"ABSTRAK
Nisin has been extensively used as a safe food preservative: therefore, the occurrence of nisin resistance in various bacteria including nisin exposed Listeria monocytogenes has increased in recent years. This problem could be overcome by using nisin in combination with other antimicrobial agents resulting in synergistic effects. Citric acid is a safe food additive granted GRAS status by the Food and Drug Administration, USA. In the present investigation, the antibacterial activity of nisin and citric acid alone or in combinations against L. monocytogenes 10403S was determined, and their potential as food preservative in food model systems was evaluated. The nisin and citric acid showed minimum inhibitory concentration (MIC) at 250 and 4,000 mg/ml, respectively. Checkerboard microdilution method using both compounds showed synergistic effect at concentration of 62.5mg/ml and 1,000 mg/ml, respectively, with the fractional inhibitory concentration index (FICI) value of 0.5. The potent anti listeria effect of nisin in combination with citric acid on the growth of L. monocytogenes in pork ham (food model) was observed during six days of storage at 4oC. This might be exploited to inhibit foodborne bacteria and minimize the nisin resistant problem of L. monocytogenes in the food industry."

"Penelitian ini dilakukan untuk menguji daya antibakteri bahan irigasi nisin 10%, klorheksidin 2%, natrium hipoklorit 2,5% terhadap pertumbuhan biofilm E. faecalis secara in vitro. Bakteri E. faecalis ATCC 29212 dibiakkan pada media agar BHI kemudian diinkubasi 24 jam pada suhu 370C. Bakteri diinokulasi pada membran filter selulosa nitrat selama 72 jam agar terbentuk biofilm. Dilakukan uji kontak langsung antara bahan uji dan biofilm selama 10 menit. Dilakukan kuantifikasi DNA bakteri yang hidup melalui penambahan PMA pada Real time PCR. Hasilnya nisin 10% mempunyai daya antibakteri terhadap biofilm E.faecalis, namun tidak sebanding dengan klorheksidin 2% dan natrium hipoklorit 2,5%.

The aim of this study was to evaluate antibacterial efficacy of 10% nisin, 2% chlorhexidine, 2.5% sodium hypochlorite against E.faecalis biofilm in vitro. Petri dishes containing BHI agar were seeded with E. faecalis ATCC 29212, incubated overnight at 370C. Celullose nitrate filter membrane was inoculated with E.faecalis for 72 hours to grown a biofilm. Direct contact test was performed between the test solutions and biofilm for 10 minutes. DNA quantification was performed using Real time PCR with PMA additive to count the lived cell. It was concluded that 10% nisin possessed antibacterial effect against E.faecalis biofilm, but not comparable with 2% chlorhexidine and 2.5% sodium hypochlorite."

"Penelitian ini berhasil memodifikasi tepung tapioka dengan asam sitrat melalu reaksi ikat silang. Tapioka dan asam sitrat direaksikan dengan mengatur keadaan selama reaksi seperti konsentrasi asam sitrat, waktu reaksi, pH, dan temperature. Asam sitrat dipilih sebagai agen pemodifikasi karena aman untuk makanan dan bisa membentuk reaksi ikat silang dengan pati melalui gugus karboksilat yang akan berubah menjadi gugus anhidrida dengan menaikan suhu reaksi. Hasil karakterisasi dengan FT-IR menunjukan terbentuk peak pada 1720 cm-1 yang menunjukan terbentuknya pati sitrat. Karakterisasi dengan XRD menunjukan bahwa pati termodifikasi mempunyai kristalinitas yang lebih rendah. Berdasarkan hasil uji produk pati sitrat, sifat fungsional dari pati menurun seperti swelling power, kelarutan, daya serap terhadap air, viskositas, dan meningkatnya kejernihan pasta. Daya cerna dari pati sitrat juga menurun secara signifikan hingga 25% dibandingkan dengan pati murni. Produk ini bisa menjadi pangan fungsional seperti penderita diabetes atau kanker usus besar. Perubahan dari sifat fungsional pati juga diharapkan bisa menjadi acuan untuk industri dalam memenuhi kebutuhan pati termodifikasi.

This study was successful modifying tapioca starch with citric acid through crosslinking reaction. Tapioca and citric acid are reacted by setting the conditions during the reaction such as the concentration of citric acid, time reaction, pH, and temperature. Citric acid was chosen as a modifying agent because it is safe for food and can form a crosslinking reaction with starch through the carboxylic group that will be converted to anhydrous by raising the reaction temperature. The results characterization with FT-IR shows that there is a peak at 1720 cm-1 which is shows the formation of starch citrate by the presence of ester bonds on starch. Characterization with XRD shows that modified starch has a lower level of crystallinity. Based on several results of testing starch citrate products, the functional properties of starch also change such as decreased swelling power, solubility, water absorption, viscosity, and increased clarity of starch paste. Digestibility of starch citrate also decreased significantly by 25% compared to normal starch. This product is can be a functional food such as for people with diabetes or colon cancer. The changes in the functional properties of starch are also expected to be a reference for the industry fulfill the needs of modified starch.

"Pseudomonas aeruginosa merupakan salah satu patogen terpenting yang menyebabkan infeksi nosokomial. Isolat P. aeruginosa sudah banyak yang resisten terhadap carbapenem yang juga berkaitan dengan resistensi terhadap antibiotik lain. Adanya P. aeruginosa yang resisten multiobat menyebabkan kesulitan dalam memilih pengobatan yang tepat. Terapi kombinasi dapat menjadi salah satu alternatif untuk menanggulangi keterbatasan monoterapi. Tujuan penelitian ini untuk mengetahui adanya sinergisme pada beberapa kombinasi antibiotik yang digunakan pada penelitian ini terhadap bakteri P. aeruginosa resisten carbapenem. Spesimen klinis yang terdiri dari sputum, bilasan bronkoalveolar, apusan luka, dan urin diambill dari ruang perawatan intensif Rumah Sakit Cipto Mangunkusumo. Isolat bakteri yang berasal dari spesimen klinis kemudian diidentifikasi dan diuji kepekaan. Isolat yang memenuhi kriteria inklusi diuji dengan metode checkerboard mikrodilusi untuk mempelajari efek kombinasi antibiotik antara amikacin dan ceftazidime, ceftazidime dan ciprofloxacin, serta amikacin dan ciprofloxacin. Dari 187 spesimen yang berasal dari 140 pasien diperoleh 16 isolat P. aeruginosa resisten carbapenem. Dari 16 Isolat P. aeruginosa yang resisten carbapenem yang diperiksa dengan uji checkerboard mikrodilusi sebanyak 13 isolat. Sebanyak 3 isolat yang terpapar kombinasi ceftazidime dan amikacin menunjukkan sinergisme dan 1 isolat yang terpapar kombinasi ceftazidime dan ciprofloxacin. Pada semua isolat yang terpapar kombinasi amikacin dan ciprofloxacin memperlihatkan indifference. Tidak ada antagonisme ditemukan pada ketiga kombinasi antibiotik ini. Berdasarkan penelitian ini, kombinasi ceftazidime dan amikacin masih dapat dipertimbangkan pada pasien dengan infeksi P. aeruginosa resisten carbapenem.

Pseudomonas aeruginosa is one of the important nosocomial pathogens. Currently, many P. aeruginosa isolates are resistant to carbapenem and other antibiotics. The occurrence of multidrug resistance in P. aeruginosa, causes difficulties in choosing the appropriate treatment of infection by this bacteria. Combination therapy could be an alternative to overcome the limitations of monotherapy. The objective of this study is to observe the occurrence of synergistic effect in the antibiotic combinations that are used in this study in carbapenem resistant P. aeruginosa. Clinical specimens consisting of sputum, bronchoalveolar lavage, wound swab, blood and urine were obtained from the ICU of Cipto Mangunkusumo Hospital. Bacterial isolates from the clinical specimens were identified and examined for susceptibility pattern. Isolates that fulfill inclusion criteria was tested with checkerboard microdilution method to study the antibiotic combination effect between amikacin and ceftazidime, ceftazidime and ciprofloxacin, amikacin and ciprofloxacin. There From 187 specimens that were collected from 140 patients, 16 carbapenem resistant P. aeruginosa isolates were obtained. From 16 carbapenem resistant P. aeruginosa isolates, there were 13 isolates that tested with checkerboard microdilution method. The results showed synergistic effect in 3 isolates that were exposed to ceftazidime and amikacin combination, and in 1 of the isolates that were exposed to ceftazidime and ciprofloxacin combination. Indifference was observed in all isolates that were exposed to amikacin and ciprofloxacin combination. No antagonism was found among the three antibiotic combinations. Based on this study, ceftazidime and amikacin combination could be considered in patient with carbapenem resistant P. aeruginosa infection.

"Latar belakang: Karies pada gigi sulung antara usia 0-72 bulan dikenal sebagai Early Childhood Caries (ECC) dan merupakan salah satu penyakit dengan prevalensi tinggi yang disebabkan oleh biofilm. Pada dekade terakhir, jamur Candida albicans banyak ditemukan bersama-sama dengan Streptococcus mutans dalam biofilm yang diambil dari plak gigi anak dengan ECC. Jamur C. albicans dapat meningkatkan derajat keparahan ECC. Pemahaman mengenai hubungan C. albicans dan S. mutans, memberikan perspektif baru untuk terapi yang efektif dalam mengkontrol ECC. Salah satu usaha untuk mencegah dan mengurangi tingkat ECC pada anak adalah dengan menggunakan bahan antimikroba. Bawang putih (Allium sativum) merupakan bahan herbal yang memiliki kemampuan antibakteri dan antijamur. Tujuan: Menganalisis efektivitas ekstrak bawang putih terhadap viabilitas biofilm Candida albicans anak ECC. Metode Penelitian: Penelitian dilakukan secara in vitro dengan isolat klinis dari plak gigi anak ECC. Ekstrak bawang putih dalam konsentrasi 10%, 25%, 50%, dan 100% dengan kontrol positif berupa CHX 0,2%. Uji viabilitas biofilm dilakukan dengan MTT assay. Hasil: Data statistik dianalisis dengan uji One Way ANOVA. Terdapat  perbedaan yang signifikan secara statistik dari viabilitas biofilm ekstrak bawang putih dibandingkan dengan kontrol negatif. Kesimpulan: Penelitian ini menunjukkan bahwa ekstrak bawang putih efektif terhadap viabilitas biofilm Candida albicans.

Background: Dental caries on primary teeth in a child 72 of months age or younger is defined as Early Childhood Caries (ECC). ECC has a high prevalences and caused by biofilms. In the past decade, Candida albicans has been frequently detected together with S. mutans in oral biofilms collected from children with ECC. Candida albicans might enhance degree of ECC. Understanding of C. albicans and S. mutans relationship give a new perspective for effective therapy to control ECC. Antimicrobial agent can be used to prevent or as a therapy for ECC. Garlic (Allium sativum) is one of the traditional medicine that has antibacterial and antifungal effect. Purpose: To analyzed the effectivity of garlic extract against the viability of C.albicans biofilms in children with ECC. Method: Laboratorium research (in vitro), with plaque sample from children with ECC. Garlic extract in 10%, 25%, 50%, 100%, and CHX 0,2% as positive control. MTT assay were used to assess biofilms viability. Statistical data were analyzed with the One Way ANOVA test. Result: There was a statistically significant difference in the viability of C.albicans biofilms after garlic extract application. Conclusion: This study showed that garlic extract has a positive effect on the viability of C.albicans."

"Staphylococcus aureus merupakan bakteri yang bersifat anaerob fakultatif, membentuk pigmen kuning, umumnya tumbuh berpasangan maupun berkelompok. Bakteri ini seringkali ditemukan pada saluran pernapasan atas dan kulit. Infeksi S. aureus diasosiasikan dengan beberapa kondisi patologi, diantaranya seperti bisul, jerawat, pneumonia, meningitis, dan arthritits. S. aureus merupakan salah satu bakteri Gram positif yang banyak ditemukan di lingkungan medis yang berpotensi menimbulkan infeksi tak berbahaya maupun gejala lebih serius terhadap pasien rawat inap di rumah sakit. Sebagai upaya pencegahan terjadinya infeksi ringan maupun serius akibat bakteri S. aureus, maka dilakukan metode sterilisasi bakteri. Dalam studi kali ini dilakukan perbandingan efikasi menggunakan metode sterilisasi UV-C dan ozon terhadap Staphylococcus aureus secara in vitro. Bakteri dipaparkan ozon dan sinar UV-C dengan durasi 15, 30 dan 45 menit. Dilakukan pula uji lanjutan sterilisasi stetoskop. Selanjutnya dilakukan analisa Total Plate Count (TPC) untuk mengetahui tingkat kematian bakteri dan dilakukan uji statistik berupa ANOVA, Mann-Whitney, dan Kruskal-Wallis sehingga dapat ditentukan metode sterilisasi yang paling efektif terhadap S. aureus. Penelitian menunjukkan bahwa sterilisasi menggunakan sinar UV-C dan ozon paling efektif pada durasi paparan 30 menit.

Staphylococcus aureus is a facultatively anaerobic bacteria, producing yellow pigments, generally grown in pairs or groups. These bacteria are often found in the upper respiratory tract and skin. S. aureus infection is associated with several pathological conditions, including ulcers, acne, pneumonia, meningitis, and arthritis. S. aureus is one of the Gram-positive bacteria found in many medical environments that have the potential to cause harmless infections or more serious symptoms to hospitalization patients. As an effort to prevent the occurrence of mild and serious infections due to S. aureus bacteria, the method of sterilization of bacteria is carried out. In this study, efficacy comparisons were conducted using UV-C and ozone sterilization methods against Staphylococcus aureus bacteria. Bacteria are exposed to ozone and UV-C light with durations of 15, 30 and 45 minutes. Further tests of stethoscope sterilization are also carried out. Furthermore, Total Plate Count (TPC) analysis is performed to determine bacterial mortality rates and statistical tests in the form of ANOVA, Mann-Whitney, and Kruskal-Wallis can be determined so that the most effective sterilization methods against S. aureus can be determined. Research shows that sterilization using UV-C light and ozone is most effective at an exposure duration of 30 minutes."

"Pseudomonas aeruginosa merupakan salah satu jenis bakteri Gram negatif yang sering ditemukan pada peralatan medis yang terkontaminasi. P. aeruginosa menjadi penyebab beberapa infeksi nosokomial seperti penyakit otitis eksterna, dermatitis, folikulitis, selain itu dapat menyebabkan infeksi pada mata, luka bakar, saluran napas bagian bawah, saluran kemih, dan organ lainnya.  Pencegahan terhadap penyebaran P.  aeruginosa di lingkungan fasilitas kesehatan sangat dibutuhkan, salah satunya dengan memanfaatkan paparan ozon dan radiasi UV-C untuk mensterilkan alat kesehatan untuk mencegah penyebaran bakteri P. aeruginosa. Tujuan dari penelitian ini ialah untuk melihat efektivitas paparan ozon dan radiasi UV-C dengan variasi waktu 0, 15, 30, dan 45 menit terhadap bakteri P. aeruginosa. Penelitian lanjutan diberikan perlakuan terhadap alat medis yaitu thermo-gun dengan variasi waktu 0, 10, 15, dan 30 menit untuk melihat efektivitas dari metode sterilisasi yang digunakan. Selanjutnya dilakukan persentase pengurangan jumlah koloni dan uji statistik menggunakan ANOVA, Mann-Whitney, dan Kruskall-Wallis. Hasil penelitian menunjukan persentase pengurang koloni bakteri 100% setelah diberikan paparan UV-C dengan durasi 15 menit pada media padat. 96% koloni bakteri berkurang setelah diberikan paparan ozon dengan kapasitas 2000 mg/h selama 45 menit. Sedangkan pada uji lanjutan didapatkan pengurangan bakteri sebanyak 75% dan 74% pada paparan UV-C dan Ozon oleh karena itu kedua perlakuan tidak menunjukan perbedaan signifikan untuk mensterilisasi thermo-gun

Pseudomonas aeruginosa is a type of Gram-negative bacteria that is often found in contaminated medical equipment and is not cleaned properly. P. aeruginosa is the cause of several infections such as externa otitis disease, dermatitis, folliculitis, in addition it can cause infections of the eyes, burns, urinary tract, and other organs. Prevention against the spread of P. aeruginosa in the hospital environment is needed, the study using plasma and radiated UV to sterilize medical devices is considered to prevent the spread of P. aeruginosa bacteria. The purpose of this study was to determine the effectiveness of exposure to ozone and UV-C radiation with time variations of 0, 15, 30, and 45 minutes against P. aeruginosa bacteria. Further research was given treatment of medical devices, namely thermo-guns with time variations of 0, 10, 15, and 30 minutes to see the effectiveness of the sterilization method used. Furthermore, the percentage reduction in the number of colonies and statistical tests using ANOVA, Mann-Whitney, and Kruskall-Wallis were carried out. The results showed that the percentage of bacterial colony reduction was 100% after being exposed to UV-C with a duration of 15 minutes on solid media. 96% of bacterial colonies were reduced after being exposed to ozone with a capacity of 2000 mg/h for 45 minutes. Meanwhile, in the follow-up test, it was found that there was a 75% and 74% reduction in bacteria on exposure to UV-C and Ozone. Therefore, the two treatments did not show a significant difference to sterilize the thermo-gun."

"Tujuan penelitian ini untuk membandingkan efikasi kombinasi phormoterol/budesonid turbuhaler dengan kombinasi nebulisasi salbutamoi/ipratropium bromid. Pengobatan utama asma akut adalah inhalasi agonis fa kerja singkat. Pasien asma saat ini menggunakan 2 obat, untuk pelega dan pengontrol. Saat ini sudah ada inhaler berisi kombinasi agonis- kerja lama onset cepat dan konikosteroid. Kombinasi ini bisa digunakan untuk pengontrol dan pelega. Uji klinis acak terbuka dilakukan pada 76 pasien berusia anlara 12 - 60 tahun yang datang ke RS Persahabatan dengan skor asma 8 - 12. Pasien lersebut dibagi menjadi 2 kelompok masing-masing 38 pasien. Kelompok pertama mendapat kombinasi formoterol/budesonid 4,5/160 {lg turbuhaler (kelompok T), kelompok kedua mendapat kombinasi nebulisasi salbutamoi/ipratropium bromid 2,5/0,25 mg (kelompok N). Setiap kelompok mendapat inhalasi tiap 20 menit, total 3 kali pemberian. Tidak ada perbedaan bermakna padajenis kelamin, tinggi dan berat badan, APE awal dan skor asma awal antar kedua kelompok. Tampak peningkatan APE dan penurunan skor asma yang bermakna pada tiap kelompok, namun perubahan ini tidak berbeda bermakna antar kedua kelompok pada tiap interval waktu yang diamati. Efek samping, nyeri tenggorok, tremor, berdebar, terjadi pada kedua kelompok namun hanya ringan. Satu pasien pada kelompok T mengalami 3 efek samping sekaligus, 5 lainnya hanya tremor. Enam orang pada kelompok N hanya mengalami tremor. Kombinasi formoterol/budesonid turbuhaler dan kombinasi salbutamoi/ipratropium bromid nebulisasi secara klinis tidak berbeda bermakna dalam mengobati asma akut sedang, namun kombinasi salbutamoi/ipratropium bromid nebulisasi memiliki efek samping lebih ringan. The aim of this study was to compare efficacy combination of phormoterol/budesonide turbuhaler vs. salbutamoi/ipratropium bromide nebulization. Main therapy for acute asthma is inhaled short acting ^2-^Sonist. Asthma patients are using two drugs, controller and reliever. Recently there is device-containing combination of long-acting p2-ugonist with rapid onset and corticosteroid. This combination can act as reliever and controller. An opened randomized clinical trial of 76 patients between the ages of 12 and 60 years presenting to Persahabatan Hospital with asthma score between 8-12 participated in this study. After initial evaluation, patients were divided into two groups. Thirty-eight patients were administered combination offonnaterol/budesonide 4.5/160 {lg via turbuhaler (T-group) every 20 minutes, total of three doses, and another 38 of salbutamoi/ipratropium bromide 2.5/0.25 mg via nebulizer (N~group) also with the same manner. There were no statistical difference in sex, mean age, high, weight, initial PEFR, and asthma score between two groups. The significant increased of PEFR and decreased of asthma score were observed in both groups. However, there were no significant difference of PEFR and asthma score between the two groups within every time-interval. Adverse events were mild including hoarseness, tremor and palpitation. Of T-group, 1 subject was suffered from 3 adverse events simultaneously (hoarseness, tremor and palpitation), 5 subjects were only tremor. Of N-group, all 6 subjects were only suffered from tremor. A combination of formoterol/budesonide turbuhaler and a combination of nebulized salbutamoi/ipratropium bromide are clinically equivalent for treatment moderate acute asthma. However, nebulized salbutamoi/ipratropium bromide had less adverse effects. (Med J Indones 2006; 15:34-42)."
