Marga Excoecaria banyak terdapat di Indonesia dan mempunyai potensi sebagai sumber senyawa kimia bioaktif. Salah satu tanaman marga ini adalah Sambang Darah (E. cochinchinensis Lour.). Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari ekstrak n-heksan, etil asetat, dan metanol daun E. cochinchinensis Lour. serta mengetahui golongan senyawa kimia yang terkandung dari fraksi teraktif. Daun E. cochinchinensis Lour. dimaserasi menggunakan n-heksana, etil asetat, dan metanol. Aktifitas antioksidan diukur menggunakan metode DPPH. Ekstrak teraktif difraksinasi dengan kromatografi kolom dipercepat. Kemudian dilakukan identifikasi golongan senyawa pada fraksi dengan aktifitas antioksidan teraktif. Hasil uji menunjukkan nilai IC50 dari ekstrak metanol, etil asetat, dan n-heksana secara berturut-turut adalah 2,81 ppm; 3,21 ppm; dan 48,77 ppm. Fraksi teraktif adalah fraksi M2 dengan nilai sebesar IC50 2,49 ppm dengan kandungan flavonoid, tanin, antrakuinon, glikosida, dan saponin. ......Excoecaria genus widely available in Indonesia and has potential as a source of bioactive chemical compounds. One of the plant of this genus is Sambang Darah (E. cochinchinensis Lour). This research aims to determine the antioxidant activity of the extracts of n-hexane, ethyl acetate, and methanol E. cochinchinensis Lour. leaves and to know the group of chemical compounds which contained the most active fraction. E. cochinchinensis Lour. leaves macerated using n-hexane, ethyl acetate, and methanol. Antioxidant activity was measured using DPPH method. The most active extract fractionated by accelerated column chromatography. Phytochemical screening were done at the most acive fraction. Test results showed IC50 value of the extract of methanol, ethyl acetate and n-hexane respectively were 2.81 ppm, 3.21 ppm, and 48.77 ppm. Fraction M2 is the most active fraction with IC50 value of 2.49 ppm and it contained flavonoids, tannins, anthraquinone, glycosides and saponins
