"Senyawa turunan kuinazolinon mempunyai berbagai aktivitas biologis. Salah satunya adalah 6-amino-3-benzil-4(3H)-kuinazolinon, yang memiliki aktivitas antibakteri berspektrum luas. Dalam rangka memperoleh senyawa turunan 6- amino lainnya dari 4(3H)-kuinazolinon, telah dilakukan nitrasi pada senyawa 3- (4-klorofenil)-2-metil-4(3H)-kuinazolinon. Nitrasi dilakukan dengan berbagai metode nitrasi dengan tujuan memperoleh metode nitrasi terbaik untuk senyawa tersebut. Metode nitrasi tersebut antara lain : nitrasi dengan asam nitrat berasap dan asam sulfat pekat, nitrasi dengan asam nitrat pekat dan asam sulfat pekat, nitrasi dengan perak nitrat dan N-bromosuksinimid, nitrasi dengan serium amonium nitrat dan silika asam sulfat, dan nitrasi dengan serium amonium nitrat dan asam sulfat pekat. Hasil penelitian menunjukkan bahwa nitrasi dengan asam nitrat berasap dan asam sulfat pekat menghasilkan senyawa 3-(4’-kloro-3’-nitrofenil)-2-metil-6-nitro-4(3H)-kuinazolinon dengan rendemen sebesar 49,74%. Struktur molekul tersebut telah dikonfirmasi dengan spektrum FT-IR dan 1HNMR. Nitrasi juga berhasil dilakukan dengan menggunakan asam nitrat pekat dan asam sulfat pekat, akan tetapi produk yang dihasilkan jauh lebih sedikit dibandingkan menggunakan asam nitrat berasap dan asam sulfat pekat. Metode nitrasi yang lain tidak memberikan hasil yang diinginkan. Dari penelitian ini dapat disimpulkan bahwa metode nitrasi terbaik untuk 3-(4-klorofenil)-2-metil-4(3H)-kuinazolinon adalah dengan menggunakan asam nitrat berasap dan asam sulfat pekat.

Some of quinazolinone derivatives have various biological activities. One of them is 6-amino-3-benzyl-quinazolin-4-one reported to have broad-spectrum antibacterial activity. In order to obtain other 6-amino derivatives of quinazolinone, the experiment on the nitration of 3-(4-chlorophenyl)-2-methylquinazolin-4-one with various methods of nitration have been performed to get the best nitration methods for these compounds. Implemented nitration methods were: nitration with fuming nitric acid and concentrated sulfuric acid, nitration with concentrated nitric acid and concentrated sulfuric acid, nitration with silver (I) nitrate and N-bromosuccinimide, nitration with ceric ammonium nitrate and silica supported sulfuric acid, and nitration with ceric ammonium nitrate and concentrated sulfuric acid. The results showed that nitration with fuming nitric acid and concentrated sulfuric acid generating compound 3-(4'-chloro-3'-nitrophenyl)-2-methyl-6-nitro-quinazolin-4-one with yield 49.74%. The structure was confirmed using FT-IR and 1H-NMR. Nitration was also successfully performed using concentrated nitric acid and concentrated sulfuric acid, however the resulting product was less than using fuming nitric acid and concentrated sulfuric acid. The other nitration methods were not give the desired results. From these research, it could be concluded that the best nitration method for 3-(4-chlorophenyl)-2-methyl-quinazolin-4-one is using fuming nitric acid and concentrated sulfuric acid."
