zn menunjukkan bahwa resveratrol, gnetol, isorhapontigenin, gnetin c, trans-?-viniferin, gnemonol K, gnemonol M, dan aglikon gnemonosida B berpotensi memiliki aktivitas penghambat ACE. Beberapa ligan tersebut mampu berikatan dengan ion Zn2 melalui ikatan kation-phi. Selanjutnya, dilakukan simulasi dinamika molekuler pada ligan-ligan tersebut dalam rentang waktu 20 ns untuk menganalisis kestabilan ikatan ligan terhadap ion Zn2 dan ikatan ligan terhadap residu asam amino pada sisi aktif ACE. Berdasarkan hasil perhitungan nilai energi bebas ikatan dengan metode MM-GBSA dan MM-PBSA, gnetin C memiliki afinitas tertinggi terhadap ACE daripada ligan uji lain pada suhu 300 K, sedangkan pada suhu 310 K afinitas tertinggi dimiliki oleh gnemonol K.

Hypertension is a health problem, in which the prevalence remains high in Indonesia. The majority of hypertensive patients rely on ACE angiotensin converting enzyme inhibitors as their drug therapy of hypertension. ACE catalytic mechanism can be inhibited by ligands via the zinc ion or amino acids interactions in the active site of ACE. Recent study demonstrated that melinjo seed extracts have ACE inhibitory activity in vitro. In this study, we conducted molecular docking of several substances extracted from melinjo seeds to analyze which substances have ACE inhibitory activity. Molecular docking results using AutoDock4Zn indicated that resveratrol, gnetol, isorhapontigenin, gnetin C, trans viniferin, gnemonol K, gnemonol M and aglycone of gnemonosida B potentially have ACE inhibitory activity. Some of these ligands are able to bind zinc ion via cation pi interactions. We also conducted molecular dynamics simulations involving these ligands within 20 ns to analyze the stability of the interactions between zinc ligand and ligand amino acids in the active site. According to the binding free energy calculations using MM GBSA and MM PBSA methods, gnetin C showed the highest affinity for ACE among other ligands at a temperature of 300 K, while at a temperature of 310 K the highest affinity belongs to gnemonol K.

Praktik kerja profesi di Apotek Kimia Farma No 362 Cibinong Periode Bulan Maret Tahun 2018 bertujuan untuk memahami tugas dan tanggung jawab Apoteker dalam pengelolaan apotek; melakukan praktik pelayanan kefarmasian sesuai dengan ketentuan perundang-undangan dan etika yang berlaku, menambah wawasan, pengetahuan, keterampilan, dan pengalaman praktis untuk melakukan pelayanan kefarmasian di apotek; serta mengidentifikasi kesesuaian Apotek Kimia Farma No. 362 Cibinong dalam menerapkan pelayanan kefarmasian terhadap standar pelayanan kefarmasian berdasarkan perundang-undangan yang berlaku. Praktik kerja profesi di apotek ini dilaksanakan selama satu bulan empat minggu . Adapun tugas khusus yang diberikan yakni mengenai ldquo;Pemanfaatan Leaflet sebagai Media Penunjang Kegiatan Pelayanan Informasi Obat PIO Penyakit GERD di Apotek Kimia Farma No. 362 Cibinong rdquo;. Tujuan dari tugas khusus ini adalah untuk melatih Apoteker dalam membuat desain leaflet yang menarik dan mencolok sehingga dapat dijadikan sebagai media penunjang Apoteker dalam melakukan Pelayanan Informasi Obat PIO kepada pasien di apotek mengenai penyakit GERD serta membantu pasien dalam mengenal GERD dan pengobatannya yang tepat.

Apothecary internship program at Apotek Kimia Farma No. 362 Cibinong on January 2018 aimed to figure out the job descriptions and responsibilities of the apothecary in managing pharmacy and practising pharmaceutical care in accordance with applicable laws and ethics; gain the insight, knowledge, skills and practical experience to perform pharmaceutical care in pharmacy; and identify the pharmaceutical care service of Apotek Kimia Farma No. 362 in compliance with the regulation of good pharmaceutical practice. The internship was conducted for one month four weeks . In this internship was also undertaken the special assignment entitled Utilization of Leaflets as Supporting Media of Drug Information Services of GERD at Apotek Kimia Farma No. 362 Cibinong . The purpose of this special assignment was to train an Apothecary in creating attractive leaflet design that can be used as a supporting media in performing Drug Information Services of GERD to patients at a pharmacy and guide patients to learn more about GERD and its treatment.

Praktik kerja profesi di PT Agrinusa Jaya Santosa Periode Bulan Maret-April Tahun 2018 bertujuan untuk mengetahui peran dan tanggung jawab Apoteker di industri obat hewan serta untuk mengidentifikasi penerapan Cara Pembuatan Obat Hewan yang Baik CPOHB di industri obat hewan. Praktik kerja profesi ini dilakukan selama 2 bulan 8 minggu . Adapun tugas khusus yang diberikan selama praktik kerja profesi yakni mengenai ldquo;Analisis Penyimpangan pada Penerapan Prosedur Produksi Induk Dan CPOHB di Ruang Pengolahan PT Agrinusa Jaya Santosa rdquo;. Tujuan dari pelaksanaan tugas khusus ini adalah untuk menilai efektivitas penerapan Prosedur Produksi Induk di PT Agrinusa Jaya Santosa, menilai kepatuhan operator dalam mengikuti instruksi dari Prosedur Pengolahan Induk PPI , dan menganalisis penerapan CPOHB di ruang pengolahan PT Agrinusa Jaya Santosa. Secara garis besar, penerapan PPI masih perlu dievaluasi efektivitasnya, sedangkan penerapan CPOHB di ruang pengolahan telah baik namum masih ada beberapa aspek dalam CPOHB yang harus dievaluasi.

Apothecary internship Program at PT Agrinusa Jaya Santosa on March ndash; April 2018 aimed to figure out the roles and responsibilities of the apothecary in veterinary medicine industry and to identify the application of good manufacturing practice for veterinary medicinal products. The internship was carried out for 2 months 8 weeks . The special assignment was given during internship entitled ldquo;Analysis of Deviation on Application of Master Processing Procedure and Good Manufacturing Practice for Veterinary Medicinal Products in Processing Room of PT Agrinusa Jaya Santosa rdquo;. The objective of this special assignment was to assess the effectiveness of the application of the master processing procedure at PT Agrinusa Jaya Santosa, to asses the operator 39;s compliance in following the instructions of the master processing procedure, and to asses good manufacturing practice for veterinary medicinal products in processing room of PT Agrinusa Jaya Santosa. In general, the application of master processing procedure was still needed to be evaluated its effectiveness, while the application of in good manufacturing practice for veterinary medicinal products in processing room was good, but there were still some aspects in good manufacturing practice for veterinary medicinal products that must be evaluated.

Praktik kerja profesi di PT Anugerah Pharmindo Lestari Periode Bulan Mei Tahun 2018 bertujuan untuk mengetahui peran dan tanggung jawab Apoteker di Pedagang Besar Farmasi PBF serta Penyalur Alat Kesehatan PAK dan mengidentifikasi penerapan Cara Distribusi Obat yang Baik CDOB dan Cara Distribusi Alat Kesehatan yang Baik CDAKB di Pedagang Besar Farmasi PBF dan Penyalur Alat Kesehatan PAK . Praktik kerja profesi ini dilaksanakan selama dua minggu. Selama praktik kerja profesi diberikan tugas khusus mengenai ldquo;Optimasi Waktu Prekondisi Metode Pengemasan Passive Packaging Styrofoam dengan Ice Pack untuk Produk dengan Suhu Penyimpanan 2-8 C rdquo;. Tujuan dari pelaksanaan tugas khusus ini adalah untuk mendapatkan waktu prekondisi optimal yang dibutuhkan styrofoam untuk mencapai suhu konstan 2-8 ?C. Diharapkan waktu tersebut lebih cepat bila dibandingkan dengan waktu yang tercantum dalam instruksi kerja, yakni 30 menit. Prekondisi metode pengemasan passive packaging styrofoam dengan ice pack yang relatif paling optimal adalah dengan menbiarkan ice pack berada di lingkungan suhu kamar 25 oC selama 10 menit sebelum dimasukkan ke dalam styrofoam.

The internship was conducted for two weeks. The special assignment given entitled Optimization of the Precondition Time of Styrofoam Passive Packaging Methods using Ice Pack for Products stored at 2-8 C . The purpose of this special assignment was to obtain the optimal precondition time required by styrofoam to achieve a constant temperature between 2-8 ?C. It was expected that the time was faster compared to the time stated in the work instructions, which was 30 minutes. Optimization result exhibited that the most optimal time was by leaving ice pack at room temperature environment 25 oC for 10 minutes before putting it into the styrofoam.
