Abstrak :
Tuberkulosis merupakan salah satu penyakit yang menyerang saluran pernapasan dan salah satu obat yang digunakan adalah antibiotik rifampisin. Walaupun terapi oral rifampisin untuk pasien tuberkulosis sudah tersedia, terapi secara inhalasi yang menargetkan langsung ke paru dan alveolus diharapkan memberikan hasil yang lebih baik. Oleh karena itu diperlukan inovasi serbuk kering inhalasi yang dapat digunakan dalam regimen terapi tuberkulosis. Penelitian terhadap pengembangan obat TB dalam bentuk serbuk kering sudah ada, namun belum ada yang mengombinasikan kitosan dengan gelatin. Pada penelitian ini dilakukan formulasi, karakterisasi, dan evaluasi sitotoksisitas dari lima serbuk kering inhalasi rifampisin dengan pembawa kitosan-gelatin dengan variasi konsentrasi kitosan dan gelatin. Serbuk dibuat menggunakan metode semprot kering, kemudian dianalisis bentuk dan ukuran partikel geometris dan aerodinamisnya, lalu diuji pelepasannya dalam larutan dapar fosfat pH 7,4 dan SLS 0,05%, dan dalam larutan KHP pH 4,5. Formulasi dengan profil pelepasan yang paling baik diambil untuk diuji sitotoksisitasnya. Berdasarkan hasil penelitian dipilih F3 (Kit-Gel 2:1) sebagai formula terbaik dengan bentuk partikel sferis, rentang diameter partikel geometris 0,825-1,281 μm dan ukuran partikel aerodinamis 11,857 ± 1,259 μm, dengan persentase rifampisin kumulatif yang terdisolusi dalam dapar fosfat sebesar 45,894 ± 0,876% dan dalam larutan KHP sebesar 42,117 ± 0,912%. Uji sitotoksisitas dilakukan dengan metode MTT assay dan parameter viabilitas sel. Serbuk F3 lebih aman digunakan daripada rifampisin dalam konsentrasi 0,1 mg/mL dengan viabilitas sel sebesar 91,68%. ......Tuberculosis is a respiratory tract disease in which one of the first-line agents used is rifampicin. Oral rifampicin for TB patients is widely available, but inhalation therapy that targets the lungs and alveolus directly could give a better outcome. This drives the need for developing a dry inhalation powder that could be incorporated into the therapeutic regimen of TB. Previous studies on the development of TB medication have been performed, but the application of combination of excipients of chitosan and gelatin as carrier has not been studied. In this study, formulation, characterization, and cytotoxicity evaluation of five rifampicin dry inhalation powders with various concentration of its chitosan-gelatin carrier were studied. The powder was produced with dry-spraying method. Its shape, aerodynamic and geometric particle size were then analyzed. Its releasing profile in phosphate buffer (pH 7.4 and SLS 0.5%) and potassium hydrogen phthalate (pH 4.5) was also tested. Formulation with the best releasing profile was used for cytotoxicity test. F3 (Chit-Gel 2:1) showed the best profile with spherical particle shape and geometric and aerodynamic particle size 0.825-1.281 μm and 11.857 ± 1.259 μm respectively, the cumulative rifampicin percentage dissolved in phosphate buffer and potassium hydrogen phthalate (KHP) were 45.894 ± 0.876% and 42.117 ± 0.912%, respectively. Cytotoxicity evaluation was conducted using MTT assay method with cell viability as the parameter. F3 is less cytotoxic than rifampicin in 0.1 mg/mL with cell viability 91.68%.

Profesi apoteker memegang peranan penting dalam pekerjaan kefarmasian. Salah satu hal penting yang harus dilakukan untuk menjadi seorang apoteker yang profesional dan kompeten adalah berpartisipasi secara langsung dalam praktik kefarmasian. Oleh karena itu, calon apoteker dituntut untuk menjalani praktik profesi sebagai bekal dan pengalaman untuk memahami peran apoteker dan meningkatkan kompetensi sebelum memasuki dunia kerja. Praktik Kerja Profesi Apoteker dilaksanakan di PT. Kalbio Global Medika periode Januari – Februari 2022 dan Apotek Kimia Farma Jatirahayu periode Mei 2022. Melalui proses PKPA di industri farmasi dan apotek tersebut, calon apoteker diharapkan mampu memperoleh wawasan, keterampilan, pengetahuan, dan pengalaman yang sesuai untuk melakukan pekerjaan kefarmasian. ......Pharmacists hold an important role in pharmaceutical practices. One of the most crucial things that should be done to be a professional and competent pharmacist is to participate directly in the practice of pharmacy. Therefore, prospective pharmacists are required to undergo professional practice to gather knowledge and experience to understand the role of pharmacists and increase competence before entering the world of work. Professional Practice of Pharmacist are held at PT. Kalbio Global Medika from January to February 2022 and Apotek Kimia Farma on May 2022. Through these activities, prospective pharmacists should be able to obtain the proper insight, skills, knowledge, and experience to perform well in pharmaceutical practice.
