"Kementerian Kelautan dan Perikanan Indonesia menetapkan Lutjanus boutton sebagai salah satu spesies kakap yang diprioritaskan dalam manajemen perikanan Indonesia pada tahun 2021. Akan tetapi, L. boutton tidak termasuk ke dalam daftar Lutjanus sp. yang diperjualbelikan di pasar ikan Indonesia, melainkan hanya L. rufolineatus. Penelitian bertujuan untuk mengonfirmasi keberadaan kedua spesies berdasarkan jarak genetik dan potensi hibridisasi. Total 43 sampel telah melalui tahap ekstraksi DNA, PCR, elektroforesis, dan sekuensing. Analisis dilakukan dengan identifikasi, peta haplotipe, rekonstruksi pohon filogenetik, analisis sekuens, dan analisis morfometrik. Sebanyak 90,7% sekuens COI memiliki kemiripan hingga 100% dengan L rufolineatus, didukung oleh pembentukan klaster pada haplotipe, pohon filogenetik, dan analisis morfometrik PCA (principal component analysis). Analisis sekuens RAG1 menemukan ada lima situs polimorfik Y (C/T). Basa polimorfik Y di situs ke-5 yang ditemukan pada empat individu dari populasi Maluku Utara berpotensi sebagai indikasi adanya hibridisasi. Hal tersebut didukung oleh keberadaan empat dan dua individu Maluku Utara yang secara berturut-turut memiliki basa C dan basa T di situs ke-5. Meski demikian, empat individu Maluku Utara dengan potensi hibrid tidak dapat dikonfirmasi tanpa memvalidasi keberadaan L. boutton dengan galur murni sebagai parental dari hibrid generasi pertama.

In 2021, the Ministry of Marine and Fisheries of Indonesia considered Lutjanus boutton one of the prioritized species in Indonesia's fishery management. However, L. boutton was not included in the list of Lutjanus sp. traded in Indonesia's fish markets, but instead, L. rufolineatus. Therefore, this research aims to confirm the occurrence of both species based on genetic distance and their hybridization potential. A total of 43 samples have proceeded through DNA extraction, PCR, electrophoresis, and sequencing. The analysis includes moleculr identification, haplotype networks, phylogenetic tree reconstruction, sequence analysis, and morphometric analysis. Identification result shows that 90,7% of COI sequences have high similarity up to 100% with L. rufolineatus, supported by clustering group type in haplotype network, phylogenetic tree, and PCA. Analysis of samples RAG1 sequence shows a total of five polymorphic sites of Y (C/T) base observed. The polymorphic base Y at the 5th site that was found in four individuals from the North Maluku population has the potential to be an indication of hybridization. This is supported by the presence of four and two North Maluku individuals who respectively have base C and base T at the 5th site. However, four individuals from North Maluku with hybrid potential could not be further confirmed in this research without validating the pure breed of L. boutton as the parent of the first hybrid generation."

"Atternaria solani adalah jamur nekrotof penyebab tiga gelaja penyakit pada tanaman tomat. Pengelolaan pathogen ini dengan menggunakan varietas tahan memerlukan informasi keragaman genetic A. solani. Penelitian bertujuan untuk mengisolasi A. solani dari daerah produksi utama tomat dan kentang di Indonesia dan mempelajari keragaman keganasan dan genetiknya. Sebanyak 22 isolat A. solani diisolasi dari daun-daun tomat dan kentang bergejala hawar dini di Jawa Tengah dan Jawa Barat. A. alternate juga diperoleh dari daun tomat bergejala hawar dini di Jawa Barat dan Sumatra Utara, sehingga di Indonesia penyakit ini mungkin disebabkan oleh lebih dari satu spesies Alternaria. Uji ketahanan empat genotype tomat terhadap isolat . A. solani terpilih di rumah kaca menunjukan bahwa isolate local lebih agresif dalam menyebabkan penyakit hawar dini dan lesion batang dibandingkan dengan isolate asal Amerika Serikat. Implikasinya adalah galur galur tomat introduksi harus diuji terhadap isolate local untuk mendapatkan gen ketahanan yang efektif. Pengelompokan genotif marka amplified fragment length polymorshipsm (AFLP) hasil amplikasi DNA menggunakan kombinasi primer EcoRI+AG dan Msel+C memisahkan 28 isolat A. solani asal Indonesia dan Taiwan dari isolate asl Amerika Serikat, yang sesuai dengan pemisah derajat keganasan antara isolate local da isolate Amerika Serikat. Tiga gerombol genotype marka AFLP yang tidak berkaitan dengan asal geografis isolate terbentuk di antara isolat Indonesia dan Taiwan. Keragaman genetic yang rendah antar isolate A. solani Indonesia menunjukan adanya struktur populasi klonal yang tersebar luas. Keberhasilan program pemulihan local ketahanan tomat terhadap A. solani memerlukan ketersediaan koleksi isolate local yang terkterisasi keganasan dan genetiknya dengan bai agar diperoleh gen ketahanan yang efektif. "

"Penyakit dengue masih merupakan masalah kesehatan penting di sebagian besar Negara tropis dan subtropis. Dalam dua dekade terakhir terjadi lonjakan drastis baik jumlah kasus maupun daerah endemik di samping peningkatan keparahan penyakit (DHF/DSS). Meski telah dipelajari secara intensif, belum dipahami benar bagaimana mekanisme infeksi dengue berkembang menjadi DHF/DSS. Sejauh ini diketahui baik faktor inang maupun virulensi virus terlibat dalam menentukan keparahan penyakit. Studi-studi terbaru melaporkan perbedaan struktural genom di antara virus dengue yang memberikan manifestasi klinis berbeda. Perbedaan ini kemungkinan berhubungan dengan patogenesis penyakit. Dengue disease are reemerging disease and major health concern in tropical and subtropical regions because of the increasing number of patients, expanding endemic areas and increased occurrence of severe clinical manifestation (DHF/DSS) in the last two decades. Despite extensive studies, it is not fully elucidated mechanism by which dengue infection progress to DHF/DSS. Information obtained so far indicates that both host-related factor and virus virulence are involved. Recent studies have shown several structural differences of dengue virus genome between those associated with DF only and those with the potential to cause DHF. "Modularization is one of the key strategies for increasing responsiveness to customers. In modular product architecture a wide variety of product configurations can be generated by altering a limited number of modules and components. Product modules are identified by grouping highly coupled components in the same module. A Design Structure Matrix (DSM) is a compact presentation of the interaction between the components. In this paper, Analytical Hierarchy Process (AHP) and Genetic Algorithm (GA)-based methodology is proposed for the clustering of highly coupled DSM components in modules. Multi-criteria DSMs are proposed, which are aggregated by using weights generated by AHP. A genetic algorithm is designed to change the order of components in DSM and to bring highly coupled interactions near the diagonal. An illustrative case study is also made to validate the proposed algorithm. Two large sized and two small size modules are identified by selecting high density clusters around the diagonal. The clustered DSM also shows independent components and loose coupling between the two modules."

"Ikan gupi (Poecilia reticulata) merupakan ikan hias air tawar yang telah menjadi komoditas ekspor utama di Indonesia. Persilangan antar strain ikan gupi sudah banyak dilakukan, namun masih sedikit ketersediaan informasi genetik intraspesies ikan gupi. Penelitian ini bertujuan untuk mengetahui keragaman genetik intraspesies ikan gupi dan hubungan kekerabatan antar populasi di wilayah Jakarta, Bogor, Depok, Tangerang, dan Bekasi menggunakan marka gen Cytochrome Oxidase Subunit 1 (CO1). Amplifikasi PCR dan sekuensing menggunakan marka gen CO1 (F): 5 TCAACCAACCACAAAGACATTGGCAC-3 (26 pb) dan CO1 (R): 5 TAGACTTCTGGGTGGCCAAAGAATCA-3 (26 pb). Analisis yang dilakukan terhadap sekuen DNA ikan gupi antara lain; homologi BLAST, jarak genetik, dan analisis filogenetik. Hasil analisis homologi berdasarkan BLAST menunjukkan persentase similaritas 98-99% terhadap mtDNA Poecilia reticulata dan Poecilia wingei. Hasil analisis didapatkan jarak terbesar dan kekerabatan terjauh adalah populasi Bekasi strain Albino Full Red dengan populasi Tangerang strain Cobra. Sekuens tersebut mungkin dapat digunakan untuk menghasilkan benih dengan keragaman tinggi.

Guppy fish (Poecilia reticulata) is fresh water ornamental fish which have been the top export commodity in Indonesia. A lot of guppy strains had been hybridized, yet there are still few information about intraspecies genetic variation. Research was conducted to study intraspecies genetic variation and phylogenetic relationship among guppy population in region Jakarta, Bogor, Depok, Tangerang and Bekasi using Cytochrome Oxidase Subunit 1 (CO1) as genetic marker. Amplification and sequencing were using CO1 (F): 5 TCAACCAACCACAAAGACATTGGCAC-3 (26 pb) and CO1 (R): 5 TAGACTTCTGGGTGGCCAAAGAATCA-3 (26 pb). Some analysis which had been done with guppy DNA were; BLAST homology, genetic distance, and phylogenetic analysis. BLAST homology resulted 98-99% similarity according to mtDNA Poecilia reticulata and Poecilia wingei. The biggest distance and farthest relationship belong to Albino Full Red strain in Bekasi with Cobra strain in Tangerang. Those sequences might be used to produce potential germ with high genetic variation."
