"Oregano (Origanum vulgare) adalah tanaman yang berasal dari daratan Eropa, khususnya di daratan Yunani. Selain digunakan sebagai rempah, sejak dahulu kala oregano telah digunakan oleh masyarakat Eropa untuk mengatasi berbagai penyakit seperti gangguan pernapasan dan pencernaan. Oregano diyakini memiliki kandungan karvakrol dan timol yang sudah terbukti memiliki efek farmakologis sebagai antimikroba. Oleh karena itu, penelitian ini menjadi penting untuk mengetahui kadar karvakrol dan timol sebagai antimikroba dalam oregano. Pada penelitian ini, dilakukan penetapan kadar karvakrol dan timol dalam minyak esensial oregano menggunakan kromatografi gas dengan detektor ionisasi nyala. Isolasi dilakukan terhadap simplisia oregano kering dengan metode Solvent-free Microwave Extraction (SFME) sehingga diperoleh yield sebesar 0,00422 gram/gram simplisia atau 0,422%. Kondisi optimum dalam analisis penetapan kadar karvakrol dan timol dalam minyak esensial oregano, yaitu pada suhu terprogram 150-1700C dengan kenaikan 20C/menit dimana suhu injektor dan detektor diatur pada 2000C dan laju alir fase gerak Helium 1,2 mL/menit. Hasil penetapan kadar timol pada minyak hasil isolasi 0,156% dan karvakrol 3,714%. Sedangkan dalam sampel yang diperoleh di pasaran didapatkan kadar timol 0,119% dan karvakrol 0,143%.

Oregano (Origanum vulgare) is a European aromatic plant that come from Greece. Oregano is widely used as food additive, and also used to heal digestive disorder a long time ago. Researchers have assume that oregano has high content of carvacrol and thymol, two widely used drugs. This research studied about determination of carvacrol and thymol using GC-FID. The isolation of volatile oil has been done using Solvent-free Microwave Extraction (SFME) method.

The yield of the isolation is 0,00422 gram/gram herbs or 0,422%. The optimum analysis condition for determination of carvacrol and thymol is at column program temperature 150-1700C, 20C/menit when the temperature of detector and injector is set at 2000C and the flow rate of Helium as carrier gas is 1,2 mL/minute. The result of determination of thymol from isolated oil is 0,156% while carvacrol is 3,714%. The determination of carvacrol and thymol is also done from oil sample and the result is 0,119% thymol and 0,143% carvacrol."
