Abstrak :
Peran protein retinoblastoma (pRb) dalam pencegahan pembentukan tumor diinhibisi oleh interaksinya dengan protein E7 HPV pada kanker serviks. Oleh sebab itu, perlu dilakukan strategi pengembangan uji in vitro untuk analisis interaksi pRb dan E7, terutama dalam pengembangan vaksin HPV berbasis antigen E7. Protein pRb dapat diperoleh dalam bentuk protein rekombinan yang diproduksi pada bakteri Escherichia coli. Penelitian bertujuan untuk memperoleh klona gen RB1 dalam vektor pQE_80L. Sintesis gen RB1 (2787 pb) dilakukan dengan metode PCR overlap extension. Fragmen gen RB1 dan vektor didigesti dengan enzim restriksi BamHI dan SalI kemudian diligasikan dengan enzim T4 ligase. Hasil ligasi ditransformasi ke dalam Escherichia coli TOP10 secara kejut panas. Hasil transformasi diseleksi menggunakan PCR koloni untuk mengidentifikasi keberadaan DNA sisipan. Sebanyak 1 dari 27 koloni yang diseleksi mengandung plasmid rekombinan. Plasmid rekombinan kemudian diisolasi dan diverifikasi dengan digesti dan sekuensing. Hasil analisis digesti dan sekuensing menunjukkan gen RB1 berhasil disisipkan ke vektor pQE_80L. Namun terdapat beberapa mutasi, yaitu substitusi (c.117G>A dan c.2316T>C) serta mutasi delesi (c.719_724delAAACAG).

The role of human retinoblastoma protein (pRb) as tumor suppressor is inhibited by its interaction with HPV E7 protein in cervical cancer. Therefore, it is interesting to develop strategy for development of in vitro assay to analyze pRb and E7 interaction, especially in the development of therapeutic HPV vaccine that is based on E7 antigen. The pRb protein can be provided in the form of recombinant protein that is poduced in Escherichia coli. The study objective was to obtain RB1 gene clone in pQE_80L vector. The synthesis of RB1 gene (2787 pb) was performed by using overlap extension PCR. The RB1 gene fragment and vector was digested by BamHI and SalI restriction enzyme then ligated by T4 ligase enzyme. The ligation product was transformed into Escherichia coli TOP10 with heat shock. The transformation result was screened using colony PCR to identify the presence of insert DNA. There was 1 out of 27 selected colonies that carried the recombinant plasmid. The recombinant plasmid then isolated and verified with digestion and sequencing. The results of digestion and sequencing analysis showed that RB1 gene was successfully inserted into pQE_80L vector. However, there were mutations which were substitution (c.117G>A and c.2316T>C) and deletion (c.719_724delAAACAG).

Praktik kerja profesi di Direktorat Standardisasi Obat, Narkotika, Psikotropika, Prekursor, dan Zat Adiktif Badan Pengawas Obat dan Makanan Periode Bulan Maret Tahun 2018 bertujuan untuk memahami tugas dan tanggung jawab apoteker di BPOM, memiliki pengetahuan tentang tupoksi BPOM, memiliki wawasan, pengetahuan, keterampilan dan pengalaman praktis untuk melakukan pekerjaan di BPOM, dan memiliki gambaran nyata tentang permasalahan kefarmasian di BPOM. Praktik kerja profesi ini dilaksanakan selama empat minggu dengan tugas khusus yaitu ldquo;Kajian Mengenai Elemental Impurities atau Uji Cemaran Logam pada USP 40 rdquo;. Tujuan dari tugas khusus ini adalah agar calon apoteker dapat mengetahui tujuan dari uji cemaran logam dan potensi implementasi dari metode uji cemaran logam kompendial USP 40 di Indonesia.

ABSTRACT
Internship at the Directorate of Drug Standardization, Narcotics, Psychotropics, Precursors, and Addictive Substances of National Agency of Drug and Food Control NADFC Period March 2018 Period aims to understand the duties and responsibilities of the pharmacist and have knowledge about the duties and functions of NADFC, have insight, knowledge, skills and practical experience to do work, and have a real picture of pharmacy issues at NADFC. This professional work practice is carried out for four weeks with a special task, namely Study of Elemental Impurities Testing of USP 40 . The purpose of this special task is so that prospective pharmacists can find out the purpose of the metal contamination test and the potential implementation of the USP 40 compendial elemental impurities testing method in Indonesia.

Praktik kerja profesi di Apotek Atrika Periode Bulan April Tahun 2018 bertujuan untuk memahami tugas dan tanggung jawab apoteker dalam pengelolaan apotek, serta melakukan praktik pelayanan kefarmasian sesuai dengan ketentuan perundang-undangan dan etika yang berlaku, memiliki wawasan, pengetahuan, keterampilan dan pengalaman praktis untuk melakukan praktik kefarmasian di apotek, memiliki gambaran nyata tentang permasalahan praktik kefarmasian serta mempelajari strategi dan kegiatan-kegiatan yang dapat dilakukan dalam rangka pengembangan praktik kefarmasian. Praktik kerja profesi ini dilaksanakan selama empat minggu dengan tugas khusus yaitu 'Rancangan Kerjasama Apotek dengan Praktik Dokter yang Melayani Program Rujuk Balik BPJS di Tanjung Priok'. Tujuan dari tugas khusus ini adalah agar calon apoteker dapat membuat dan mengetahui kelayakan rancangan apotek yang bekerjasama dengan praktik dokter yang melayani program rujuk balik.

Internship at Atrika Pharmacy on April Period aims to understand the duties and responsibilities of pharmacists in pharmacy management, as well as to practice pharmaceutical services in accordance with applicable laws and ethics, have the insight, knowledge, skills and practical experience to undertake pharmaceutical practices in pharmacies, can also have the insight of pharmaceutical practice issues and learn strategies and activities that can be undertaken in the course of pharmaceutical practice development. This internship at was conducted for four weeks with special assignment 'Plan of Pharmacy Partnership with the Practices of Doctors Who Serve the Referral Program of BPJS at Tanjung Priok'. The purpose of the special assignment is that the prospective pharmacist can create and know the feasibility of pharmacy designs that work with the practice of doctors who serve referral program.

Praktik kerja profesi di PT AstraZeneca Indonesia Cikarang Periode Bulan Januari ndash; Februari Tahun 2018 bertujuan untuk mengetahui tugas dan tanggung jawab apoteker di industri farmasi dan memahami penerapan Cara Pembuatan Obat yang Baik CPOB di Industi Farmasi. Selain itu calon apoteker juga dapat memiliki gambaran nyata tentang permasalahan pekerjaan kefarmasian di industri farmasi, memiliki wawasan, pengetahuan, keterampilan, dan pengalaman praktis untuk melakukan pekerjaan kefarmasian di industri farmasi. Tugas khusus yang diberikan yaitu berjudul ldquo;Pembuatan Spesifikasi Fungsional Printer Zebra Series S4M untuk Pencetakan Label pada Kemasan Tersier rdquo;. Tujuan dari pelaksanaan tugas khusus adalah untuk membuat dokumen spesifikasi fungsional dari sistem Printer Zebra Series S4M dalam rangka memenuhi validasi sistem komputerisasi. Internship at PT AstraZeneca Indonesia Cikarang Plant Period January - February 2018 aims to understand the duties and responsibilities of pharmacists in the pharmaceutical industry and the understanding of the application of good manufacturing practice GMP in the pharmaceutical industry. In addition, the pharmacist candidate can also have the insight, knowledge, skills and practical experience to undertake pharmaceutical work in the pharmaceutical industry. The special assignment given is ldquo;The Creation of Zebra Series S4M Functional Specifications for Tertiary Packaging Labels rdquo;. The purpose of this special assignment is to create a functional specification document for Zebra Series S4M Printer system in order to meet computerized system validation. In general, PT AstraZeneca Indonesia has applied 12 aspects of GMP well and correctly, the authors also have the ability to understand the roles, duties, insights and responsibilities of pharmacists and provide solutions on products in the industry pharmacy.
