"Diabetes melitus dianggap sebagai suatu masalah kesehatan yang serius. Komplikasi diabetes melitus seperti komplikasi mikrovaskular dan makrovaskular, sering menimbulkan kecacatan dan kematian. Inhibitor α-glukosidase dapat digunakan sebagai terapi diabetes melitus karena memiliki potensi untuk menormalkan kadar gula darah. Penelitian sebelumnya menunjukkan bahwa ekstrak etanol dari herba meniran (Phyllanthus niruri L.) memiliki penghambatan aktivitas α-glukosidase yang kuat dengan IC50 = 2,32 μg/mL. Penelitian lainnya menunjukkan bahwa fraksi metanol dari ekstrak etanol herba meniran merupakan fraksi paling aktif (IC50 = 1,67 μg/mL). Penelitian ini bertujuan untuk mengetahui kemampuan penghambatan aktivitas α-glukosidase pada fraksi-fraksi dari ekstrak metanol herba meniran dan golongan senyawa dari fraksi paling aktif. Serbuk herba meniran direfluks berturut-turut dengan pelarut n-heksana, etil asetat, dan metanol. Ekstrak metanol dikromatografi menggunakan fase diam Sephadex LH-20 dan metanol 50% sebagai eluennya. Enam fraksi terpilih diuji penghambatan aktivitas α-glukosidase. Uji penghambatan aktivitas α-glukosidase dilakukan dengan metode spektrofotometri menggunakan microplate reader. Nilai IC50 dari fraksi paling aktif adalah18,82 μg/mL. Fraksi ini memiliki jenis penghambatan nonkompetitif. Penapisan fitokimia menunjukkan bahwa fraksi paling aktif mengandung flavonoid dan glikosida.

Diabetes mellitus is recognized as a serious global health problem. The complications of diabetes mellitus such as microvascular and macrovascular complications, often resulting in morbidity and mortality. α-Glucosidase inhibitors have been used as therapy of diabetes mellitus because of the potential to normalize blood-glucose level. The previous study showed the ethanolic extract of Phyllanthus niruri herb had a potent α-glucosidase inhibitory activity with IC50=2.32 μg/mL. The other study showed the methanolic fraction from ethanolic extract of Phyllanthus niruri herb as the most active fraction (IC50= 1.67 μg/mL). This research aims to know α-glucosidase inhibitory activity from fractions of methanolic extract of Phyllanthus niruri herb and the class compounds from the most active fraction. The powder of Phyllanthus niruri herb was refluxed by nhexane, ethyl acetate, and methanol, successively. The methanolic extract was chromatographed by Sephadex LH-20 as stationery phase and 50% methanol as mobile phase. Selected six fractions were assayed for α-glucosidase inhibition activity. The α-glucosidase inhibition assay was performed by spectrophotometric method with microplate reader. The IC50 value of the most active fraction was 18.82 μg/mL. This fraction had a noncompetitive inhibitory activity. Phytochemical screening showed the most active fraction contained flavonoid and glycoside."
