"Metode fused deposition modelling (FDM) merupakan salah satu teknik pencetakan tiga dimensi (3D-printing) yang sangat berpotensi untuk diaplikasikan dalam pembuatan tablet. Metode ini diawali proses ekstrusi untuk menghasilkan filamen menggunakan polimer termoplastik sebagai bahan pembentuknya, namun pemanfaatan polimer farmasi dalam bidang ini masih perlu dikembangkan lebih lanjut. Penelitian ini bertujuan untuk mendapatkan filamen mengandung teofilin berbasis campuran polimer polivinil pirolidon – poli asam laktat (PVP-PLA) serta polikaprolakton (PCL) melalui proses ekstrusi dengan konsentrasi bahan aktif maksimal dan mengamati pengaruhnya terhadap kekuatan mekanis filamen serta pelepasan obat. Filamen yang dibuat menggunakan campuran PVP-PLA dipreparasi dengan alat twin-screw extruder, sementara filamen berbasis PCL dibuat menggunakan alat single-screw extruder. Seluruh filamen hasil ekstrusi dikarakterisasi meliputi bentuk dan diameter, morfologi permukaan (SEM), kekuatan mekanis, analisis kandungan obat dan uji pelepasan obat. Tiga filamen berbasis PVP berhasil dibuat dengan diameter rata-rata berkisar antara 1,51 ± 0,11 mm hingga 1,60 ± 0,10 mm, sementara filamen berbahan dasar PCL berhasil diekstrusi dalam 12 formulasi dengan rentang diameter antara 1,38 ± 0,07 mm hingga 1,56 ± 0,07 mm. Tampilan SEM menunjukkan filamen yang dihasilkan memiliki bentuk permukaan yang cukup halus dengan bahan aktif terlihat tersebar di dalam filamen. Seluruh filamen memiliki kekuatan mekanis yang memenuhi syarat dengan rentang kekerasan antara 0,611 ± 0,044 kg hingga 2,478 ± 0,135 kg dan diamati terjadi penurunan kekuatan mekanis seiring dengan meningkatnya konsentrasi teofilin dalam filamen. Kapasitas pemuatan teofilin maksimal dalam filamen sebesar 18,25 ± 0,29% (b/b) untuk formula berbasis PVP dan 5,09 ± 0,07% (b/b) untuk formula berbasis PCL. Hasil uji pelepasan obat menunjukkan bahwa filamen dengan bahan dasar PVP memiliki karakteristik pelepasan segera, sementara filamen berbasis PCL memiliki karakteristik pelepasan diperlambat. Secara keseluruhan, filamen yang dihasilkan memiliki kualitas yang baik dan berpotensi untuk diaplikasikan dalam pembuatan sediaan farmasi dengan bantuan teknologi 3D-printing.

Fused deposition modelling (FDM) is one of the 3D-printing techniques which is potential to be applied in tablet manufacturing. This method started with the hot melt extrusion process which requires thermoplastic polymer as the filament-forming material, but the use of pharmaceutical polymers still has to be developed. This study aims to obtain theophylline-loaded filaments using the combination of polyvinyl pyrrolidone and polylactic acid (PVP-PLA) as well as polycaprolactone (PCL) with maximum drug-loading capacity through extrusion process and observing their impact to the mechanical properties and drug release profile. Filaments made of the PVP-PLA mixture were prepared using twin-screw extruder, while the PCL-based filaments were fabricated using the single-screw extruder. All extruded filaments were characterized for shape and diameter, surface morphology (SEM), mechanical properties, drug content, and drug release profile. Three PVP-based filaments were successfully prepared with average diameter ranging in 1.51 ± 0.11 mm to 1.60 ± 0.10 mm and 12 PCL-based filament formulations were successfully made with the range of diameter between 1.38 ± 0.07 mm and 1.56 ± 0.07 mm. SEM images showed that all filaments have fairly smooth surface and the active ingredients were spread evenly inside the filaments. All filaments have proper mechanical properties with the hardness value ranging in 0.611 ± 0.044 kg to 2.478 ± 0.135 kg and it was observed that the strength was decreased as the amount of theophylline in filaments increased. Maximum drug-loading capacity for the PVP-based filaments was 18.25 ± 0.29% (w/w) and 5.09 ± 0.07% (w/w) for the PCL-based filaments. The results of drug release study showed that filaments using PVP have the immediate-release profile, while the PCL-based filaments have slow release profiles. Overall, the extruded filaments have excellent properties and potential to be applied in the manufacturing of pharmaceutical preparations using the 3D-printing technology."

"This book presents the comprehensive results of experimental and numerical investigations of glass façade breakage behavior under fire conditions. First of all, full-scale frame and point-supported glass facades, incorporating single, double and coated glazing, were tested under pool fire conductions. The results determined the effects of different glass frames, types of glass, and thermal shocks on breakage behavior. Small-scale tests, using the Material Testing System (MTS) 810, Netzsch Dilatometer and FE-SEM, were also performed at different temperatures to determine the basic mechanical properties of glazing.
In addition, a three-dimensional dynamic model was developed to predict stress distribution, crack initiation and propagation, and has since been employed to identify the breakage mechanisms of different types of glass facade. The numerical results showed very good agreement with the experimental results and verified the models ability to accurately predict breakage. Lastly, a theoretical model based on incident heat flux was developed to predict the breakage time and heat transfer in glazing, which served to reveal the nature of interactions between fire and glass."
