"Kanker serviks merupakan salah satu jenis kanker yang paling banyak diderita wanita di Indonesia. Pengobatan kanker serviks seperti operasi, kemoterapi, dan radioterapi masih terus diupayakan, namun tingkat kesembuhan penderita kanker serviks masih rendah, yaitu dengan 1-year survival rate hanya sekitar 10-20%. Salah satu tanaman terapeutik di Indonesia yang memiliki aktivitas antikanker adalah bajakah tampala (Spatholobus littoralis Hassk). Nanopartikel bajakah tampala dilaporkan memiliki sitotoksisitas sedang terhadap sel HeLa. Namun, belum diketahui konsentrasi optimal ekstrak bajakah tampala terhadap sel HeLa berdasarkan viabilitas dan ultrastruktur sel. Tujuan dari penelitian ini adalah mengetahui pengaruh variasi konsentrasi ekstrak bajakah tampala (100, 150, 200 dan 250 µg/mL) terhadap viabilitas sel HeLa menggunakan trypan blue dan ultrastruktur sel HeLa dengan scanning electron microscope (SEM). Hasil analisis statistik dengan tingkat kepercayaan 0,05 menunjukkan terdapat perbedaan signifikan nilai persentase viabilitas sel antara kontrol dan perlakuan konsentrasi ekstrak bajakah tampala. Hasil pengamatan ultrastruktur dengan scanning electron microscope menunjukkan terdapat perbedaan ultrastruktur sel HeLa antara sampel kontrol dengan perlakuan. Hasil tersebut menunjukkan bahwa konsentrasi ekstrak bajakah tampala 100 μg/mL, 150 μg/mL, 200 μg/mL dan 250 μg/mL memiliki pengaruh terhadap viabilitas dan ultrastruktur sel HeLa. Konsentrasi 200 μg/mL cenderung lebih mampu menekan viabilitas sel dan menunjukkan karakteristik apoptosis secara ultrastruktur.

Cervical cancer is one of the cancer that most women suffer from in Indonesia. Treatment for cervical cancer such as surgery, chemotherapy and radiotherapy is still being attempted, but the cure rate for cervical cancer sufferers is still low, with a 1-year survival rate of only around 10-20%. One of the therapeutic plants in Indonesia that has anticancer activity is bajakah tampala (Spatholobus littoralis Hassk.). Bajakah tampala nanoparticles were reported to have moderate cytotoxicity against HeLa cells. However, the optimal concentration of bajakah tampala extract for HeLa cells is not yet known based on cell viability and ultrastructure. The aim of this research was to determine the effect of varying concentrations of bajakah tampala extract (100, 150, 200 and 250 µg/mL) on HeLa cell viability using trypan blue and HeLa cell ultrastructure using a scanning electron microscope (SEM). The results of statistical analysis with a confidence level of 0.05 show that there is a significant difference in the percentage value of cell viability between the control and the bajakah tampala extract concentration treatment. The results of ultrastructural observations using a scanning electron microscope showed that there were differences in the ultrastructure of HeLa cells between control and treated samples. These results indicate that the concentrations of bajakah tampala extract of 100 μg/mL, 150 μg/mL, 200 μg/mL and 250 μg/mL have an influence on the viability and ultrastructure of HeLa cells. A concentration of 200 μg/mL tends to be more capable of suppressing cell viability and shows ultrastructural characteristics of apoptosis.

"Kanker serviks menjadi salah satu penyebab kematian tertinggi di seluruh dunia setiap tahunnya. Upaya untuk mengobati penyakit kanker baik dengan cara kemoterapi dan imunologik juga telah dilakukan, namun kemoterapi berkepanjangan dapat memicu resistensi sel kanker karena kemoterapi kurang selektif terhadap sel kanker. Eucalyptol telah dilaporkan memiliki beberapa sifat obat yang meliputi antibakteri, antijamur, dan aktivitas antheimintik. Eucalyptol telah dilaporkan dapat menginduksi kematian sel karsinoma kulit (A431), osteosarcoma (MG-63) dan keratinosit manusia (HaCaT). Akan tetapi pengaruh senyawa eucalyptol pada sel HeLa belum diketahui. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi eucalyptol (25 µg/mL, 50 µg/mL, 100 µg/mL, 200 µg/mL) terhadap viabilitas sel HeLa dengan metode Trypan Blue, ScepterTM 3.0, dan ultrastruktur sel HeLa dengan metode Scanning Electron Microscope (SEM). Hasil analisis statistik dengan tingkat kepercayaan 0,05 menunjukkan terdapat perbedaan signifikan nilai persentase viabilitas dengan metode trypan blue antara kontrol dan perlakuan. Hasil pengamatan dengan ScepterTM 3.0 menunjukkan bahwa senyawa eucalyptol dapat menekan konsentrasi sel dan menyebabkan penyusutan sel. Hasil pengamatan kualitatif dengan scanning electron microscope menunjukkan perubahan ultrastruktur sel HeLa pada sampel perlakuan. Hal tersebut menunjukkan bahwa konsentrasi eucalyptol sebesar 25 µg/mL, 50 µg/mL, 100 µg/mL, dan 200 µg/mL berpengaruh terhadap viabilitas, konsentrasi sel, diameter sel, volume sel, dan ultrastruktur sel HeLa. Konsentrasi senyawa eucalyptol 200 µg/mL cenderung paling tinggi menekan viabilitas konsentrasi sel, diameter sel, volume sel, dan ultrastruktur sel HeLa dibanding konsentrasi lain.

Cervical cancer is one of the leading causes of death worldwide every year. Efforts to treat cancer both by chemotherapy and immunologic methods have also been carried out, but prolonged chemotherapy can trigger cancer cell resistance because chemotherapy is less selective against cancer cells. Eucalyptol has been reported to have several medicinal properties which include antibacterial, antifungal, and anthelmintic activity. Previous studies have reported that eucalyptol can induce cell death of skin carcinoma (A431), osteosarcoma (MG-63), and human keratinocytes (HaCaT). Nevertheless, its effects on HeLa cell viability and ultrastructure have not been evaluated. This study aims to determine the effect of eucalyptol concentration (25 g/mL, 50 g/mL, 100 g/mL, 200 g/mL) on the viability of HeLa cells using the Trypan Blue, ScepterTM method. 3.0, and ultrastructure of HeLa celss using Scanning Electron Microscope (SEM). The results of statistical analysis with a confidence level of 0.05 showed that there was a significant difference in the percentage of viability with the trypan blue method between the control and treatment. The results of observations with ScepterTM 3.0, eucalyptol can suppress cell concentration and cell shrinkage. The results of qualitative observations using a scanning electron microscope showed changes in the ultrastructure of HeLa cells in the treatment samples. This shows that the eucalyptol concentrations of 25 g/mL, 50 g/mL, 100 g/mL, and 200 g/mL affected the viability, cell concentration, cell diameter, cell volume, and morphology of HeLa cells. The concentration of eucalyptol at 200 g/mL tends to affect the viability, cell concentration, cell diameter, cell volume, and morphology of HeLa cells compare to other concentrations."

"Morbiditas dan mortalitas kanker paru yang tinggi serta tantangan kemoterapi (resistensi sel kanker dan efek samping serius) menimbulkan perlunya ekplorasi agen antikanker alami. Jojoba (Simmondsia chinensis (Link) Schneider) adalah salah satu tumbuhan yang menjadi sumber agen antikanker potensial. Ekstrak dan minyak biji jojoba telah dilaporkan memiliki efek sitotoksik terhadap viabilitas sel kanker kolorektal HCT-116, sel kanker payudara MCF-7, dan sel melanoma MV-3. Akan tetapi, pengaruh minyak biji jojoba terhadap viabilitas dan ultrastruktur sel kanker paru A549 belum diketahui sebelumnya. Maka dari itu, penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi minyak biji jojoba (50, 100, 200, 300, 400, dan 500¼g/mL) terhadap viabilitas sel A549 dengan metode Trypan Blue serta ultrastruktur sel A549 dengan metode Scanning Electron Microscopy. Hasil uji dan analisis statistik membuktikan bahwa minyak biji jojoba dengan konsentrasi 100, 200, 300, 400, dan 500 μg/mL memiliki pengaruh yang signifikan terhadap penurunan viabilitas sel A549. Konsentrasi 300¼g/mL minyak biji jojoba mampu mengakibatkan perubahan ultrastruktur sel A549 berupa blebbing membran dan pemisahan sel dari jaringan di sekitarnya serta hilangnya sitoskeleton aktin. Analisis induksi apoptosis, potensi antikanker senyawa simmondsin murni, serta pengaruh secara in vivo perlu diuji untuk membuktikan potensi antikanker minyak biji jojoba terhadap kanker paru secara lebih lanjut.

The high morbidity and mortality rate of lung cancer and the challenges of chemotherapy (cancer cell resistance and serious side effects) have led to the urgency to explore natural anticancer agents. Jojoba (Simmondsia chinensis (Link) Schneider) is one of the plants that is known for its unique seed wax ester oil and has various potentials, one of which is anticancer. Jojoba seed oil and extracts has been reported to exert cytotoxic effects on the viability of HCT-116 colorectal cancer cells, MCF-7 breast cancer cells, and MV-3 melanoma cells. However, the effect of jojoba seed oil on the viability and ultrastructure of A549 lung cancer cells has not been previously determined. This study aims to determine the effect of various concentrations of jojoba seed oil (50, 100, 200, 300, 400, and 500 μg/mL) on A549 cell viability by Trypan Blue method and ultrastructure by Scanning Electron Microscopy method. The test results and statistical analysis proved that jojoba seed oil with concentrations of 100, 200, 300, 400, and 500¼g/mL had a significant effect on reducing A549 cell viability. The jojoba seed oil concentration of 300¼g/mL can cause membrane blebbing, detachment from surrounding tissue, and loss of cytoskeleton on A549 cells. Analysis of apoptosis induction, anticancer potential of pure simmondsin compound, and in vivo effects need to be evaluated to further prove the anticancer potential of jojoba seed oil towards lung cancer."

"Kanker payudara merupakan salah satu jenis kanker dengan dampak yang paling signifikan di dunia. Saat ini, pengobatan kanker payudara terbatas pada kemoterapi yang memiliki efek samping berbahaya bagi tubuh. Minyak biji jojoba (Simmondsia chinensis (Link) Scneider) dikenal memiliki senyawa bioaktif seperti simmondsin, kuinon, dan fenolik yang memiliki sifat antikanker sehingga potensial digunakan untuk pengobatan alami. Namun, belum ada penelitian yang memaparkan tentang efek sitotoksik dari minyak biji jojoba terhadap sel kanker payudara MDA-MB-231 yang dikenal agresif. Penelitian ini bertujuan untuk mengetahui efek sitotoksik dari minyak biji jojoba terhadap viabilitas, apoptosis, dan ultrastruktur sel MDA-MB-231. Viabilitas sel MDA-MB-231 diperoleh dengan menggunakan metode trypan blue, apoptosis dengan menggunakan flow cytometry, dan ultrastruktur dengan menggunakan Scanning Electron Microscopy (SEM). Hasil analisis menunjukkan viabilitas paling rendah terdapat pada konsentrasi 200 μg/mL. Hasil uji apoptosis menunjukkan bahwa total apoptosis paling tinggi terdapat pada konsentrasi 400 μg/mL dengan efek sitotoksik bersifat dose-dependent. Sementara, hasil analisis SEM menunjukkan terdapat perbedaan ultrastruktur berupa penyusutan sel dan terbentukknya blebbing sebagai hallmark apoptosis. Keseluruhan pengujian menunjukkan pengaruh terhadap penurunan viabilitas, peningkatan apoptosis, dan perubahan ultrastruktur sel MDA-MB-231. Namun, diperlukan pengujian lanjutan untuk mengetahui mekanisme penginduksian apoptosis dengan menggunakan ELISA serta komposisi senyawa bioaktif dalam minyak biji jojoba dengan menggunakan HPLC.

Breast cancer is one type of cancer with the most significant impact in the world. Currently, breast cancer treatment is limited to chemotherapy which has dangerous side effects for the body. Jojoba (Simmondsia chinensis (Link) Scneider) seed oil is known to contain bioactive compounds such as simmondsin, quinones and phenolics which have anticancer properties so they have the potential to be used for natural treatment. However, there has been no research that describes the cytotoxic effect of jojoba seed oil on MDA-MB-231 breast cancer cells which are known to be aggressive. This study aims to determine the cytotoxic effect of jojoba seed oil on viability, apoptosis, and ultrastructure of MDA-MB-231 cells. MDA-MB-231 cell viability was obtained using the trypan blue method, apoptosis using flow cytometry, and ultrastructure using Scanning Electron Microscopy (SEM). The analysis results showed that the lowest viability was at a concentration of 200 µg/mL. The results of the apoptosis test showed that the highest total apoptosis was at a concentration of 400 µg/mL with the cytotoxic effect being dose-dependent. Meanwhile, the results of SEM analysis show that there are ultrastructural differences in the form of cell shrinkage and the formation of blebbing as a hallmark of apoptosis. Overall testing showed an effect on decreasing viability, increasing apoptosis, and changes in the ultrastructure of MDA-MB-231 cells. However, further testing is needed to determine the mechanism of induction of apoptosis using ELISA and the composition of bioactive compounds in jojoba seed oil using HPLC."
