"ABSTRAKTesis ini membahas pesan ideologis yang disampaikan oleh Presiden Soeharto di awal kepemimpinannya. Presiden Soeharto adalah Presiden RI ke-2 yang memimpin Indonesia selama 32 tahun. Wacana pidato ini disampaikan di Manado pada bulan Oktober 1967. Presiden Soeharto melakukan kunjungan ke Manado, Sulawesi Utara, sebagai upaya untuk menyampaikan berbagai usaha yang sedang dan akan dilakukan oleh pemerintahan baru. Di awal kepemimpinannya, pidato merupakan salah satu cara berkomunikasi Presiden Soeharto kepada rakyat Indonesia yang bertujuan menyampaikan nilai ideologi dan cita-cita Orde Baru. Penelitian ini menganalisis berbagai pesan ideologis yang termuat dalam transkripsi wacana pidato Presiden Soeharto dengan menggunakan analisis wacana kritis Norman Fairclough. Pengonstruksian frasa, klausa, dan kalimat merupakan strategi Presiden Soeharto dalam menyampaikan pesan ideologis, baik secara tersurat, maupun secara tersirat. Hasil penelitian menunjukkan bahwa strategi yang dibangun oleh Presiden Soeharto dalam pidatonya memperlihatkan hubungan kuasa antara dirinya dan rakyat Sulawesi Utara.

ABSTRACTThe thesis analyses the ideological messages delivered by President Soeharto at the beginning of his presidency. Soeharto was the second president ruling Indonesia for 32 years. The speech was delivered in Manado in October 1967. President Soeharto visited Manado, North Sulawesi, as an effort to tell about the things that had been and would be carried out by the new government. At the early period of his presidency, one of the ways used by President Soeharto to communicate to Indonesian people was delivering speech. This way was aimed at conveying the ideological values and ideals of the New Order. This research analyses various ideological messages contained in the transcript of the speech discourse of President Soeharto by using critical discourse analysis of Norman Fairclough. The construction of the phrases, clauses, and sentences was the strategy used by President Soeharto in conveying the ideological messages, either implicitly or explicitly. The result of the research indicates that the strategy used by President Soeharto in his speech showed the power relations between him and the people of North Sulawesi."

"3,4-Metilendioksimetamfetamin MDMA atau di Indonesia dikenal dengan ekstasi merupakan recreational drugs golongan stimulant dan halusinasi dari golongan amfetamin yang masih banyak disalahgunakan hingga saat ini. Untuk membuktikan seseorang menyalahgunakan MDMA maka diperlukan uji MDMA dalam tubuh. Selama ini kadar MDMA di dalam tubuh biasanya ditentukan di dalam darah dan urin. Dried Blood Spot sebagai metode yang lebih sederhana dan tidak invasif bila dibandingkan dengan pengambilan darah langsung dari vena yang lebih invasif atau dari urin yang masih diragukan kebenaran pengambilan sampel dikarenakan masalah privasi. Penelitian ini bertujuan untuk mengembangkan metode analisis MDMA dalam sampel DBS mulai dari kondisi kromatografi gas spektrometri massa yang optimum, metode preparasi sampel DBS yang optimum, hingga validasi metode analisis. Kondisi kromatografi optimum adalah kolom kapiler HP-5 MS dengan panjang 30 m, diameter dalam 0,25 mm; fase gerak gas Helium 99,999 ; laju alir 1,2 mL/menit; deteksi MS pada nilai m/z 58,00 dan 135,00 dan efedrin HCl sebagai baku dalam. Preparasi sampel menggunakan metode mikroekstraksi cair-cair dengan pelarut methanol lalu reesidunya dikeringkan dan direkonstitusi dengan etil asetat sebanyak 40 ? L. hasil validasi terhadap metode analisis MDMA yang dilakukan memenuhi persyaratan validasi berdasarkan EMEA Bioanalytical Guideline tahun 2011. Metode yang diperoleh linear pada rentang konsentrasi 25,0-500,0 ng/mL dengan r> 0,9999.

3,4 Metilendioxymethamphetamine or in Indonesia known as ecstasy is one of recreational drugs that causing stimulant and hallucinogen of the amphetamine group which is still widely abused. To prove a person abusing MDMA then MDMA test is required in the body. MDMA concentration in the body are usually determined in the blood and urine. Dried Blood Spot as a simple and non invasive method compared to blood taking directly from the vene that usually is invasive and urine that can be still doubtful because privacy issue. This study aims to develop analytical methods for MDMA in DBS sample from the conditions of gas chromatography mass spectrometry optimum, optimum DBS preparation methods, to the validation of analytical methods. The optimum chromatography conditions were HP MS 5 capilarry columns with a length of 30 m 0.25 mm inner diameter mobile phase Helium gas 99.999 flow rate 1.2 mL min detection of MS at m z values of 58.00 and 91.00 and ephedrine HCl as an internal standard. Sample preparation using liquid liquid microextraction with methanol solvent and the residue is dried and reconstituted with about 40 L of ethil acetate. The results of the validation of analytical methods for MDMA that satisfies the validation by the EMEA Guideline 2011. Bioanalytical Methods obtained linear in the concentration range from 25.0 to 500.0 ng mL with r 0.9999."
