"Latar Belakang: Tindakan peritomi konjungtiva meningkatkan penanda inflamasi pada permukaan okular dan menyebabkan penurunan densitas sel goblet yang berimplikasi pada ketidakstabilan lapisan air mata. Pemberian HP-guar diharapkan mampu memperbaiki keadaan permukaan okular pasca peritomi konjungtiva 360° dibandingkan kontrol.
Tujuan: Untuk menilai efektivitas artificial tears HP-guar dalam melindungi permukaan okular pasien pasca peritomi konjungtiva 360°.
Desain: Penelitian prospektif, uji klinis open label.
Hasil: Terdapat 23 subjek yang berpartisipasi dalam penelitian ini. Kelompok HP-guar menunjukkan hasil yang lebih baik dibandingkan dengan kelompok kontrol secara signifikan dalam hal TFBUT (9,43+2,20 vs 5,95+1,93, p=0,001), densitas sel goblet (68,73 ±97,49 vs 10,00 ± 22,09,p =0,012), skor epitel (1,73±1.00 vs 4,55±1.57, p=0.000). Tidak terdapat perbedaan bermakna antara kedua kelompok dalam hal skor lissamine green (1,33±1,55 vs 2,18±1,72, p=0,146) dan IL-6 air mata (60,32 ±17,86 vs 57,44 ±24,67, p=0,782).
Kesimpulan: Pemberian HP-guar lebih memperbaiki nilai TFBUT, densitas sel goblet dan epitel konjungtiva dibandingkan kontrol. HP-guar tidak berpengaruh terhadap skor lissamine green dan kadar IL-6 air mata.

Background: Conjunctival peritomy, a step commonly done in ocular surgery, increases ocular surface inflammation and decreases goblet cell density (GCD) that could lead to tear film instability. Hydroxypropyl-guar (HP-guar) is expected to improve ocular surface related to conjunctival peritomy.
Purpose: To evaluate HP-guar effectiveness in protecting ocular surface after conjunctival peritomy.
Methods: Randomized controlled trial, open label study, on subjects underwent scleral buckling surgery.
Result: Twenty-three participants were involved in this study. The HP-guar group showed statistically better results compared to the control group regarding Tear-film break-up time (TFBUT) (9,43±2,20 vs 5,95±1,93, p=0.001), GCD (68,73 ±97,49 vs 10,00 ± 22,09,p =0.012), and epithelial score (1.73±1.00 vs 4.55±1.57, p=0.000). No statistical differences were found between groups in lissamine green score (1.33±1.55 vs 2.18±1.72, p=0.146) and IL-6 tear fluid level (60.32 ±17.86 vs 57.44 ±24.67, p=0.782).
Conclusion: The addition of HP-Guar to regular treatment after conjunctival peritomy increases TFBUT, goblet cell density, and improves conjunctival epithelial cells. HPguar has no effect on ocular surface inflammation.

"Tujuan Mengembangkan cara spot sederhana untuk melekatkan suspensi sel hasil kultur pada kaca objek. Metode Kami membandingkan tiga cara, masing-masing pada kaca objek biasa dan khusus (Shandon-Polysin). Ketiga cara tersebut adalah membuat sediaan spot kecil secara langsung, atau dengan menambahkan 3 atau 10 μl fetal bovine serum (FBS) per 20 μl sampel. Dengan demikian, secara keseluruhan ada 6 cara. Hasilnya dinilai secara kualitatif dalam hal warna latar belakang, warna dan keutuhan batas spot, dan bagian yang terlipat dan terlepas. Selanjutnya, untuk tiap kaca objek, jumlah sel utuh yang melekat dihitung, dan persentase sel utuh yang melekat per jumlah sel yang dibuat spot juga dihitung. Perbedaan sel utuh yang melekat di antara keenam cara di atas dianalisis dengan ANOVA menggunakan program SPSS 13.0 for Windows. Hasil Tidak ada perbedaan bermakna di antara keenam cara di atas dalam hal persentase sel utuh yang melekat (P= 0,804), walaupun cara yang menggunakan kaca objek khusus tanpa penambahan FBS (kadar FBS final 5%) menghasilkan persentase sel utuh melekat yang paling tinggi, dengan latar belakang bersih tanpa lipatan. Kesimpulan Kami telah mengembangkan cara spot sederhana untuk membuat sediaan sitologi suspensi sel hasil kultur, dan hasil terbaik didapat dengan menggunakan kaca objek khusus dengan suspensi sel berkadar FBS 5%.

Aim To develop a simple spot method to attach cultured cells in suspension on to a glass slide. Methods We compared three approaches using both conventional and special glass slide (Shandon-Polysin)., either without additional fetal bovine serum (FBS), or with addition of 3 or 10 μl of FBS to a 20 μl sample (altogether there were six approaches). The slides were examined qualitatively for the background color, boundary color and intactness, and whether there were folded and detached parts. Further, for each slide, the attached intact cells were counted, and the percentage of attached intact cells per number of spotted cells was calculated. The difference in attach intact cells between different approaches was analyzed by ANOVA using SPSS 13.0 for windows. Results There were no significant difference in the percentage of attached intact cells between the six approaches (P= 0.804), though the approach using special glass slide without additional FBS (FBS final concentration 5%) yield the highest percentage of attached intact cells, showed clean background without folded parts. Conclusions We have developed a simple spot method for cultured cell suspension, and the best approach to make spot specimen is using special glass slide with 5% FBS in the cell suspension.

"Circulating tumor cell (CTC) merupakan intermediet proses metastasis kanker yang dapat dimanfaatkan untuk diagnosis, prognosis, dan target pengobatan kanker. Pengembangan pemanfaatan CTC dapat dilakukan dengan penelitian yang umumnya melibatkan proses kultur. Medium bebas serum dinilai lebih baik dibanding medium berserum karena dapat menghasilkan data yang lebih konsisten, sehingga lebih cocok digunakan untuk penelitian yang mengkaji aktivitas fisiologi sel dan persinyalan molekular. Namun, medium bebas serum memerlukan suplemen agar sel dapat tumbuh optimum. Penambahan suplemen insulin-transferrin-selenium (ITS) telah diketahui memiliki peran penting dalam kultur sel keratosit, ovarium, dan keratinosit. Namun, belum diketahui peran ITS dalam medium bebas serum untuk kultur CTC. Penelitian ini mengkaji perbedaan efek FBS dan ITS dengan konsentrasi 1X dan 10X dalam medium bebas serum terhadap CTC yang diisolasi dengan metode eritrolisis. Kultur dilakukan selama 18 hari. Dinamika CTC dan leukosit diamati dengan meninjau viabilitasnya pada 6 hari pertama kutur. Selain itu, observasi morfologi dilakukan seiring dengan pengukuran morfometri sel. Pada hari ke-18, keberadaan CTC diverifikasi dengan imunofluoresens menggunakan marka cytokeratin 20 (CK20) dan plastin 3 (PLS3). Hasil penelitian menunjukkan bahwa CTC yang dikultur pada medium dengan penambahan 10X ITS memiliki diameter sel yang lebih besar dari yang dikultur pada medium dengan penambahan 1X ITS dan 10% FBS. Hal tersebut menunjukkan bahwa ITS memiliki peran penting dalam kultur CTC dalam medium bebas serum dan dalam konsentrasi 10X dapat meningkatkan pertumbuhan CTC kanker kolorektal.

Circulating tumor cells (CTCs) are intermediates in the cancer metastasis process and hold potential for use in cancer diagnosis, prognosis, and treatment targeting. The development of CTC applications typically involves research incorporating cell culture processes. In cell culture, serum-free media are considered superior to serum-containing media as they yield more consistent data, making them more suitable for studies examining cell physiological activity and molecular signaling. However, serum-free media require supplementation to ensure optimal cell growth. The addition of insulin-transferrin-selenium (ITS) supplements is known to play a crucial role in the culture of keratocytes, ovarian cells, and keratinocytes. However, the role of ITS in serum-free media for CTC culture remains unknown. This study investigates the differential effects of fetal bovine serum (FBS) and ITS at concentrations of 1X and 10X in serum-free media on CTCs isolated via erythrolysis. Cultures were maintained for 18 days, with CTC and leukocyte dynamics observed by assessing cell viability during the first six days of culture. Additionally, morphological observations and cell morphometric measurements were conducted. On the 18th day, the presence of CTCs was verified using immunofluorescence with cytokeratin 20 (CK20) and plastin 3 (PLS3) markers. The results indicated that CTCs cultured in media supplemented with 10X ITS exhibited larger cell diameters compared to those cultured with 1X ITS and 10% FBS. This finding suggests that ITS plays a critical role in the successful culture of CTCs in serum-free media and that a 10X concentration of ITS can enhance the growth of colorectal cancer CTCs."
