"Radikal bebas adalah molekul atau senyawa kimia sangat reaktif yang menyebabkan stres oksidatif, yang didefinisikan sebagai ketidakseimbangan antara jumlah oksidan dan antioksidan, berpotensi memicu kanker. Senyawa alkaloid dari Phoebe sp. diketahui sebagai antioksidan yang potensial dan sebagai agen sitoprotektif. Oleh karena itu, penelitian kali ini hendak dilakukan isolasi dan uji aktivitas antioksidan terhadap ekstrak daun Phoebe declinata Nees. Daun Phoebe declinata Nees dimaserasi dengan n-heksana, kemudian diekstraksi kembali dengan metode refluks menggunakan diklorometana. Ekstrak diklorometana diisolasi dengan kromatografi kolom, fraksi yang dihasilkan dimurnikan dengan metode rekristalisasi dan KLT preparatif. Isolat diidentifikasi dengan penyemprot Dragendorff dan diuji aktivitas antioksidan dengan metode DPPH. Dari penelitian ini diperoleh 26,9 mg kristal yang positif terhadap reaksi alkaloid. Nilai IC50 dari kristal tersebut adalah 147,62 μg/ml.

Free radicals are highly reactive molecules or chemical substances that cause oxidative stress, which is defined as an imbalance between oxidants and antioxidants, potentially leading to cancer. Alkaloids from Phoebe sp. is known as a potential antioxidant and cyto-protective agent. Therefore, at this research the leaves extract of Phoebe declinata Nees was isolated and tested its antioxidant activity. Leaves of Phoebe declinata Nees were macerated by n-hexane and then re-extracted by reflux method using dichloromethane. Dichloromethane extract was isolated by column chromatography and then its fraction was purified by recrystallization and TLC-preparative method. The isolate was identified by Dragendorff sprayer and its antioxidant activity was tested by DPPH method. From this research, 26.9 mg crystal was obtained and it was positive of alkaloid reaction. IC50 of this crystal was 147.62 μg/ml."
