"Latar belakang : Nigella sativa dilaporkan memiliki efek antijamur terhadap Candida albicans. Candida albicans adalah jamur yang menyebabkan kandidiasis oral.
Tujuan : Menganalisis efektivitas antijamur ekstrak biji Nigella sativa terhadap viabilitas Candida albicans.
Metode : Candida albicans ditambahkan pada 96-microwell plate yang telah dilapisi saliva buatan, kemudian dipaparkan ekstrak biji Nigella sativa dengan konsentrasi 6.25%-50% dan amphotericin B 1µl (250mg/ml) sebagai kontrol positif. Viabilitas Candida albicans dihitung dengan uji MTT.
Hasil : Nilai optical density Candida albicans lebih rendah setelah pemberian ekstrak biji Nigella sativa dibandingkan dengan kelompok tanpa perlakuan.
Kesimpulan : Viabilitas Candida albicans setelah dipaparkan ekstrak biji Nigella sativa turun dibandingkan dengan kelompok tanpa perlakuan dan viabilitas Candida albicans menurun seiring dengan peningkatan konsentrasi ekstrak biji Nigella sativa.

Back ground : Nigella sativa has antifungal effect against Candida albicans. Candida albicans is a fungi that causes oral candidiasis.
Objective : To analyze the antifungal effectiveness of the extract of Nigella sativa seed on the viability of Candida albicans.
Methods : Candida albicans was added on 96-well plate that had lined been by artificial saliva and exposed by the extract of Nigella sativa seed 6.25%-50% and amphotericin B 1µl (250mg/µl) was used as positive control. The viability of Candida albicans was determined by MTT assay.
Result : The optical density value of Candida albicans after exposed by the extract of Nigella sativa seed was lower than the negative control.
Conclusion : The viability of Candida albicans after exposed by the extract of Nigella sativa seed was decreased than the negative control and viability of Candida albicans was decreased as increasing concentrations of the extract of Nigella sativa seed.

"Candida glabrata adalah spesies kedua yang terisolasi pada kandidiasis oral setelah Candida albicans. Nigella sativa (jintan hitam) dilaporkan memiliki efek antijamur. Tujuan penelitian ini adalah menganalisis efek antijamur ekstrak biji Nigella sativa terhadap viabilitas Candida glabrata. Candida glabrata ditambahkan ke dalam 96 microwell plate yang telah dilapisi saliva buatan kemudian dipajankan dengan ekstrak biji Nigella sativa 6.25%-50% dan amphotericin B 250 mg/ml. Dilakukan pengujian viabilitas Candida glabrata dengan uji MTT. Viabilitas Candida glabrata lebih rendah setelah pamajanan dengan ekstrak biji Nigella sativa dibandingkan kontrol negatif. Viabilitas Candida glabrata semakin menurun seiring dengan peningkatan konsentrasi ekstrak biji Nigella sativa.

Candida glabrata is the second most frequently isolated species from Oral Candidiasis, after Candida albicans. Nigella sativa (black cumin) is reported has antifungal effect. This research was to analyze Nigella sativa antifungal effect Seeds’ extract to the Candida glabrata viability. "[ABSTRAK
Penyakit diare masih merupakan masalah serius di Indonesia, terutama pada anak-anak. Salah satu patogen tersering penyebab diare adalah Escherichia coli, terutama ETEC. Penggunaan Nigella sativa untuk berbagai penyakit sudah diteliti, namun hasil penelitian terhadap Escherichia coli belum konklusif. Oleh karena itu dilakukan percobaan untuk mengetahui potensi antibakteri Nigella sativa Linn. terhadap Escherichia coli. Penelitia dilakukan di Laboratorium Mikrobiologi FKUI secara in vitro, menggunakan metode sumuran. Konsentrasi ekstrak N. sativa yang digunakan ialah 1000 mg/ml, 500 mg/ml, 250 mg/ml, 125 mg/ml, dan 62,5 mg/ml, dengan siprofloksasin sebagai kontrol positif dan akuades sebagai kontrol negatif. Percobaan dilakukan tiga kali dengan pengulangan masing-masing empat kali. Pada penelitian ini tidak ditemukan adanya efek antibakteri Nigella sativa Linn. terhadap Escherichia coli. Hal ini dapat disebabkan oleh karakteristik Nigella sativa di Indonesia

"Latar belakang: Propolis memiliki efek antifungal. Universitas Indonesia sedang mengembangkan permen mengandung propolis yang diduga dapat menghambat pertumbuhan C.albicans.
Tujuan: Penelitian ini bertujuan untuk mengetahui efek ekstrak propolis dan permen propolis terhadap pertumbuhan C.albicans.
Metode: Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM) ekstrak propolis diuji menggunakan metode spektrofotometri serta jumlah koloni C.albicans pasca pemaparan ekstrak dan permen propolis dihitung secara langsung.
Hasil: Nilai KHM dicapai pada konsentrasi 10%, nilai KBM dicapai pada konsentrasi 15%; serta terdapat penurunan jumlah koloni jamur pasca pemaparan ekstrak dan permen propolis.
Kesimpulan: Ekstrak dan permen propolis terbukti efektif menghambat pertumbuhan C.albicans.

Background: Propolis has antifungal effect. Universitas Indonesia has been developing propolis candy which can inhibit C.albicans growth.
Objective: This research was aimed to analyze effect of propolis extract and candies to C.albicans growth.
Methods: After C.albicans were exposed to propolis extract and candies, Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) were determined by spectrophotometry and post-exposure colonies of C.albicans were counted.
Result: MIC of propolis extract against C.albicans were determined at 10% and MBC at 15%, also the amount of C.albicans colonies were decreased after propolis extract and candies exposure.
Conclusion : Propolis extract and candies were effective to inhibit C.albicans growth.

"Biofilm C. albicans memiliki matriks ekstraseluler yang mempersulit penetrasi agen antifungal sintetik. Matriks ini diproduksi pada fase filamentasi dan terakumulasi pada fase maturasi. Temulawak merupakan obat herbal yang banyak digunakan di Indonesia dan ekstraknya telah dilaporkan memiliki efek antifungal terhadap C. albicans planktonik karena memiliki senyawa aktif yaitu xanthorrhizol. Penelitian ini dilakukan dengan MTT assay untuk menghitung viabilitas biofilm C. albicans setelah pemaparan dengan ekstrak etanol temulawak secara in vitro. Hasil yang didapatkan menunjukkan ekstrak etanol temulawak memiliki efek antifungal yang setara dengan nystatin terhadap biofilm C. albicans fase filamentasi dan maturasi pada konsentrasi 35%.

Extracellular matrix in C. albicans biofilm preventing access of synthetic antifungal agents to C. albicans biofilm. This matrix is produced during filamentation phase and accumulated on maturation phase. Java turmeric is a common Indonesian herbal medicine and has been reported to have antifungal effect against planktonic C. albicans for its active component, xanthorrhizol. This research was conducted using MTT assay to count C. albicans biofilm viability after in vitro exposure to Java turmeric ethanol extract. The result showed it has an equal antifungal effect to nystatin against C.albicans biofilm on filamentation and maturation phase in 35% of concentration.

"Salah satu faktor virulensi Candida albicans adalah kemampuannya dalam membentuk biofilm sehingga meningkatkan resistensi terhadap agen antifungal. Fase awal merupakan prasyarat terbentuknya biofilm serta ditandai dengan adhesi dan proliferasi sel C. albicans. Temulawak merupakan tanaman khas Indonesia dan dilaporkan memiliki efek antifungal karena mengandung zat aktif yaitu xanthorrhizol. Penelitian ini dilakukan dengan MTT assay untuk mengukur viabilitas C. albicans pada biofilm setelah pemaparan ekstrak etanol temulawak secara in vitro.
Hasil penelitian menunjukkan ekstrak dengan konsentrasi 35% dapat menurunkan viabilitas C. albicans setara dengan nystatin. Ekstrak etanol temulawak terbukti memiliki efek antifungal terhadap C. albicans pada biofilm fase adhesi dan proliferasi.

Candida albicans has the ability to form biofilm that increase resistance to antifungal agents. Early phase is a prerequisite, characterized by adhesion and proliferation. Java turmeric is an Indonesian medicinal plants and reported to have antifungal effect due to its active component, xanthorrhizol. This study was conducted using MTT assay to measure viability of C. albicans in biofilm after exposure to Java Turmeric ethanol extract.
The result showed extract in 35% concentration can reduce the viability of C. albicans equal to nystatin’s capability. Java Turmeric ethanol extract has antifungal effect against C. albicans in adhesion and proliferation phase of biofilm.

"Faktor serum yang bersifat antimikroba seperti komplemen dapat menghambat pertumbuhan C. albicans. Konsumsi xylitol dilaporkan mampu menekan pertumbuhan C. albicans.
Tujuan: Menganalisis efek xylitol 1%, 5%, 10% selama 3 hari atau 7 hari terhadap resistensi C. albicans dalam serum in vitro, dan menganalisis peran faktor serum dalam menghambat C. albicans dalam serum.
Metode: Deteksi C. albicans yang diambil dari lesi mulut pasien kandidiasis oral dilakukan dengan menggunakan media CHROMagar dan dikonfirmasi dengan uji pembentukan germ tube. Setelah melalui tahap pengenceran, C. albicans dipaparkan dengan larutan xylitol 0% (kontrol), 1%, 5%, dan 10% yang dilarutkan dalam media Sabouraud Dextrose Broth (SDB) selama 3 hari atau 7 hari. Tiap konsentrasi dan durasi kemudian dipaparkan dalam serum aktif (Fetal Bovine Serum/FBS) atau serum inaktif (FBS yang sudah dipanaskan pada suhu 65°C selama 30 menit untuk inaktivasi komplemen) pada suhu 37°C selama 2 jam. Jumlah koloni C. albicans pada Sabouraud Dextrose Agar (SDA) dihitung 2 hari kemudian. Prosedur yang sama dilakukan pada C. albicans strain ATCC 10231. Analisis data menggunakan uji one-way ANOVA dengan 0,05.
Hasil: Pada kultur C. albicans 3 hari, jumlah koloni dalam serum aktif secara bermakna lebih rendah daripada dalam serum inaktif, baik dengan maupun tanpa paparan xylitol (p = 0.032). Peningkatan konsentrasi xylitol meningkatkan jumlah koloni C. albicans klinis dalam serum aktif, walaupun secara statistik tidak bermakna (p = 0.689). Hanya paparan xylitol 10% selama 7 hari yang meningkatkan jumlah koloni C. albicans secara bermakna (p = 0.034). Faktor serum tidak mempengaruhi jumlah koloni C. albicans usia 7 hari. (p = 0.404).
Simpulan: Pemberian xylitol 1%, 5%, dan 10% selama 3 dan 7 hari tidak mempengaruhi efek inhibisi C. albicans oleh faktor serum. Efek inhibisi C. albicans oleh faktor serum hanya bermakna pada kultur usia 3 hari dan tidak terlihat pada kultur usia 7 hari.

Serum factor with antimicrobial effect like complement, could inhibit C. albicans growth. Xylitol is reported to inhibit the growth of C. albicans.
Objectives: Investigating the effect of 1%, 5%, 10% xylitol for 3 or 7 days on C. albicans resistance in serum in vitro, and investigating whether serum factor plays role in inhibiting the growth of C. albicans.
Methods: Identification of C. albicans taken from oral swab of candidiasis patient was conducted using CHROMagar, and confirmed by germ tube test. The cultures were serially diluted and inoculated in Sabouraud Dextrose Broth (SDB) contained 0% (control), 1%, 5%, or 10% xylitol and kept for 3 or 7 days. These inoculations were then exposed to either active or inactive serum (Fetal Bovine Serum heated in 65°C for 30 minutes to inactivate the complement) for 2 hours in 37°C. The Colony Forming Unit (CFU) of C. albicans in Sabouraud Dextrose Agar (SDA) were counted after 2 days. The same procedure was conducted for C. albicans ATCC 10231 strain. Data was analyzed using one-way ANOVA with 0.05.
Results: After 3 days cultured in media with or without xylitol, the CFU of C. albicans exposed to active serum were significantly lower than those exposed to inactive serum (p = 0.032). Increased concentration of xylitol lead to increased resistance of C. albicans in active serum, though it was not significant statistically (p = 0.689). Only 7 days exposure of 10% xylitol resulted in significantly higher growth of C. albicans (p = 0.034), but no significant difference on C. albicans CFU between in active or inactive serum (p = 0.404).
Conclusion: Exposure of 1%, 5%, or 10% xylitol for 3 or 7 days has no significant effect on C. albicans resistance in serum. The inhibition effect of serum factor to C. albicans growth was significant after 3 days, but not effective anymore after 7 days.

"Manifestasi kandidiasis oral berhubungan dengan pembentukan biofilm pada permukaan mukosa. Candida albicans merupakan jamur penyebab utama kandidiasis oral. Propolis dilaporkan berpengaruh terhadap pembentukan biofilm C.albicans. Tujuan: Menilai efektifitas permen dengan kandungan propolis terhadap pembentukan biofilm C.albicans dibandingkan dengan permen madu. Metode: C. albicans dipaparkan dengan larutan permen X, permen propolis madu, dan permen madu 50% pada 96-well plate yang sudah dicoating saliva dan serum. Untuk menganalisis pembentukan biofilm C. albicans dilakukan uji dengan MTT assay. Data dianalisis dengan uji one-way ANOVA. Hasil: Terdapat peningkatan yang signifikan pada perlakuan dengan permen X baik dengan coating saliva (p=0.000) maupun serum (p=0.000). Tidak terdapat perubahan yang signifikan pada pembentukan biofilm C. albicans yang ditambahkan permen propolis madu dengan coating saliva (p=0.187) maupun serum (p=0.386) serta permen madu dengan coating saliva (p=0.062) maupun serum (p=0.396). Simpulan: Pemberian larutan permen X bermakna dalam meningkatkan pembentukan biofilm C.albicans. Pemberian larutan permen propolis madu dan permen madu tidak mempengaruhi pembentukan biofilm C.albicans.

Manifestations of oral candidiasis related with biofilm formation on mucosal sufaces. Candida albicans is the main microbial culprit in oral candidiasis. Propolis is reported to have an effect on biofilm formation of C.albicans. Objective: To evaluate effect of candy that contains propolis on in-vitro biofilm formation of C.albicans compared with honey candy. Methods: C. albicans was exposed with 50% X candy solutions, propolis honey candy solutions, and honey in 96-well plate that had been coated with saliva and serum. To analyze formation of C. albicans biofilm MTT assay was used. Data was analyzed with one-way ANOVA. Result: There were significant increases on biofilm formation of C.albicans with X candy treatment either coated with saliva (p=0.000) or serum (p=0.000). There were no significant differences of C. albicans biofilm formation with addition of propolis honey candy either coated with saliva (p=0.187) or serum (p=0.386) and honey candy either coated with saliva (p=0.062) or serum (p=0.396). Conclusion: Treatment with propolis honey candy and honey candy solutions has no significant effect for biofilm formation of C.albicans. Effect of treatment with X candy solution was significant in increasing C.albicans biofilm formation."

"Latar belakang: Infeksi jamur oportunistik oleh Candida sp,. di dunia terus bertambah setiap tahunnya. Candida albicans menjadi jamur dengan tingkat infeksi kandidiasis paling banyak. Penggunaan antibiotik spektrum luas dan kateter urin dalam waktu lama menyebabkan obat standar seperti flukonazol mayoritas menjadi resisten terhadap kandidiasis. Propolis ternyata memiliki senyawa bioaktif yang mampu menghambat pertumbuhan Candida albicans. Propolis Lombok memiliki zat aktif senyawa fenolik seperti flavonoid yang berperan sebagai antijamur. Penelitian ini bertujuan untuk membandingkan efektivitas propolis Lombok dengan flukonazol, serta membandingkan metode well diffusion dengan microbroth dilution. Metode: Penelitian secara in vitro ini menggunakan design penelitian eksperimental yaitu metode well diffusion dan microbroth dilution. Perlakuan dibagi menjadi kelompok kontrol positif: flukonazol, kelompok kontrol negatif: DMSO, serta tiga kelompok propolis dengan kadar 50mg/mL, 70mg/mL, dan 100mg/mL. Setiap prosedur diulang 3 kali. Data dianalisis menggunakan uji statistik Kruskal-walis dan One Way ANOVA. Hasil: Pemberian propolis Lombok menunjukkan zona hambatan dengan rentang rata-rata 7,9 mm–10 mm pada metode agar difusi, sedangkan menunjukkan zona hambatan rata–rata sebesar 15 mm (p < 0,05). Jika dibandingkan dengan metode microbroth dilution ditemukan hasil yang serupa. Pada analisis statistik tidak ditemukan adanya perbedaan bermakna di antara ketiga konsentrasi propolis Lombok (p > 0,05). Kesimpulan: Propolis Lombok memiliki efek antijamur terhadap pertumbuhan Candida albicans berdasarkan dua metode yang sudah digunakan, namun efektivitasnya masih lebih rendah dibandingkan flukonazol.

Introduction: Opportunistic fungal infection by Candida sp,. in the world continues to grow every year. Candida albicans is the fungus with the highest candidiasis infection rate. The use of broad-spectrum antibiotics and urinary catheters for a long time have caused most of the standard drugs such as fluconazole to become resistant to candidiasis. Propolis turns out to have bioactive compounds that can inhibit the growth of Candida albicans. Lombok propolis has active phenolic compounds such as flavonoids which act as antifungals. This study aims to compare the effectiveness of Lombok propolis with fluconazole, and to compare the well diffusion method with micro broth dilution. Method: This in vitro study used an experimental research design, namely the well diffusion and microbroth dilution methods. The treatment was divided into a positive control group: fluconazole, a negative control group: DMSO, and three groups of propolis with levels of 50mg/mL, 70mg/mL, and 100,g/mL. Each procedure was repeated 3 times. Data were analyzed using the Kruskal-Wallis statistical test and One-Way ANOVA. Result: The provision of propolis in Lombok showed an inhibition zone with an average range of 7.9 mm–10 mm on the agar diffusion method, while the inhibition zone showed an average of 15 mm (p < 0.05). When compared with the microbroth dilution method, similar results were found. Statistical analysis did not find any significant difference between the three concentrations of Lombok propolis (p > 0.05). Conclusion: Lombok propolis has antifungal effect on the growth of Candida albicans based on two methods that have been used, but its effectiveness is still lower than fluconazole."

Metode: Uji MTT digunakan untuk menguji viabilitas C. albicans pada biofilm dan dikonversikan persen hambat ekstrak etanol temulawak KHBM50 . Efek EET terhadap gambaran mikroskopis setiap fase perkembangan biofilm C. albicans diamati dengan Scanning Electron Microscopy.
Hasil: Nilai Konsentrasi Hambat Biofilm Minimal KHBM50 EET terhadap biofilm C. albicans isolat klinis pada fase awal, menengah, dan maturasi secara berturut-turut adalah 20 , 30 , dan 35 . Gambaran mikroskopis pada setiap fase perkembangan biofilm C. albicans terlihat penurunan jumlah sel dan densitas C. albicans, serta terhambatnya pembentukan filamen dibandingkan dengan kelompok tanpa perlakuan.
Kesimpulan: EET mampu menghambat perkembangan fase awal, menengah, dan maturasi biofilm C. albicans isolat klinis. Semakin matur fase perkembangan biofilm, C. albicans akan semakin resisten terhadap ekstrak temulawak. Paparan ekstrak temulawak memengaruhi kemampuan C. albicans isolat klinis dalam membentuk filamen serta menurunkan jumlah sel dan densitas biofilm.

Background: Javanese turmeric which contains xanthorrhizol is known to have antifungal effect. Xanthorrhizol is reported to be able to eradicate Candida albicans' biofilm formation.
Objective: Analyze the correlation between inhibition concentration of Javanese turmeric ethanol extract JTEE and each development phase of C. albicans' biofilm, and observing microscopic appearance of each phase of C. albicans biofilm.
Method: MTT assay was used to test the viability of C. albicans towards biofilm and converted to Minimum Biofilm Inhibitory Concentration MBIC50 . JTEE' s effect on each phase of microscopic appearance of C. albicans' biofilm is observed by Scanning Electron Microscopy.
Result: MBIC50 of JTEE towards development of clinical isolate of C. albicans' biofilm in the early adhesion and proliferation , intermediate, and maturation phase as follows 20, 30, and 35 respectively. The microscopic appearance on each phase of C. albicans' biofilm development shows decrease in cell number and density, as well as inhibiton of filament formation compared with control group.
Conclusion: JTEE can inhibit the development phases of C. albicans' biofilm. The potency of JTEE to inhibit development of C. albicans' biofilm was decreased along with the maturation of biofilm. The JTEE' s exposure leads to changes of microscopic appearance of C. albicans' biofilm development.
