Cool room, cold storage, dan frozen storage merupakan fasilitas yang digunakan pada cold chain management. Fasilitas tersebut didukung oleh ruangan dan alat yang harus memenuhi spesifikasi dari hasil kualifikasi. Kualifikasi alat atau mesin meliputi kualifikasi desain, kualifikasi instalasi, kualifikasi operasional, dan kualifikasi kinerja (CDOB, 2020). PT Enseval Putera Megatrading DC 1 memiliki freezer yang akan digunakan untuk mendukung cold chain management yang ditandai sebagai F.16. Freezer F.16 telah dilakukan kualifikasi desain dan kualifikasi instalasi yang selanjutnya akan dilakukan kualifikasi operasional pada kondisi kosong. Dari hasil kualifikasi operasional yang dilakukan, Freezer F.16 memenuhi spesifikasi kualifikasi operasional dengan pengaturan posisi pada beberapa titik yang akan dipastikan kembali pada kualifikasi kinerja.

"Penelitian ini bertujuan untuk mengkualifikasi kinerja Combi Cooler Freezer PF-20 F.16 guna memastikan bahwa freezer tersebut dapat mendistribusikan suhu dingin sesuai dengan spesifikasi operasional. Tujuan penelitian ini guna mengevaluasi kemampuan freezer dalam mematangkan material pendingin (ice gel) yang digunakan dalam pengiriman cold chain product. Penelitian dilakukan dengan menaruh thermometer data logger ke-12 titik pada freezer dan memperhatikan kematangan dari ice gel. Hasil penelitian menunjukkan rentang suhu distribusi dingin yang dihasilkan dari ke-12 posisi adalah -29°C hingga -4°C. Rentang suhu tersebut tidak memenuhi persyaratan yang ditetapkan (-30°C hingga -20°C) dan diperlukan justifikasi atas penyimpangan tersebut. Penyimpangan terjadi pada posisi 1 dan 2 yang berada pada tray pertama, serta posisi 3 dan 4 yang berada pada tray keempat. Namun, hasil pengujian distribusi pembekuan ice gel menunjukkan bahwa semua ice gel mengalami pembekuan dan tidak ada ice gel yang mencair. Penyimpangan keempat posisi tersebut dapat terjadi karena posisi tersebut paling dekat dengan bagian mesin freezer dan juga evaporator yang bekerja menyerap panas, sehingga memungkinkan terjadinya perpindahan kalor yang dapat meningkatkan suhu. Kesimpulan dari penelitian ini adalah pendistribusian suhu dingin pada Combi Cooler Freezer PF-20 F.16 telah memenuhi spesifikasi, dan penyimpangan suhu pada posisi 1, 2, 3, dan 4 dapat dikondisikan dengan justifikasi, serta freezer dapat digunakan untuk membuat ice gel dengan suhu -25°C dan tidak ada ice gel yang mencair.

This research aims to qualify the performance of the Combi Cooler Freezer PF-20 F.16 to ensure that the freezer can distribute cold temperatures in accordance with the operational specifications. The objective of this study is to evaluate the freezer's capability to mature the cooling material (ice gel) used in cold chain product shipments. The research was conducted by placing twelve data logger thermometers at various points within the freezer and monitoring the maturity of the ice gel. The results indicated that the temperature distribution range obtained from the twelve positions ranged from -29°C to -4°C. This temperature range did not meet the established requirements (-30°C to -20°C); therefore, justifications for the deviations were necessary. The deviations were observed in positions 1 and 2, located on the first tray, as well as positions 3 and 4, situated on the fourth tray. However, the ice gel freezing distribution test results showed that all ice gel samples were freezing without any melting. The deviations observed in the positions could be attributed to their proximity to the freezer's machinery and evaporator, which absorb heat and may cause heat transfer, leading to increased temperatures. In conclusion, the research confirmed that the distribution of cold temperatures in the Combi Cooler Freezer PF-20 F.16 met the specifications, and the temperature deviations in positions 1, 2, 3, and 4 could be justified. Additionally, the freezer can effectively produce ice gel at a temperature of - 25°C without any melting."

"Cold Chain Product (CCP) adalah produk yang sensitif terhadap temperatur sehingga penyimpanan dan pengirimannya memerlukan kontrol temperatur yang tak terputus mulai dari pabrik sampai distribusi ke pengguna akhir, hal ini harus dilakukan guna menghindari risiko penurunan khasiat dan keamanannya. Dibutuhkan freezer yang mumpuni dalam membekukan ice pack atau ice gel untuk diletakkan di dalam shipping box produk CCP. Oleh karena itu perlu dilakukan kualifikasi operasi untuk memastikan bahwa peralatan yang digunakan sesuai dengan kriteria keberterimaan dan dapat berfungsi dengan baik. Tujuan dari pembuatan laporan praktik kerja ini diantaranya, untuk mengetahui hasil kualifikasi operasi 1: menyalakan dan mematikan daya freezer combi cooler PF-20 F.15; Untuk mengetahui hasil kualifikasi operasi 2: pengaturan suhu freezer combi cooler PF-20 F.15; Untuk mengetahui hasil kualifikasi operasi 3: distribusi dingin pada freezer keadaan kosong freezer combi cooler PF-20 F.15; Untuk mengetahui hasil kualifikasi operasi 4: operasi defrost freezer combi cooler PF-20 F.15. Metodologi penelitian yang digunakan yaitu menggunakan rancangan observasional dengan memeriksa kualifikasi operasional distribusi dingin freezer dalam keadaan kosong. Didapatkan kesimpulan diantaranya, Hasil kualifikasi operasi 1, 2, dan 4 memenuhi persyaratan sedangkan hasil kualifikasi operasi 3 memenuhi persyaratan dengan adanya justifikasi pada posisi 1, 2, 3 dan 4.

Therefore, it is necessary to carry out operational qualifications to ensure that the equipment used meets the acceptance criteria and can function properly. The purpose of preparing this work practice report, among other things, is to find out the results of operation qualification 1: turning on and off the power of the combi cooler PF-20 F.15 freezer; To find out the results of operation qualification 2: setting the temperature of the combi cooler PF-20 F.15 freezer; To find out the results of operation qualification 3: cold distribution in an empty freezer freezer combi cooler PF-20 F.15; To find out the results of operation qualification 4: defrost freezer combi cooler PF-20 F.15 operation. The research methodology used was an observational design by examining the operational qualifications of the cold freezer distribution in an empty state. The conclusions obtained include the results of operational qualifications 1, 2 and 4 meet the requirements while the results of operational qualification 3 meet the requirements with justification in positions 1, 2, 3 and 4."

Salah satu aspek penting dalam pendistribusian produk CCP adalah pengontrolan suhu produk dalam penyimpanan selama proses distribusi terjaga dalam rentang yang dipersyaratkan. Untuk memenuhi persyaratan kontrol suhu produk CCP, PT Enseval Megatrading Tbk menggunakan metode passive packaging dengan menggunakan wadah penyimpanan yang suhunya terkontrol dan termonitoring selama pengiriman. Penelitian dilakukan dengan mengikuti prosedur pengujian kualifikasi kinerja freezer mengikuti protokal yang berlaku di PT Enseval Putera Mega Trading DC 1 Jakarta. Hasil pengujian kualifikasi kinerja freezer yang telah dilakukan, ditemukan beberapa data distribusi suhu dari termometer yang berada diluar rentang suhu yang dipersyaratkan yaitu -25°C s/d -15°C. Suhu yang didapatkan cenderung lebih dingin dari rentang suhu yang dipersyaratkan. Walaupun begitu pendistribusian suhu pada kinerja freezer terbilang sudah merata sehingga kualifikasi kinerja ini dinyatakan lulus. Selain itu, tidak ada ice gel yang mencair pada pengaturan suhu -18°C sehingga freezer tersebut dapat digunakan lebih lanjut untuk membuat material pendingin (ice gel) dengan pengaturan suhu -18°C. Hasil laporan ini dapat dijadikan sebagai sumber informasi dalam melakukan proses Kualifikasi Kinerja (KK) untuk fasilitas pendingin seperti freezer oleh pedagang besar farmasi atau distributor farmasi.

The research was carried out by following freezer performance qualification testing procedures following the applicable protocols at PT Enseval Putera Mega Trading DC 1 Jakarta. As a result of the freezer performance qualification tests that had been carried out, it was found that several temperature distribution data from the thermometer were outside the required temperature range, namely -25°C to -15°C. The temperature obtained tends to be colder than the required temperature range. However, the temperature distribution in the freezer performance is fairly even so this performance qualification is declared passed. Apart from that, no ice gel melts at a temperature setting of -18°C so that the freezer can be further used to make cooling material (ice gel) with a temperature setting of -18°C. The results of this report can be used as a source of information in carrying out the Performance Qualification (PQ) process for cooling facilities such as freezers by pharmaceutical wholesalers or pharmaceutical distributors."

Dalam pendistribusian produk rantai dingin PBF Enseval menggunakan box Styrofoam dan, polyurethane box, yang dilengkapi dengan satu data logger dan ice pack yang diletakan di sekeliling bagian dalam dari masing masing wadah obat yang digunakan. Ice pack di atas dibuat dengan menggunakan alat Combi Cooler Freezer PF-20, sehingga Freezer yang digunakan harus dapat menjamin kualitas ice pack yang dihasilkanya. Agar menjamin kinerja freezer berjalan sesuai spesifikasi yang diinginkan, maka perlu dilakukan kualifikasi kinerja secara berkala. kualifikasi kinerja merupakan prosess yang dilakukan untuk mendapatkan dan mendokumentasikan sistem dan atau alat bekerja sesuai dengan metode dan spesifikasi yang diinginkan dan telah disetujui. Kualifikasi kinerja dilakukan dengan cara meletakan thermometer data logger yang telah dibungkus plastik ke dalam freezer di beberapa lokasi selama 3 x 24 jam. Berdasarkan hasil kualifikasi kinerja diketahui bahwa pendistribusian dingin pada combi cooler Freezer merk Gea model PF-20 F.15 tidak merata dan terjadi Penyimpangan suhu di beberapa lokasi pada freezer. Tindak lanjut yang dapat dilakukan yaitu pembuatan ice pack dilakukan pada posisi yang memenuhi kriteria kualifikasi kinerja pada freezer.

In this report, the compilation of operational and performance qualification protocols for the stand freezer at PT Enseval Putera Megatrading Tbk Jakarta-1 Branch was conducted. The results indicate that the components of the operational and performance qualification protocols include objectives, scope, responsibilities, machine descriptions, and testing procedures for the stand freezer. Operational qualification testing includes powering on/off, temperature adjustment, cold distribution in empty condition, and defrost operation. Furthermore, performance qualification testing involves cold distribution testing with load on the freezer."

Penelitian dilaksanakan pada tanggal 11 September sampai dengan 22 September 2023. Hasil dari Kualifikasi Instalasi dan Kualifikasi Opeasional yang dilakukan pada Upright Medical Freezer DC 1 Jakarta telah memenuhi kriteria penerimaan tanpa ada penyimpangan dengan nilai rata-rata suhu yaitu -18,91 pada setting suhu -22°C. Titik pemantauan suhu akan diletakkan pada posisi 5 karena memiliki suhu maksimal dan rata-rata paling rendah yaitu -13.20°C dan -17.23°C.

The results of the Installation Qualification and Operational Qualification carried out on the Upright Medical Freezer DC 1 Jakarta met the acceptance criteria without any deviation with an average temperature value of -18.91 at the temperature setting - 22°C. The temperature monitoring point will be placed in position 5 because it has the lowest maximum and average temperatures, namely -13.20°C and -17.23°C."

Pemetaan suhu dilakukan saat container chiller belum terisi oleh produk rantai dingin yang akan disimpan. Hasil pemetaan suhu menunjukkan bahwa kondisi suhu pada container chiller yang diuji memenuhi kriteria yang dibutuhkan dalam penyimpanan produk rantai dingin karena masih berada pada rentang yang dipersyaratkan yaitu 2-8oC, dengan nilai MKT (Mean Kinetic Temperature) tertinggi dan terendah secara berturut-turut adalah 5,14 (Upper) dan 4,37 (Lower). Suhu tertinggi terdapat pada titik 8, sedangkan suhu terendah berada pada titik 11. Hasil kualifikasi operasional menunjukkan bahwa sistem refrigerasi container chiller dapat berfungsi dengan baik. Alarm pada container chiller juga dapat beroperasi dengan baik.

Cold Chain Products (CCP) must always be maintained within the required temperature range to avoid product damage. Therefore, adequate storage facilities are needed. Container chiller is one of the storage facilities for cold chain products used at PT. Enseval Putera Megatrading. According to CDOB (2020), chiller / cold room / freezer storage facilities must be qualified first at the start of use. Apart from that, to ensure that the container chiller can operate at a temperature that meets the requirements, temperature mapping activities need to be carried out. This paper discusses operational qualifications and temperature mapping of container chillers at PT. Enseval Putera Megatrading. Temperature mapping is carried out when the chiller container is not yet filled with the cold chain products to be stored. The temperature mapping results show that the temperature conditions in the chiller container tested meet the criteria required for storing cold chain products because they are still within the required range, namely 2-8oC, with the highest and lowest MKT (Mean Kinetic Temperature) values respectively being 5,14 (Upper) and 4,37 (Lower). The operational qualification results show that the container chiller refrigeration system can function well. The alarm on the container chiller can also operate properly."
